Project description:The plasma multimeric glycoprotein von Willebrand factor (VWF) plays a critical role in primary hemostasis by tethering platelets to exposed collagen at sites of vascular injury. Recent studies have identified additional biological roles for VWF, and in particular suggest that VWF may play an important role in regulating inflammatory responses. Critically however, the molecular mechanisms through which VWF exerts its immuno-modulatory effects remains poorly understood. In this study, we report for the first time that VWF binding to macrophages triggers direct downstream MAPkinase signaling leading to NF-kB activation and production of pro-inflammatory cytokines and chemokines. In keeping with these observations, VWF binding also promotes macrophage M1 polarization and shifts macrophage metabolism towards glycolysis in a p38 dependent manner. Cumulatively, our findings define an entirely new biological role for VWF in modulating macrophage function, and thereby establish a novel link between primary hemostasis and innate immunity.

Project description:Synthesis of the hemostatic protein Von Willebrand factor (VWF) drives formation of endothelial storage organelles called Weibel-Palade bodies (WPBs). In the absence of VWF, angiogenic and inflammatory mediators that are co-stored in WPBs are subject to alternative trafficking routes. In Von Willebrand disease (VWD) patients, the partial or complete absence of VWF/WPBs may lead to additional bleeding complications, such as angiodysplasia. Studies addressing the role of VWF using VWD patient-derived blood outgrowth endothelial cells (BOECs) have reported conflicting results due to the intrinsic heterogeneity of patient-derived BOECs. To study the role of WPBs in endothelial cells using CRISPR-mediated knockout of VWF in BOECs. We used CRISPR/Cas9 gene editing in single donor cord blood-derived BOECs (cbBOECs) to generate clonal VWF-/- cbBOECs. Clones were selected using high-throughput screening, VWF mutations were validated by sequencing and cells were phenotypically characterized. Two VWF-/- BOEC clones were obtained and were entirely devoid of WPBs, while overall cell morphology was unaltered. Several WPB proteins, including CD63, syntaxin-3 and the cargo proteins Ang-2, IL-6 and IL-8 showed alternative trafficking and secretion in absence of VWF. Interestingly, Ang-2 changed localization to the cell periphery and colocalized with Tie-2. CRISPR editing of VWF provides a robust method to create VWF deficient BOECs that can be directly compared to their wild-type counterparts. Results obtained with our model system confirmed alternative trafficking of several WPB proteins in the absence of VWF and support the theory that increased Ang-2/Tie-2 interaction contributes to angiogenic abnormalities in VWD patients.

Project description:von Willebrand factor (VWF) is the protective carrier of procoagulant factor VIII (FVIII) in the shear forces of the circulation, prolonging its half‐life and delivering it to the developing thrombus. VWF∙FVIII complex formation is characterized by catch‐bond behavior in which force first decelerates then accelerates bond dissociation. Patients with mutations in VWF at the FVIII binding site phenocopies hemophilia A and the most common mutations involve cysteine residues of multiple disulfides. Thirteen VWF disulfide bonds at the FVIII binding site exist in formed and unformed states, and binding of FVIII results in partial formation of 12 of the VWF bonds. The VWF∙FVIII bond stiffens in response to force and this property is ablated when VWF disulfide bonds are prevented from forming, resulting in slip‐bond behavior. These findings demonstrate that FVIII binding to VWF involves dynamic changes in the covalent states of several VWF disulfides that are required for productive interaction.

Project description:Endothelial cells store von Willebrand factor (VWF) in rod-shaped secretory organelles, called Weibel-Palade bodies (WPBs). WPB exocytosis is coordinated by a complex network of Rab GTPases, Rab-effectors and SNARE proteins. We have previously identified STXBP1 as the link between the Rab27A-Slp4-a complex on WPBs and the SNARE proteins syntaxin-2 and -3. In this study we investigate the function of syntaxin-3 in VWF secretion. In human umbilical vein endothelial cells (HUVECs) and in blood outgrowth endothelial cells (BOECs) from healthy controls endogenous syntaxin-3 immunolocalized to WPBs. A detailed analysis of BOECs isolated from a patient with variant microvillus inclusion disease (MVID), carrying a homozygous mutation in STX3 (STX3-/-), showed a loss of syntaxin-3 protein and absence of WPB-associated syntaxin-3 immunoreactivity. Ultrastructural analysis revealed no detectable differences in morphology or prevalence of immature or mature WPBs in control versus STX3-/- BOECs. VWF multimer analysis showed normal patterns in plasma of the MVID patient, and media from STX3-/- BOECs, together indicating WPB formation and maturation are unaffected by absence of syntaxin-3. However, a clear defect in Ca2+ and cAMP-mediated VWF secretion was found in the STX3-/- BOECs. Co-immunoprecipitation studies showed that syntaxin-3 associates with the WPB SNAREs SNAP23 and VAMP8. Our data reveal syntaxin-3 as a novel WPB-associated SNARE controlling VWF secretion and highlight the complex regulation of WPB exocytosis by multiple SNARE complexes.

