Glycine Chloramine directed gene expression changes in Jurkat T cell lines

ABSTRACT: The goal of the current study was to determine whether chloramine exposure results in long-term alterations in transcriptional output. Proliferating Jurkat T-lymphoma cells were exposed to sublethal doses of glycine chloramine and RNA expression was profiled at 4, 24, 48 and 72 hours.

Epigenome analysis of glycine chloramine treated Jurkat T cells

Project description:Genome wide DNA methylation profiling of Jurkat T lymphoma cell lines, using the human DNA methylationEPIC 850K array. All cells were blocked using a thymidine block, and DNA methylation was analysed 4 hrs post release from the block (S-Phase) or 72 hours later. Samples are either untreated control (PBS) or treated with Glycine chloramine. Cells were exposed to Glycine chloramine in two independent time points: pre replication (same time as release from block, 3 replicates) or during S-phase (four replicates, 2 hrs post block release). These two exposures were assessed in two independent experiments and therefore have their own controls. Each replicate used cells from a different passage number, and was performed independently.

The human allicin-proteome: global protein S-thioallylation of by the garlic defence substance allicin and its biological effects

Project description:A single clove of edible garlic (Allium sativum L.) produces up to 5 mg of allicin (diallylthiosulfinate), a thiol-reactive sulfur-containing defence substance that gives injured garlic tissue its characteristic smell. Allicin induces apoptosis or necrosis in a dose-dependent manner but sublethal doses influence cellular metabolism and signalling cascades. Oxidation of protein thiols and depletion of the glutathione pool are thought to be responsible for allicin’s physiological effects. Here, we studied the effect of Allicin on post-translational thiol-modification in human Jurkat cells using shotgun LC-MS/MS analyses. We identified 332 proteins in the human Jurkat cell proteome that were modified by S-thioallylation which causes a mass shift of 72 Dalton on cysteines. Many S-thioallylated proteins are high abundant proteins, including the cytoskeletal proteins tubulin, actin, cofilin, filamin and plastin-2, the heat shock chaperones HSP90 and HSPA4, the glycolytic enzymes GAPDH, ALDOA, PKM as well the protein translation factor EEF2. We confirmed that Allicin disrupted the actin cytoskeleton in mouseL929 fibroblasts. Allicin further resulted in Zn2+ release from proteins and stimulated the Zn2+-dependent IL1-triggered release of IL2 in murine EL4 T-cells. Allicin further caused inhibition of enolase activity. In conclusion, our study revealed the overall extent of widespread S-thioallylation in the human Jurkat cell proteome after allicin exposure which affects essential cellular functions of selected allicin targets.

Translatome and transcriptome profiling of FK866 response in Jurkat cells

Project description:We treated Jurkat cells for 48 hr with a sublethal dose of FK866 (5 nM) and DMSO (Mock, control treatment). RNA samples for microarrays derived from fractionated samples by sucrose gradient (sub-polysomes, polysomes), giving us the chance to perform an analysis among polysomal/subpolysomal distribution in treated or untreated cells and the possibility to identify the multi-level gene expression regulation effects of FK866. We are interested to find differentially expressed genes, in the early phase of cell response to FK866, and genes that account for a specific post-transcriptional regulation exerted by the cell in response to the drug. Keywords: polysomal profiling, translatome profiling, polysomal RNA, subpolysomal RNA, translational profiling, polysome profiling, post-transcriptional regulation, FK866, translational efficiency. Gene expression signals derived from the polysomal and subpolysomal RNA populations were compared by microarrays analysis to those obtained from total RNAs (derived from the sum of all the fractions in the polysomal gradient). Polysomal RNA, subpolysomal RNA and total RNA were isolated from Jurkat cells treated with FK866 5 nM or DMSO (mock, control treatment) for 48 hr. Cells lysates were collected from control cells (mock) and from treated cells (FK866). All experiments were run in biological quadruplicates.

Project description:Soybean (Glycine max, cv Williams82) leaf petiole explants exposed to 25 ul/l ethylene for 0 to 72 h. Explants were prepared from 21 day-old greenhouse grown plants. Leaf abscission zones (LAZ) consisted of 2 mm of tissue collected below the leaf blade. The petioles (NAZ) consisted of approximately 3 to 4 mm of petiole tissue with the AZ removed. Explants and tissue were collected in February, March and April of 2013. Tissue and RNA were collected at USDA, Beltsville, MD (Mark L Tucker, Joonyup Kim and Ronghui Yang). Library construction and sequencing was completed at Univ of Cornell, Itheca, NY using a Illumina HiSeq 2000 (James J Giovannoni and Zhangjun Fei).

Genome-wide analysis of histone modifications in Jurkat cells upon ELF-MF exposure

Project description:To address whether ELF-MF influences the epigenetic landscape in the T cell lymphoma cell line Jurkat , we exposed Jurkat cells in the exponential growth phase to ELF MF or a sham control for 72 h, applying intermittently (5’ on/10’ off) a 50 Hz sine ELF-MF at a flux density of 1 mT. Additionally, cells were treated with the histone deacetylation inhibitor trichostatin A (TSA) at a subtoxic dose of 10 nM. Genome-wide profiling of active and repressive histone modifications did not reveal significant alterations. However, ELF-MF exposure has an influence on the robustness of the epigenetic landscape in leukaemic cells. Our data suggests that ELF-MF has a stochastic effect on the epigenetic landscape of individual cells.
