Project description:Physical activity is essential for maternal health during pregnancy, with WHO guidelines recommending 150 minutes of moderate-intensity aerobic exercise per week. Despite the established benefits, only 35% of pregnant women meet these guidelines, often due to fatigue, lack of time, and pregnancy discomfort. High-Intensity Interval Training (HIIT), characterized by short bursts of intense exercise followed by recovery periods, offers a time-efficient alternative that has been shown to be safe and enjoyable for both mother and fetus. This study aims to analyze the changes induced by a HIIT program in the urine proteome of pregnant women using the SWATH-MS technique, providing insights into molecular processes influenced by exercise.We observed significant alterations in the urinary proteome after a single HIIT session, with a higher scale of differentially expressed proteins compared to after 8 weeks of regular HIIT training. Notable changes included proteins involved in calcium ion binding, enzyme inhibitory activity, and immune system pathways, particularly neutrophil degranulation. Proteins such as ARSA, NPC2, MME, and CD177 showed significant regulation, indicating metabolic adaptations and immune responses. The observed downregulation of proteins like TFF2, associated with insulin resistance, suggests potential benefits of HIIT in preventing gestational diabetes.Our findings demonstrate that while a single session of HIIT induces immediate metabolic and immune changes, regular HIIT leads to adaptations, reflected in a decreased number of altered proteins over time. This research provides a novel proteomic perspective on the impact of HIIT during pregnancy, underscoring its potential to improve maternal health through targeted metabolic and immune system adaptations. Further studies are needed to confirm these findings and explore the long-term benefits of HIIT for pregnant women.

Project description:The microRNA (miRNA) expression profile of plasma exosome in pregnant women complicated with gestational diabetes mellitus (GDM) has not been fully clarified. In this study, differentially expressed miRNAs in plasma exosomes were identified by high-throughput small RNA sequencing in 12 GDM and 12 normal glucose tolerance (NGT) pregnant women and validated in 102 GDM and 101 NGT pregnant women. A total of 22 exosomal miRNAs were found and five of them were verified by qRT-PCR. Exosomal miR-423-5p was upregulated, while miR-99a-5p, miR-192-5p, miR-148a-3p, and miR-122-5p were downregulated in pregnant women complicated with GDM.

Project description:Lactate, a major exercise-derived metabolite, has been proposed to couple cellular metabolism to gene regulation, yet its direct impact on skeletal muscle remains unclear. Male mice underwent six-week interventions (Control, lactate, HIIT, MCT1/2 inhibition, or HIIT+inhibition). We profiled gastrocnemius DNA methylation, mRNA-seq and miRNA-seq, and assessed signaling proteins, metabolites, and running performance. Lactate and HIIT each induced broad, site-specific remodeling of the methylome and transcriptome with substantial overlap at promoter CpGs and among DEGs. Promoter methylation changes showed weak coupling to steady-state mRNA, whereas integrative analyses revealed robust anti-directional miRNA–mRNA networks and enriched for chromatin/epigenetic regulators, identifying a lactate-driven miRNA axis as a proximate regulator of transcriptional output. At the protein level, lactate selectively increased TET2 and DNMT3A and activated PAX7, VEGF, and AKT–S6 signaling, consistent with integrative miRNA–mRNA findings. HIIT increased TET1/2 and DNMT3A with DNMT3B reduction and uniquely enhanced mitochondrial/antioxidant signaling. Blocking MCT1/2 abrogated HIIT-induced methylome and miRNA remodeling and blunted transcriptomic and protein adaptation, demonstrating that intact lactate flux is required for exercise-evoked epigenetic and transcriptomic reprogramming. Despite molecular convergence, chronic lactate did not improve running performance, indicating that lactate is necessary but not sufficient for the full physiological adaptation of training. These findings position a lactate–miRNA axis linking metabolic perturbation to epigenetic and transcriptional remodeling in skeletal muscle.

