RNA-seq of MV4;11 cells treated with Methyl Bestatin or Brefeldin A

ABSTRACT: We report the application of RNA-seq for profiling gene transcription upon treatment of methyl bestatin (MeBs, aminopeptidase inhibitor) or brefeldin A (BFA, ER-to-Golgi transport inhibitor), compared to DMSO control. We found MeBs and BFA similarly upregulated genes pertinent to proteotoxic stress.

Whole genome sequencing of brefeldin A adaptors in Cryptococcus neoformans

Project description:Cryptococcus neoformans lab strain H99 was grown in YPD broth supplemented with 16ug/ml brefeldin A for 72h at 30C. We used 3 biological repeats. Then the culture was washed and diluted with distilled water, then plated on YPD plate. From each culture, we randomly tested 120 colonies to see if they obtained tolerance to brefeldin A. In total, we got 30 tolerant adaptors. We sequenced the adaptors as well as the parent.

Project description:In Candida albicans, the lab strain SC5314 was grown in YPD broth supplemented with 4ug/ml brefeldin A for 24h. The culture was washed, diluted and spread on YPD. Randomly 120 colonies were tested for tolerance to brefeldin A. Two adaptors (FY200 and FY201) obtained tolerance. These two adaptors were sequenced.
