Characteristic of the transcriptomics in the early subcortical vascular cognitive impairment [miRNA-Seq]

ABSTRACT: Two cohorts with cerebrovascular disease are included in the present study, i.e., SCD group and VCI group. Participants in SVD group has the normal cognitive function and others in VCI group show the worse cognitive function. The whole-blood is collected from each participant, and RNA sequencing is conducted in whole-blood samples. Characteristic of circRNA, lncRNA, miRNA and mRNA are identified in the present blood samples.

Extracellular vesicle proteome of breast cancer patients with and without cognitive impairment following anthracycline-based chemotherapy

Project description:Cognitive impairment due to cancer and its therapy is a major concern among cancer survivors. Extracellular vesicles (EVs) composition altered by cancer and chemotherapy may affect neurological processes such as neuroplasticity, potentially impacting the cognitive abilities of cancer survivors. We investigated the EV proteome of breast cancer patients with and without cognitive impairment following anthracycline-based chemotherapy from longitudinally collected plasma. EVs were cup-shaped and positive for Flotillin-1 and TSG-101. We identified 517 differentially expressed EV proteins between the cognitive impaired and non-impaired groups during and post-chemotherapy. The observed decreased expression of p2X purinoceptor, cofilin-1, ADAM 10, and dynamin-1 in the plasma EVs of cognitive impaired group may suggest alterations in mechanisms underlying synaptic plasticity. The reduced expression of tight junction proteins among cognitive-impaired patients may imply weakening of the blood-brain barrier. These EV protein signatures may serve as a fingerprint that underscore the mechanisms underlying cognitive impairment in cancer survivors.

Determination of the serum miRNA expression profile in human moyamoya disease (MMD)

Project description:Moyamoya disease (MMD) is a cerebrovascular disease characterized by progressive stenosis of the intracranial internal carotid arteries and their proximal branches. However, the etiology of this rare disease remains widely unknown. Serum microRNA (miRNA) profiles have been screened to identify novel biomarkers for disease diagnosis and prognosis. Here, we identified important serum miRNAs that might play important roles in contributing to MMD pathogenesis through microarray analysis. 10 MMD patients and 10 controls were consecutively recruited at Shanghai Changhai Hospital. Five-ml venous blood was collected from each participant and separated into serum and cellular fractions. We pooled serum samples from 10 MMD patients and 10 controls. Agilent Human 8 x 60K miRNA Array was performed on the two pooled samples.

Identification of different miRNAs in Alzheimer's disease and vascular cognitive impairment and dementia

Project description:There are many subtypes of dementia without any minimally invasive, low-cost and efficient diagnostic biomarkers. MicroRNA (miRNA) are easily gained and accessible from patients, which may be a promising biomarker for dementia diagnosis and differential diagnosis. We make a completely expression analysis of serum miRNA from 24 participants including post-stroke cognitive impairment (PSCI) group (n=6), post-stroke non-cognitive impairment (PSCI) group, Alzheimer's Disease (AD) group (n=6) and normal control group (NC) group (n=6). Different expression miRNA (DE -miRNA) were screened by LIMMA ebayes method, Benjamini-Hochberg check and fold change (P < 0.05, |log2FC|≥1), we construct a DE-miRNA-key gene-signaling pathway network, the roles of DE-miRNA and related genes and signaling pathways in AD and PSCI were deduced through comprehensive network construction and literature search.
