An investigation into the effects of ATV:Trem2 on AD model mice using scRNA-seq

Project description:WT hTfR-KI mice and APP/SAA hTfR-KI mice were treated with one of: Isotype control ATV, naked 4D9 (activating Trem2 Ab), or ATV:4D9. Drugs were delivered IV. After 24 hours, mice were sacrificed, and perfused cortices were processed into single-cell suspensions. The suspensions were FACS sorted for Cd45+/Cd11b+ cells. Single-cell suspensions were labeled with CellPlex CMO per 10X Genomics protocol (CG000391 Rev A.) After each sample was labeled with CMO tags, equal numbers of cells per sample were combined into sample pools for single cell capture on the Chromium using Chromium Next GEM Single Cell 3’ v3.1 kit. After RT and cDNA amplification, each sample pool was separated into one gene expression (GE) library and one CMO tag library via dual-sided SPRI bead purification. Library preparation was performed per manufacturer’s protocol (CG000388 Rev A)

Microglia regulate cortical remyelination via ΤNFR1-dependent phenotypic polarization [scRNA-Seq]

Project description:Microglia are strongly implicated in demyelinating neurodegenerative diseases with increasing evidence for roles in protection and healing, but the mechanisms that control CNS remyelination are poorly understood. Here we show that microglia-specific deletion of TNFR1 and pharmacological inhibition of soluble TNF (solTNF) or downstream IL-1R allow maturation of highly activated disease-associated microglia with increased size and myelin phagocytosis capacity that accelerate cortical remyelination and motor recovery. Single cell transcriptomic analysis of cortex at disease onset reveal that solTNF inhibition enhances reparative IL-10-responsive, while preventing damaging IL-1-related signatures of disease-associated microglia. Longitudinal brain transcriptome analysis through disease reveal earlier recovery upon therapeutic loss of microglia TNFR1. Functional relevance of microglia inflammatory polarization pathways for disease is validated in vivo. Furthermore, disease-state microglia producing downstream IL-1/IL-18/NLRP3/CASP1 targets are identified in human demyelinating lesions. Overall, redirecting disease microglia polarization by targeting cytokines is a potential approach for improving CNS repair in demyelinating disorders.

Neuro-immune organoid model reveals a role of microglia in cell stress response and emergence of neural activity

Project description:Purpose: Many fundamental questions about microglia diversity and function during development remain unanswered as we lack experimental strategies to interrogate their interactions with other cell types and responses to perturbations ex vivo. Methods: By using 10X Genomics scRNAseq, we generated transcriptomic data across different microglia models. Additionally, we performed microglia engraftment into cortical brain organoids and analyzed molecular changes in different cell types occurring in response to the presence of microglia. Results: We compared human microglia states across culture models, including cultured primary microglia and pluripotent stem cell-derived microglia and developed a ‘report card’ of gene expression signatures differentially recruited and attenuated across these distinct experimental models.

Resulting cell suspension from Ncx1-/- and Ncx1+/+ embryos loaded onto independent channels of a Chromium chip before single cell partitioning and barcoding on Chromium Controller (10x Genomics).
