Single cell RNA-seq of mice synovium

ABSTRACT: Purpose: Joint function is impaired by disuse. We investigated mice synovium for control and MMS(minimized mechanical stress) model mice. Method: We developed a MMS model which could represent the disused joint by combining non-weight bearing with tail suspension and knee joint immobilization with a plastic cylinder. We obtained synovial cells from control and MMS day14 mice. Conclusion: Specific subsets in MMS synovium expressed many secreted proinflammatory cytokines and are thought to be the tragger of cartilage degeneration..

Project description:Purpose: Joint function is impaired by disuse. We investigated mice synovium and cartilage for control and MMS (minimized mechanical stress) model mice. Method: We developed a MMS model which could represent the disused joint by combining non-weight bearing with tail suspension and knee joint immobilization with a plastic cylinder. We followed the time course of MMS model for day 3,7,14 for synovium and day 3,7,14,28 for cartilage. In addition, we used two models to stop the immobilization; in R1, only the joint immobilization was canceled at 2 weeks, but tail suspension was continued; in R2, both the joint motion and weight bearing were allowed at 2 weeks. Conclusion: MMS synovium expressed many secreted proinflammatory cytokines. MMS cartilage expressed less cartilage anabolic genes.

Project description:Knee joint synovium was used for gene expression analysis of mouse collagen induced arthritis (CIA). Synovium was prepared at day 30 after initial sensitization from: healthy controls, CIA animals with no, with mild, with moderate, or with severe joint inflammation. Each sample group is represented by three replicates, each consisting of tissue collected from three to four animals. Experiment Overall Design: The data set consists of 15 samples: five groups with three replicates each. One sample group is from healthy controls, the other groups are from CIA animals with different degress of joint inflammation.

After harvesting the synovium and extraction of RNA, species specific microarray analyses were performed uisng Affymetrix microarrays to investigate the gene expression change of human and rat transcripts after injection.

Single-cell atlas of human infrapatellar fat pad and synovium unravels mechanisms of osteoarthritis-induced joint pathology

Project description:Infrapatellar fat pad (IPFP) and synovium, two joint tissues next to each other, play essential roles in regulating joint homeostasis and the progression of knee osteoarthritis (OA). However, the mechanisms regulating their function under healthy and diseased conditions are largely unknown. This study aims to analyze the cellular heterogeneity of these two tissues from normal and OA patients. Single cell and single nuclei transcriptomic profiling were performed on IPFP/synovium cells from four healthy donors and five OA patients. Subsequent bioinformatic analyses identified cell clusters, delineated differentiation routes of mesenchymal cells and macrophages, and identified novel cell-to-cell communication pathways in joint tissue.

Single cell RNA sequencing on synovium, meniscus and cartilage of rheumatoid arthritis

Project description:Rheumatoid arthritis (RA) induced destruction of knee joints is a common cause of total knee arthroplasty (TKA). Under single cell RNA sequencing generated by 10x Genomics, we identified CD142+ synovial fibroblasts as a novel cluster, located at the sublining layer in normal and osteoarthritis knee synovium, but elevated and distributed at the lining layer in RA knee synovium, and infiltrated in multiple RA joint synovia. Intra-articular injection of CD142+ synovial fibroblasts can quickly and drastically damage the meniscus but has a slight effect on cartilage.Long-term follow-up of the RA cohort indicated that enriched CD142+ synovial fibroblasts in the lining layer was a risk factor for severe joint destruction and eventually underwent TKA. Our results demonstrate that CD142+ synovial fibroblasts can be used as an indicator to assess prognosis and a therapeutic target to inhibit meniscal damage, thereby alleviating RA knee joint destruction.
