Expression profiling of liver tissue from C57BL6 x DBA strain

ABSTRACT: Expression profiling of liver tissue from 111 F2 female mice resulting from a cross between C57BL/6J and DBA/2J inbred mouse strains. Mice were feed a chow diet for 12 months followed by 4 months on an atherogenic high-fat, high-cholesterol diet. Parental and F2 mice were killed at 16 months of age. The livers were removed immediately and flash frozen for expression profiling. See Schadt, et al., Nature , Vol 422(6929):297-302 for details. Project description:Gene transcription in epididymal fat pads was investigated in an F2 cross of 129S6 x Balb/c mice using Illumina gene expression arrays. Expression data was determined in 5 months old male mice fed a carbohydrate diet. Expression data from livers of F2 mice (B57BL/6 X DBA/2) deficient in leptin receptor (db/db)

Project description:In several models of obesity-induced diabetes, increased lipid accumulation in the liver has been associated with decreased diabetes susceptibility. For instance, deficiency in leptin receptor (db/db) leads to hyperphagia and obesity in both C57BL/6 and C57BLKS mice but, only on the C57BLKS background do the mice develop beta-cell loss leading to severe diabetes while C57BL/6 mice are relatively resistant. Liver triglyceride levels in the resistant C57BL/6 mice are 3 to 4 fold higher than in C57BLKS. To better understand the mechanisms contributing to metabolic dysfunction in obesity-induced diabetes, we used microarrays to comprehensively profile gene expression livers of F2 mice (B57BL/6 X DBA/2) deficient in leptin receptor (db/db) DBA/2J females were mated to C57BL/6 males carrying leptin receptor deficiency (db/+) and, F1 (db/+) offspring were interbred to produce F2 mice. Offspring deficient in leptin receptor (db/db) were fed on a chow diet until 5 weeks or 12 weeks of age and then euthanized for collection of liver tissue for RNA profiling along with other diabetes-related phenotypes.

Mouse F2 Cross, Carbohydrate Diet and High Fat Diet, Heart Tissue

Project description:The effect of high fat diet feeding on heart gene transcription regulation was investigated in an F2 cross of 129S6 x Balb/c mice using Illumina gene expression arrays. Expression data was determined in 5 months old male mice fed a high fat diet (40% fat) for 15 weeks. Control mice were fed a standard carbohydrate chow. 96 animals per group were used. The files mouse-f2-chd-genotype.txt and HFD.txt contain genotyping data linked to the diets, they can be found found in the file E-MTAB-401.additional.zip on the FTP for this submission. NOTE: the file CHD.txt was replaced by the updated file mouse-f2-chd-genotype.txt on 8th April 2014.

Project description:The purpose of this experiment was to determine the expression traits in animals from F2 intercross of inbred strains C57BL/6J, C3H/HeJ. (N=309, 164 males and 145 females). Adipose from 295 F2 female and male mice were generated by intercrossing F1 mice. Mice were fed chow diet containing 4% fat (Ralston-Purina Co., St. Louis, MO) until 8 weeks of age and then were placed on a high-fat "Western" diet containing 42% fat and 0.15% cholesterol (Teklad 88137, Harlan Teklad, Madison WI) for 12 weeks. At 20 weeks mice were sacrificed, after a 12-hour fast adipose tissues were dissected and flash frozen in LN2 and stored at -80Â°C. Expression profiling of F2 progenies from C57BL/6J x DBA/2J

Project description:Expression profiling of a cohort of mice was derived from a standard F2 intercross using two standard inbred strains, C57BL/6J and DBA/2J. In this study, we selected 127 male animals that had been raised on a standard rodent chow diet (Purina Chow from Ralston-Purina Co., St. Louis, MO) for eight weeks and then switched to a high fat diet for sixteen additional weeks. At 20 weeks of age, the mice were euthanized and liver tissues were collected and flash frozen in liquid nitrogen and stored at -80 degrees C prior to RNA isolation. All procedures of housing and treatment of animals were performed in accordance with IACUC regulations.

eQTL study of bone marrow derived macrophages and endothelial cells from DBA2xAKR F2 mice

Project description:An eQTL analysis between DBA/2J and AKR/J mice strains on the ApoE null background was done in bone marrow derived macrophages and endothelial cells to identify modifier genes that affect atherosclerosis susceptibility. DBA/2J mice were crossed with AKR/J mice and their offspring crossed with each other to yield an F2 generation. Bone marrow derived macrophages were cultured and gene expression quantified using Illumina's MouseRef-8 v2 arrays in 170 F2 mice for an eQTL study. In addition primary endothelial cells derived from the aorta of 48 male F2 mice were also assayed using arrays for eQTL mapping. Lesion area in the aortic root was also quantified in these samples.
