ABSTRACT: It was our goal to discern the epigenomic signatures of the ventral hippocampus, in male and female mice in a double hit model of acute stress: one hit of acute restraint stress followed by forced swim stress one week later.

Project description:The acute stress response mobilizes energy to meet situational demands and re-establish homeostasis. Here, we used a brief forced swim exposure to trigger a strong stress response in mice. Using multiomic profiling we characterize the stress-induced molecular events in the dorsal (dHC) and ventral (vHC) hippocampus over time, across molecular scales, and down to the level of cell types and single cells.

Gene expression analysis of the hippocampus in Il18+/+ and Il18-/- with restraint stress

Project description:Interleukin-18 (IL-18) was one of the inflammatory cytokines and related to major depressive disorder (MDD) and dementia. We previously investigated IL-18 knockout (Il18−/−) mice developed impaired learning and memory, more anxiety, and less motivation comparing to Il18+/+ mice. In the Il18−/− mice with acute stress, both the immobility time in the forced swim test and the tail suspension test were decreased, even though no difference was observed in the Il18+/+ mice. To reveal the mechanism, RNA-Seq was performed.

Project description:Glucocorticoid hormones (GCs) play a critical role in physiological regulation and behavioural adaptation through mineralocorticoid (MR)- and glucocorticoid receptor (GR)-mediated actions in the hippocampus. We conducted genome-wide MR and GR ChIP-Seq studies on rat hippocampus to elucidate MR- and GR-regulated genes under physiological conditions like acute stress and circadian variation. Rat hippocampus tissue was collected under early morning baseline conditions, at 30 min after the start of a 15-min forced swim session (acute stress), or under late afternoon baseline conditions. We identified many genomic loci in which MR and/or GR binds following acute stress or due the circadian rise in GC secretion.
