Project description:To characterize plant growth in net zero energy greenhouse models with semi-transparent organic solar cell (OSC) filter roofs, RNA was extracted from red oak leaf lettuce leaves grown under 3 OPV filters (FTAZ:IT-M, PTB7-Th:IEICO-4F, FTAZ:PCBM) an clear and shaded glass controls. 2 light intensity experiments were used. PPPFD Controlled (PC) treatments had a constant light intensity and different light spectra created by the filters. Height Controlled (HC) treatments varied in both light intensity and spectra, according to the light transmission of each filter. When light intensity was controlled, plants grown under FTAZ:IT-M had a DEG profile distinct from plants grown under the other 2 filters. Key genes were up or down-regulated that indicate changes in anthocyanin accumulation, nitrogen metabolism and pest defense.

Project description:AtGenExpress: A multinational coordinated effort to uncover the transcriptome of the multicellular model organism Arabidopsis thaliana The activity of genes and their encoded products can be regulated in several ways, but transcription is the primary level, since all other modes of regulation (RNA splicing, RNA and protein stability, etc.) are dependent on a gene being transcribed in the first place. The importance of transcriptional regulation has been underscored by the recent flood of global expression analyses, which have confirmed that transcriptional co-regulation of genes that act together is the norm, not the exception. Moreover, many studies suggest that evolutionary change is driven in large part by modifications of transcriptional programs. An essential first step toward deciphering the transcriptional code is to determine the expression pattern of all genes. With this goal in mind, an international effort to develop a gene expression atlas of Arabidopsis has been underway since fall 2003. This project, dubbed AtGenExpress, is funded by the DFG, and will provide the Arabidopsis community with access to a large set of Affymetrix microarray data. As part of this collaboration, we have generated expression data from 80 biologicaly different samples in triplicate. Stress treatments Seeds of Arabidopsis thaliana Wild Type (col-0) were sown on rafts in Magenta boxes containing MS-Agar-media. After 2 days in the cold room (4°C, dark) the boxes were transferred to the long day chamber. Long day conditions were 16/8 hrs light/dark, 24°C, 50% humidity and 150 uEinstein/cm2 sec light intensity. At day 11 the rafts were transferred in Magenta boxes containing MS-liquid-media. At day 16 stress treatment started at 3 hrs of light period; samples taken at 0.5, 1, 3, 6, 12, 24 h after treatment (in selected indicated cases 0.25h and 4.0h too), control samples include 0h; roots and shoots were prepared separately; all treatments and preparations were done on the same batch of seedlings in one place (Lab J.Kudla/Ulm, Germany) by coworkers from the indicated groups. Project description:Urinary chronic pelvic pain syndrome (UCPPS) is a condition of unknown etiology characterized by pelvic pain, and urinary frequency and/or urgency. As the proximal fluid of this disease, urine is an ideal candidate sample matrix for an unbiased study of UCPPS. In this study, a large, discovery-phase, TMT-based quantitative urinary proteomics analysis of 244 subjects was performed. The subjects included patients with UCPPS (n=82), healthy controls (HC) (n=94) and other chronic pain diseases, termed positive controls (PC) (n=68). Utilizing training and testing cohorts , we identified and validated a small and distinct set of proteins that distinguished UCPPS from HC (n=7) and UCPPS from PC (n=3). Validated UCPPS:HC proteins were predominantly ECM/ECM modifying or immunomodulatory/host defense in nature. Significantly,varying proteins in the UCPPS:HC comparison were overrepresented by members of several dysregulated biological processes including decreased immune cell migration, decreased development of epithelial tissue and increased bleeding. Comparison with the PC cohort enabled evaluation of UCPPS-specific upstream regulators, contrasting UCPPS with other conditions that cause chronic pain. Specific to UCPPS were alterations in the predicted signaling of several upstream regulators, including alpha-catenin, IL6, EGF and TGFB1, among others. These findings advance our knowledge of the etiology of UCPPS and inform potential future clinical translation into a diagnostic panel for UCPPS.

Project description:Bamboo is one of the most important non-timber forestry products in the world. Light is not only the most critical source of energy for plant photosynthesis, but also involved in regulating the biological processes of plants. However, there are few reports on how blue/red light affects moso bamboo. This study investigated the growth status and physiological responses of moso bamboo (Phyllostachys edulis) to blue and red light treatments. The growth status of the bamboo plants was evaluated under different light conditions, revealing that both blue and red light treatments promoted plant height and overall growth compared to the dark treatment. Gas exchange parameters, chlorophyll fluorescence, and enzyme activity were measured to assess the photosystem response of moso bamboo to light treatments. Both blue and red light treatments significantly increased the net photosynthetic rate and stomatal conductance of the bamboo plants. Additionally, the blue light treatment led to higher chlorophyll content and enzyme activities related to photosynthesis and oxidative phosphorylation compared to the red light treatment.
