Gene expression profiles from regressive liver fibrosis mice models

ABSTRACT: C57BL/6J mice (male, eight weeks) were intraperitoneally injected with CCl4 in mineral oil twice a week for 6 weeks; control mice only received isometric mineral oil for 6 weeks. Peak liver fibrosis mice subsequently underwent spontaneous recovery for another 1, 3 or 6 weeks after the cessation of CCl4 intoxication. Livers were harvested for RNA sequencing. Total RNA of mouse liver tissue was isolated using an RNA simple Total RNA kit. Poly(A) mRNAs were enriched using magnetic oligo (dT) beads, followed by RNA sequencing library preparation using the NEBNext® Ultra RNA Library Prep Kit. A 125 bp paired-end run was performed on the Illumina HiSeq2500 platform. Clean reads were obtained by perl script, and then mapped to mouse reference genome GRCm38 using Bowtie2 and HISAT algorithms. Gene expression levels were measured using FPKM values by the Cufflinks software.
