G Protein-coupled Receptor 91 (GPR91) Indicates Prognosis and Enhances Chemoresistance by Regulating DNA Damage Pathway and ERK Signaling Pathways in Neuroblastoma

ABSTRACT: This study first reveals that GPR91 is a potential oncogene in neuroblastoma, which is significantly correlated with poor prognosis and significantly represses the sensitivity to chemotherapeutic drugs including doxorubicin and cisplatin of NB cells. It is regulated by GPR91 though the DNA damage pathway and the ERK signaling pathway.

TNFAIP8 promotes AML chemoresistance by activating ERK signaling pathway through interaction with Rac1

Project description:The tumor necrosis factor induced protein 8 (TNFAIP8), a novel anti-apoptotic molecule, has been implicated in chemoresistance of several types of neoplasms, but its role in AML remains to be elucidated. In this study, upregulated TNFAIP8 expression was found in human AML patients and cell lines. E74 like ETS transcription factor 1 (ELF1) was identified to contribute to its aberrant expression. Forced expression of TNFAIP8 protected cells from apoptosis induced by chemotherapeutic agents and increased drug resistance. Importantly, the leukemia-promoting action of TNFAIP8 was mediated by sustaining activity of the extracellular signal-regulated kinase (ERK) signaling pathway through an interaction with Rac family small GTPase 1 (Rac1). In vivo experiments showed that TNFAIP8 suppression lowered leukemia infiltration and improved survival. Taken together, our data provide a molecular basis for the role of TNFAIP8 in chemoresistance and progression of AML and highlight the unique function of TNFAIP8 as an attractive therapeutic target.

Untargeted LC-HRMS Based-Plasma Metabolomics Reveals 3-O-Methyldopa as a New Biomarker of Poor Prognosis in High-Risk Neuroblastoma

Project description:Neuroblastoma (NB) is the most common extracranial malignant tumor in children. Although the survival rate of NB has improved over the years, the outcome of NB still remains poor for over 30% of cases. A more accurate risk stratification remains a key point in the study of NB and the availability of novel prognostic biomarkers of 'high-risk' at diagnosis could help improving patient stratification and predicting outcome. In this paper we show a biomarker discovery approach applied to the plasma of 172 NB patients. Plasma samples from a first cohort of NB patients and age-matched healthy controls were used for untargeted metabolomics analysis based on high-resolution mass spectrometry (HRMS). Differential expression analysis highlighted a number of metabolites annotated with a high degree of identification. Among them, 3-O-methyldopa (3-O-MD) was validated in a second cohort of NB patients using a targeted metabolite profiling approach and its prognostic potential was also analyzed by survival analysis on patients with 3 years follow-up. High expression of 3-O-MD was associated with worse prognosis in the subset of patients with stage M tumor (log-rank p < 0.05) and, among them, it was confirmed as a prognostic factor able to stratify high-risk patients older than 18 months. 3-O-MD might be thus considered as a novel prognostic biomarker of NB eligible to be included at diagnosis among catecholamine metabolite panels in prospective clinical studies. Further studies are warranted to exploit other potential biomarkers highlighted using our approach.

DIA LC-MSMS of MCF-7 Breast Cancer Cells

Project description:The breast cancer cell line MCF-7 was treated with doxorubicin. Protein expression differences between treated and non-treated cells were semiquantitatively determined using the precursor acquisition independent from ion count (PAcIFIC) mass spectrometric method. The acquired proteomic data sets were searched for regulated Reactome pathways and Gene Ontology annotation terms using a new algorithm (SetRank). Using this approach, we described significantly changed pathways (p-value of 0.05 or less) already known to be influenced by chemotherapeutic drugs, such as chromatin organization, DNA binding, embryo development, condensed chromosome, sequence-specific DNA binding, response to oxidative stress and response to toxin. Additionally, we found pathways such as central nervous system neuron differentiation, neuron projection membrane and SNAP receptor activity explaining side-effects of doxorubicin chemotherapy, characterized as ‘chemo brain’, on a molecular level.