Methods: This study prospectively enrolled 71 patients who underwent liver biopsy at an academic hospital in Korea. Exosomes were extracted from serum samples, followed by next-generation sequencing (NGS) of miRNAs and targeted real-time quantitative polymerase chain reaction. A model was derived to discriminate advanced fibrosis based on miRNA levels and the performance of this model was evaluated. Validation of the effect of miRNA on liver fibrosis in vitro was followed. Methods: This study prospectively enrolled 71 patients who underwent liver biopsy at an academic hospital in Korea. Exosomes were extracted from serum samples, followed by next-generation sequencing (NGS) of miRNAs and targeted real-time quantitative polymerase chain reaction. A model was derived to discriminate advanced fibrosis based on miRNA levels and the performance of this model was evaluated. Validation of the effect of miRNA on liver fibrosis in vitro was followed. Results: NGS data revealed that exosomal miR-660-5p, miR-125a-5p, and miR-122 expression were changed significantly with the progression of liver fibrosis, of which miR-122 exhibited high read counts enough to be used as a biomarker. The level of exosomal miR-122 decreased as the pathologic fibrosis grade progressed and patients with biopsy-proven advanced fibrosis had significantly lower levels of exosomal miR-122 (P < 0.001) than those without advanced fibrosis. Exosomal miR-122 exhibited a fair performance in discriminating advanced fibrosis especially in combination with fibrosis-4 score and transient elastography. In a subgroup of patients with a non-viral etiology of liver disease, the performance of exosomal miR-122 as a biomarker was greatly improved. Inhibition of miR-122 expression increased the proliferation of the human hepatic stellate cell line, LX-2, and upregulated the expression of various fibrosis related proteins. Conclusion: Exosomal miR-122 may serve as a useful non-invasive biomarker for liver fibrosis, especially in patients with non-viral etiologies of chronic liver disease.

Project description:This 12-week, exercise study will assess the feasibility of a home-based high-intensity interval training (HIIT) program among colorectal cancer survivors and explore the impact of home-based HIIT compared to a standard home-based moderate-intensity continuous aerobic exercise program on physical outcomes linked with survival from colorectal cancer and surrogate blood markers of colorectal cancer recurrence.
HIIT is a type of aerobic exercise that includes short bursts (i.e. 1-4 minutes) of vigorous exercise followed by longer periods of moderate to lower intensity exercise (i.e. 1-10 minutes). Participants in this study will be randomly assigned into a personalized home based exercise program - either high-intensity interval training (HIIT) or moderate-intensity continuous aerobic exercise (MICE).
This pilot study will provide us with preliminary evidence for a larger trial aimed to compare the effectiveness of these two different types of home-based exercise programs on physical outcomes linked with survival, quality of life, and surrogate blood markers of colorectal cancer recurrence.

Project description:The molecular transducers of benefits from different exercise modalities remain incompletely defined. Here we report that 12 weeks of high-intensity aerobic interval (HIIT), resistance (RT), and combined exercise training enhanced insulin sensitivity and lean mass, but only HIIT and combined training improved aerobic capacity and skeletal muscle mitochondrial respiration. HIIT revealed a more robust increase in gene transcripts than other exercise modalities, particularly in older adults, although little overlap with corresponding individual protein abundance was noted. HIIT reversed many age-related differences in the proteome, particularly of mitochondrial proteins in concert with increased mitochondrial protein synthesis. Both RT and HIIT enhanced proteins involved in translational machinery irrespective of age. Only small changes of methylation of DNA promoter regions were observed. We provide evidence for predominant exercise regulation at the translational level, enhancing translational capacity and proteome abundance to explain phenotypic gains in muscle mitochondrial function and hypertrophy in all ages.

Project description:Renal interstitial fibrosis (RIF) is a common pathological feature and final manifestation of chronic progressive kidney disease. Although exosome-mediated intercellular communication is known to play a crucial role in RIF, the mechanism by which injured tubular epithelial cells (TECs) contribute to fibrogenesis remains incompletely understood. In this study, we investigated the role of exosomal miR-122-5p derived from TECs in modulating fibroblast activation and renal fibrosis. Exosomes were isolated from kidneys of unilateral ureteral obstruction (UUO) mice and from TGF-β1-stimulated HK-2 cells. These exosomes were either co-cultured with fibroblasts (NRK-49F cells) or injected into UUO mice via the tail vein. High-throughput miRNA profiling was used to identify differentially expressed miRNAs in exosomes derived from HK-2 cells, and miR-122-5p was selected for further investigation based on expression level and bioinformatic prediction. Functional analyses were performed using miRNA mimics, inhibitors, and target validation assays. The results showed that exosomal miR-122-5p was significantly downregulated in both fibrotic kidneys and TGF-β1-induced HK-2 cells. In vivo and in vitro, restoration of miR-122-5p levels markedly attenuated renal fibrosis, as evidenced by the reduction of fibrotic markers including α-smooth muscle actin, fibronectin, and collagen I. Mechanistically, miR-122-5p was found to directly target hypoxia-inducible factor 1-alpha (HIF-1α), thereby inhibiting activation of the TGF-β1/Smad signaling pathway. These findings suggest that decreased expression of miR-122-5p in TEC-derived exosomes promotes renal fibrosis through the HIF-1α/TGF-β1/Smad axis, while reintroduction of miR-122-5p can effectively reverse these effects. Taken together, our study provides new insights into the intercellular communication between TECs and fibroblasts via exosomal miRNAs, and identifies miR-122-5p as a potential therapeutic target for the treatment of renal interstitial fibrosis.