Project description:Von Willebrand Factor (vWF), a plasma glycoprotein, plays essential roles in primary hemostasis, in cooperation with other coagulation factors. The glycosylation affects many biological phases during the protein life-cycle. In this work, we comprehensively characterized the microheterogeneity at all probable N-glycosites in simultaneous with O-glycosites, beside the N-occupancy estimation.An RP-LC-MS/MS system functionalized with CID and HCD tandem mass techniques was utilized. Our results showed gradual glycosylation occupancy along the protein backbone. A total of 181 glycoforms, 173 N-froms and 8 O-forms, were detected and specified into plasma vWF glycosites. Given subtle characterization of site-specific glycoforms is critical for profound understanding the protein biological functions and activity.

Project description:Abstract Von Willebrand factor (VWF) is a multimeric hemostatic protein primarily synthesized in endothelial cells (ECs). VWF is stored in endothelial storage organelles, the Weibel-Palade bodies (WPBs), whose biogenesis strongly depends on VWF anterograde trafficking and Golgi architecture. Elongated WPB morphology is correlated to longer VWF strings with better adhesive properties. We previously identified the SNARE SEC22B, which is involved in anterograde ER-to-Golgi transport, as a novel regulator of WPB elongation. To elucidate novel determinants of WPB morphology we explored endothelial SEC22B interaction partners in a mass spectrometry-based approach, identifying the Golgi SNARE Syntaxin 5 (STX5). We established STX5 knockdown in ECs using shRNA-dependent silencing and analyzed WPB and Golgi morphology, using confocal and electron microscopy. STX5-depleted ECs exhibited extensive Golgi fragmentation and decreased WPB length, which was associated with reduced intracellular VWF levels, and impaired stimulated VWF secretion. However, the secretion incompetent organelles in shSTX5 cells maintained WPB markers such as Angiopoietin 2, P-selectin, Rab27A, and CD63. Taken together, our study has identified SNARE protein STX5 as a novel regulator of WPB biogenesis

Project description:An in-frame heterozygous large deletion of exons 4-34 of the von Willebrand factor (VWF) gene was identified in an index patient (IP) with type 3 von Willebrand disease (VWD), as the only mutation. The IP exhibited severe bleeding episodes despite prophylaxis treatment, with a short VWF half-life after infusion of VWF/FVIII concentrates. This study intends to elucidate the causal molecular mechanism of this large deletion. Transcript analysis confirmed transcription of normal VWF mRNA besides an aberrant deleted transcript. The amount of secreted VWF from blood outgrowth endothelial cells (BOECs) isolated from the IP was not significantly different from that of controls. However, IP-BOECs exhibited a deficiency in the assembly of VWF multimers and biogenesis of the Weibel-Palade bodies (WPBs). Furthermore, immunostaining of IP-BOECs demonstrated subcellular mislocalization of WPBs pro-inflammatory cargos angiopoietin-2 (Ang2) and P-selectin. Additionally, whole-transcriptome RNA-sequencing of the BOECs and subsequent Ingenuity Pathway Analysis indicated the significant alterations of canonical pathways in IP-BOECs related to inflammatory responses, cell adhesion, extracellular organization, and angiogenesis (e.g. granulocyte adhesion and Rho-related signaling pathways), which are known downstream signaling pathways induced by Ang2. Accordingly, the IP-BOECs exhibited an increased adhesiveness to leukocytes which may contribute to accelerated VWF clearance. In conclusion, deleted VWF has a dominant-negative impact on the elongation of multimers and biogenesis of WPBs. Aberrant WPBs lead to the alternative trafficking of its cargos in a specific way, which, in turn, may cause distinctive perturbations in cellular signaling pathways, resulting in the exceptional phenotypes in the current patient.

Project description:A type 3 von Willebrand disease (VWD) index patient (IP) remains mutation-negative after completion of conventional diagnostic analysis, including multiplex ligation-dependent probe amplification and sequencing of the promotor, exons, and flanking intronic regions of VWF gene (VWF). In this study, we intended to elucidate causitive genetic defect through screening of the whole VWF (including complete intronic region), mRNA analysis, and study of the patient-derived endothelial colony-forming cells (ECFCs). The entire VWF was analyzed by next-generation sequencing (NGS) on an Illumina platform. The NGS revealed a novel variant in VWF intron 8 (997+118 T>G). The subsequent assessments using bioinformatics tools (e.g. SpliceAl) predicted this variant creates a new donor splice site (ss) in intron 8, which could outcompete the consensus 5’ donor ss at exon/intron 8 junction. This leads to an aberrant mRNA which contains a premature stop codon, targeting it to nonsense-mediated mRNA decay. The VWF mRNA from whole blood and isolated ECFCs were quantified using the TaqMan assay on an ABI 7500 real-time PCR system. The quantitative analysis confirmed the virtual absence of VWF mRNA. Additionally, the level of secreted VWF from IP ECFCs was considerably reduced (~6% of healthy donors).
