Genome-wide analysis of gene expression during differentiation in C2C12 cells


ABSTRACT: Investigate the genome-wide gene expression profiles of 50% and 95% confluent C2C12 myoblasts and C2C12 myotubes differentiated for 24 and 48 hours. Overall design: Triplicate total RNA samples were obtained from 50% and 95% confluent C2C12 myoblasts, and C2C12 myotubes differentiated for 24 and 48 hours.

Recent studies have demonstrated that MyoD initiates a feed-forward regulation of skeletal muscle gene expression, predicting that MyoD binds directly to many genes expressed during differentiation. We have used chromatin immunoprecipitation and high-throughput sequencing to identify genome-wide binding of MyoD in several skeletal muscle cell types.
