Project description:Stem cell-derived β (SC-β) cells are an emerging regenerative therapy to compensate for loss of functional β cell mass in diabetes. Glucose-stimulated insulin secretion in SC-β cells is variable in vitro but stabilizes after transplantation and maturation under the kidney capsule of mice. We identified mechanisms correlated with functional maturation using RNA-sequencing and co-expression network analysis. In vivo maturation enhanced glucose-stimulated but not basal insulin secretion, up-regulated β cell hormones IAPP and ADCYAP1, increased expression of maturation markers MAFA, UCN3, and SIX2, and resolved endocrine identity of incompletely specified polyhormonal cells produced during differentiation. Transplantation promoted calcium signalling, induced exocytotic machinery supporting hormone secretion and improved stimulus-secretion coupling that fine-tunes insulin secretion. Growth hormone signalling emerged as candidate driver of in vivo maturation and was confirmed in vitro. Also, a large co-expression module correlated with HbA1c and was enriched in genes up-regulated during in vivo maturation but down-regulated in hyperglycaemic and palmitate stress conditions, suggesting that transcriptional maturation of SC-β cells in vivo mirrors processes lost in diabetic β cells.

Project description:Transcriptional profiling of HeLa cells comparing control untreated HeLa cells with IFN-gamma-treated HeLa cells To infect host cells, many viruses use small cellular compartments, called endosomes, for entry, and the thiol/disulfide interchange in viral envelope glycoproteins (Envs) is crucial for infection. By screening cysteine-reacting chemicals, we found a compound, 4,4’-dithiopyridine, which is active at acidic pH, efficiently restricts retrovirus vector infection. We thus hypothesized that some products of endosome-localized, interferon-stimulated genes (ISGs) exhibit anti-viral activity by inhibiting thiol/disulfide interchange in viral Envs. Among the hundreds of ISGs, gamma-interferon (IFN)-inducible lysosomal thiolreductase (GILT) is the only molecule that resides in the endosomes/lysosomes and digests the S-S bonds of proteins under acidic conditions. We now report that GILT significantly inhibits the replication of retroviruses, including HIV-1 and MLV, by restricting both the early and late phases of their life cycles. Using the VSV-G pseudotyped HIV-1 model, we found that GILT digests the S-S bonds of viral Env proteins. GILT also inhibits HIV-1 viral release by digesting the S-S bond of CD63, an endosome-localized molecule reportedly involved in HIV-1 particle release. The effect of -IFN on HIV-1 is limited, although GILT induced by gamma-IFN is supposed to have anti-HIV-1 activity. We found that while gamma-IFN effectively inhibits MLV replication through GILT, the gamma-IFN signaling is remarkably inhibited by the HIV-1 Env protein, but not the MLV Env protein. These findings suggest that GILT functions as an anti-viral host factor induced by gamma-IFN, however, HIV-1 may have evolved to inhibit gamma-IFN signaling by the Env protein.

Project description:This microarray study aimed at identifying the differences in the global gene expression growth program of adult cultured olfactory ensheathing cells (cOEC) (from the olfactory bulb) versus adult cultured Schwann cells (SC) (from the sciatic nerve) and versus adult OEC directly dissected from the olfactory nerve layer (nOEC). The aim of the comparison between cOEC and SC is to define intrinsic molecular differences that distinguish cOEC from SC (both cell types support neuronal regeneration). The aim of the comparison between cOEC and nOEC is to determine the transcriptional responses that are induced in OEC during culturing. Three-condition experiment: cOEC vs. SC, cOEC vs. nOEC, SC vs. nOEC. Biological replicates: 3 cOEC-SC replicates, 3 cOEC-nOEC replicates, 3 SC-nOEC replicates. A factorial design was used consisting of direct comparisons between the three cell types (Glonek and Solomon, 2004 (PMID 14744830).

Project description:To study immune responses in the context of human allogeneic graft rejection, we chose the Hu-PBL-NSG-MHCnull humanized mouse (Brehm et al., 2019). NOD-scid IL-2 receptor subunit γ (IL2rg)null (NSG) immunocompromised mice that lack murine MHC class I and II, were transplanted (under the kidney capsule, n=12) with 5M SC-islets (HLA-A2 positive), followed by human PBMC injection (termed ‘hPi-mice’; 50M/mouse, n=6) from healthy unmatched donors (HLA-A2 negative). The lack of murine MHC allowed us to monitor the graft function for prolonged durations without the risk of xenogeneic graft vs host disease (GVHD). Half of the SC-islet transplanted cohort (n=6 mice) was used as the control, without PBMC injection (Figure 1A). Since graft elimination by PBMCs is incomplete and residual endocrine cells remain in the hPi-mice grafts, we retrieved the SC-islet grafts for single cell RNA sequencing (scRNA-seq) analysis. These samples, along with pre-injected PBMCs as controls, were used for 10x Genomics mRNA expression library preparation and Illumina sequencing.
