Bulk RNA-seq of mice synovium

ABSTRACT: Purpose: Joint function is impaired by disuse. We investigated mice synovium and cartilage for control and MMS (minimized mechanical stress) model mice. Method: We developed a MMS model which could represent the disused joint by combining non-weight bearing with tail suspension and knee joint immobilization with a plastic cylinder. We followed the time course of MMS model for day 3,7,14 for synovium and day 3,7,14,28 for cartilage. In addition, we used two models to stop the immobilization; in R1, only the joint immobilization was canceled at 2 weeks, but tail suspension was continued; in R2, both the joint motion and weight bearing were allowed at 2 weeks. Conclusion: MMS synovium expressed many secreted proinflammatory cytokines. MMS cartilage expressed less cartilage anabolic genes.

Single-cell sequencing of murine articular cartilage

Project description:Osteoarthritis (OA) is a degenerative joint disease characterized by progressive cartilage loss, bone remodeling, synovial inflammation, and significant joint pain, often resulting in disability. Injury to the synovial joint such as the anterior cruciate ligament (ACL) tear is the major cause of OA in young adults. Currently, there are no approved therapies available to prevent joint degeneration or rebuild articular cartilage destroyed by OA, primarily because our understanding of the cellular and molecular changes that contribute to joint damage is very limited. The synovial joint is a complex structure composed of several tissues including articular cartilage, subchondral bone, synovium, synovial fluid, and tensile tissues including tendons and ligaments. In the present study, using single-cell RNA sequencing (scRNA-seq), we examined the cellular heterogeneity in articular cartilage from mouse knee joints and determined the knee joint injury-induced early molecular changes in the chondrocytes that could contribute to OA.

Characterising the effect of age and sex on post-transcriptional regulation in synovial joint tissues

Project description:Our analysis determined the effect of age on polyadenylation site usage in joint tissues. Using total RNA purified from articular cartilage or synovium, we performed QuantSeq-Rev 3’ end RNA sequencing to identify polyadenylation sites utilised in all transcripts. This 3’ end sequencing method reads 3’ to 5’, starting in the polyA tail of the transcript, and so every read identifies a polyadenylation site. We then utilised PolyAminer software, with adaptations made to the code to identify and mask genomic polyA regions so that they would contribute to the site comparisons following read alignment. We found evidence that age affected polyadenylation site usage in both of these tissues

Project description:Osteoarthritis (OA) affects all tissues in the knee joint but therapeutic targets have often been selected for their role in cartilage damage and inflammation and largely omitted consideration of mechanisms that are mediating meniscus damage and more importantly there have been insufficient efforts to determine whether pathogenic processes are shared between tissues. Several scRNA-seq analyses have been performed in OA knees but have been largely restricted to the analysis of a single joint tissue of articular cartilage, or meniscus with the exception of one study that analyzed cartilage and synovium. Here, we performed scRNA-seq analyses of healthy and OA-affected human knee cartilage and menisci to interrogate separate and shared mechanisms in cellular homeostasis and OA pathogenesis.

Cholesterol metabolism regulates cartilage metabolic-inflammatory crosstalk by triggering osteocyte mitochondria transfer as a therapeutic target for osteoarthritis

Project description:Osteoarthritis (OA) is considered as a metabolic related inflammatory disease in the joint affecting both cartilage and subchondral bone. Cholesterol metabolism has been implicated in the pathogenesis of OA, yet the underlying mechanism still remains elusive. Understanding the role of cholesterol metabolism in the joint inflammation will provide treatment alternatives of OA. Here, we found that the disordered cholesterol metabolism in joint do not directly target cartilage and synovium but target subchondral bone by transferring osteocyte mitochondria to amplify inflammatory phenotype in the cartilage. In detail, Nudt8 in osteocyte mitochondria serves as a metabolic-inflammatory switch point to alter cholesterol metabolism by degrading coenzyme A (CoA) and further increase the cytosolic mitochondrial DNA (mtDNA) stress to activate cGAS-STING pathway in chondrocytes, resulting in exacerbated inflammation and OA severity. Inhibiting the transfer of osteocyte mitochondria ameliorated joint inflammation and OA progression. Pharmacological targeting osteocyte mitochondria regulated cholesterol metabolism by supplementation of pantethine - an important precursor metabolite of CoA - ameliorated osteocyte mitochondria activated cGAS-STING pathway and OA phenotype in chondrocytes, and subsequent improved the joint damage and reduced painful behavior in OA mice. Targeting cholesterol-mitochondria regulated metabolic-inflammatory crosstalk represents a promising treatment strategy for OA.

Decreased chondrocyte features of articular cartilage during short-term repeated overloading: a synovium-cartilage interaction via inhibin subunit beta A

Project description:The primary goal of this study is to investigate the ultrashort-term responses and molecular mechanisms in both the synovium and cartilage under conditions of excessive mechanical loading, a major contributing factor in the pathogenesis of osteoarthritis (OA). While cartilage degeneration is a hallmark of OA, the involvement of the synovium in disease progression and pain, particularly in relation to synovitis, has been increasingly recognized. Despite this, the immediate molecular changes in both tissues following mechanical stress, and the interactions between them, remain unclear. To address these gaps, we developed a mouse model that simulates excessive mechanical loading through treadmill exercise combined with weight attachment. We investigated the ultrashort-term gene expression changes in the synovium and cartilage at 1, 4, and 24 hours post-exercise using RNA sequencing (RNA-seq). Additionally, we examined the effects of repeated overloading by analyzing tissue samples from mice subjected to four consecutive days of overloading. This approach allowed us to capture both the acute and repeated stress responses in these tissues. Through transcriptomic analysis, we aimed to identify key molecular mediators driving both the rapid transcriptional changes in each tissue and the synovium-cartilage interaction.
