Project description:We generated a transgenic mouse line which express EGFP in the retina driven by the Crx promoter using BAC transgenesis. We sorted EGFP-positive photoreceptor precursors at E17.5 using FACS, and subsequently performed microarray analysis of the FACS-sorted cells. In order to clarify a molecular role of Crx in developing cone photoreceptor precursors, we investigated the expression profile of the BAC-Crx-EGFP-positive cells compared with that of the BAC-Crx-EGFP-negative cells at E17.5.

Investigating cone photoreceptor development using patient-derived NRLnull retinal organoids

Project description:Photoreceptor loss is a leading cause of blindness, but mechanisms underlying photoreceptor degeneration are not well understood. Treatment strategies would benefit from an improved understanding of gene-expression patterns directing photoreceptor development, as many genes are implicated in both development and degeneration. Neural retina leucine zipper (NRL) is critical for rod photoreceptor genesis and degeneration, with NRL mutations known to cause enhanced S-cone syndrome and retinitis pigmentosa. While murine Nrl loss has been characterized, studies of human NRL can identify important insights for human retinal disease. Here we utilized human organoid models of retinal development to molecularly define developmental alterations in a human model of NRL loss. Consistent with the function of NRL in rod fate specification, human retinal organoids lacking NRL develop S-opsin dominant photoreceptor populations. We report generation of two distinct S-opsin expressing populations in NRL null retinal organoids and identify MEF2C as a candidate regulator of cone development.

Transcriptome profiling of developing photoreceptor subtypes reveals candidate genes for avian photoreceptor diversification

Project description:Purpose: Avian photoreceptors are a diverse class of neurons, comprised of four single cones, the two members of the double cone, and rods. Many distinctive features of photoreceptor subtypes, including spectral tuning, oil droplet size and pigmentation, synaptic targets and spatial patterning, have been well characterized, but the molecular mechanisms underlying these attributes have not been explored. Furthermore, the signaling events and transcriptional regulators driving the differentiation of these diverse photoreceptors are currently unknown. Methods: To identify genes specifically expressed in distinct chicken (Gallus gallus) photoreceptor subtypes, we developed fluorescent reporters that label photoreceptor subpopulations, isolated these subpopulations using fluorescence-activated cell sorting, subjected them to next-generation sequencing, and conducted differential expression analysis. Results: We identified hundreds of differentially expressed genes from photoreceptor subpopulations labeled with rhodopsin, red opsin, green opsin, and violet opsin reporters. These genes are involved in a variety of processes, including phototransduction, transcriptional regulation, cell adhesion, maintenance of intra- and extra-cellular structure, and metabolism. Of particular note are a variety of differentially expressed transcription factors, which may drive and maintain photoreceptor diversity, and cell adhesion molecules that may mediate spatial patterning of photoreceptors and act to establish retinal circuitry. Direct Reprogramming of Human Fibroblasts into Retinal Progenitor Cells by Small Molecules to Treat Photoreceptor Degeneration

Project description:Retinal progenitor cells (RPCs) hold promise for restoring vision in advanced stages of photoreceptor degeneration. We have developed a technique to directly reprogram human Tenon's fibroblasts (HTFs) into induced RPC-like cells (iRPCs) using small molecules. To explore the transcriptomic profile of iRPCs, we performed bulk RNA-seq analysis of HTFs and FACS-sorted iRPCs. The Gene Ontology (GO) enrichment analysis demonstrated significantly highly enriched GO terms include Proteinaceous Extracellular Matrix, Axon and Dendrite, Synaptic Membrane and Transport Vesicle, Presynapse and Postsynapse, Basement Membrane and Collagen Trimer, Platelet Alpha Granule and ER Lumen. The significantly down-regulated GOs were Spindle and Chromosomal Structures, Sarcomere and Muscle Fiber Components, Cell Adhesion Complexes.
