Brg1 controls stemness and metastasis of pancreatic cancer through regulating hypoxia pathway

ABSTRACT: Brg1 is a subunit of SWI/SNF chromatin remodeling complex. To clarify the role of Brg1 for the progression and metastasis of pancreatic ductal adenocarcinoma (PDAC), we generated a PDAC mouse model with a dual recombinase system (Brg1 lox/lox; Pdx1-Flp; Kras FSF-G12D; Trp53 frt/frt; Rosa26 FSF-CreERT2) which enabled to ablate Brg1 in established PDAC with tamoxifen treatment. We established cancer cell lines derived from PDAC tumors developed in those mice and investigated the effect of Brg1 ablation on PDAC cell proliferation and metastasis. In order to determine the genes expression in Brg1-deleted mouse PDAC cells, we conducted a transcriptomic analysis using microarray using Brg1 wild type and Brg1-deleted mouse PDAC cells.

Project description:Brg1 is a subunit of SWI/SNF chromatin remodeling complex. We compared the transcriptome between these two groups of tumours with the aim to understand their molecular signatures and the relevance to human pancreatic ductal adenocarcinoma subtypes.

Arginase 1 deletion in myeloid cells decreases immune suppression and tumor formation in pancreatic cancer

Project description:We deleted Arginase 1 in myeloid cells in a spontaneous pancreatic cancer mouse model. We sequenced the spontaneous pancreatic tumor generated by an 11-month-old KF mouse (Ptf1aFlpO/+;KrasFrt-STOP-Frt-G12D/+) and an 11-month-old KFCA mouse (Ptf1aFlpO/+;KrasFrt-STOP-Frt-G12D/+;LysMcre;Arg1f/f). Single cell RNA sequencing revealed macrophage reprogramming and increase CD8+ T cell infiltration upon deletion of Arginase 1 in myeloid cells.

Project description:Global mRNA expression profiles of murine primary PDAC cells following JQ1 or SAHA monotherapy as well as JQ1-SAHA combination therapy were collected using Affymetix mouse whole genome array (Mouse Genome 430A 2.0 Array) . Primary PDAC cells isolated from Ptf1aCre/+;Kras+/LSL-G12D;p53lox/lox (Kras;p53) mice were treated either with JQ1 (100 nM) or SAHA (2000 nM) or vehicle 10% (2-Hydroxypropyl)-β-cyclodextrin (Sigma-Aldrich) or as combination therapy with the indicated dosage for monotherapy. Total RNA isolation was performed after 6 hours of treatment. Primary PDAC cells from Ptf1aCre/+;Kras+/LSL-G12D;p53lox/lox (Kras;p53) mice treated either with JQ1, SAHA, vehicle or JQ1-SAHA combination were analyzed by global gene expression analysis.

Project description:Analysis of gene expression in human embryonic kidney cells HEK293 overexpressing the transcription factors HNF4a2 or mutant forms HNF4a2 C106R and HNF4a2 R154X, which were introduced by FRT/FLP recombination. The analysis was performed 24 hours following 1µg/ml tetracycline treatment to induce the expression of the genes. Results identify HNF4a regulated genes in kidney cells being several of these genes deregulated in renal cell carcinoma. Project description:Identification of genes regulated by the transcription factor HNF4a2 Experiment Overall Design: We used microarrays to identify genes regulated by the transcription factor HNF4a2. HEK293 cell lines containing doxycyclin-inducible wilt type or mutant forms of HNF4a2m which were introduced by FRT/FLP mediated recombination into FLP-In T-REx 293 cells (Invitrogen) were treated for 24 hours with 1 Âµg/ml doxycyclin. Gene expression profiles of induced and noninduced cells were compared to identify HNF4 regulated targets.

Expression data from Dicer WT and Dicer KO mouse CD4 T cells.

Project description:miRNA regulate gene expression at the post-transcriptionnal level. To gain further insight into this process, we analysed by Affymetrix microarray, the transcriptome of Dicer WT or Dicer deleted mouse CD4 T cells. Mouse CD4 T cells were sorted by flow cytometry from control CD4Cre neg Dicer lox/lox or CD4Cre pos Dicer lox/lox animals, total RNA was extracted using RNA bee and amplified before hybridization onto Mouse Gene arrays. Three biological replicate were analysed for each condition (3 Dicer WT control and 3 Dicer KO).
