Whole transcriptome RNAseq analysis of germ cells in Inhba and Inha fetal testis

ABSTRACT: We performed bulk RNA-Seq on male fetal germ cells isolated from E13.5 and E15.5 testes from activin A-deficient mice (Inhba KO), and from E13.5 testes from mice with greater activin A bioactivity (Inha KO). The aim of this expereiment was to determine how chronic altered activin A bioactivity altered the germline transcriptome.

Project description:We performed RNA-seq on testicular somatic cells from fetal activin A-deficient mice (Inhba KO) and wildtype littermates at embryonic ages E13.5 and E15.5. Activin A deficiency predominantly affects the Sertoli cell transcriptome. New candidate targets include Minar1, Sel1l3, Vnn1, Sfrp4, Masp1, Nell1, Tthy1 and Prss12. Importantly, the testosterone (T) biosynthetic enzymes present in fetal Sertoli cells, Hsd17b1 and Hsd17b3, were identified as activin-responsive. Activin-deficient testes contained elevated androstenedione (A4), displayed an Inhba gene dose-dependent A4/T ratio, and contained 11-keto androgens.

Bulk RNA-Seq of Inha KO mouse placenta exposed to DEHP

Project description:Introduction: In the placenta, activin A affects decidualisation and trophoblast growth and invasion. Levels increase during pregnancy to peak at parturition, while premature elevation is a hallmark of several pregnancy disorders. Here we examine the impact of elevated activin A bioactivity in the inhibin α knockout (Inha KO) mouse placenta. We also investigate whether simultaneous exposure to the endocrine disruptor, diethyl-hexyl phthalate (DEHP), which impairs normal placental function, exacerbates effects on placental morphology and gene expression in this model. Methods: Litters of Inha WT, Het and KO fetuses were collected at E15.5 from time-mated Inha heterozygous pregnant mice randomly assigned to three groups: untreated, corn oil (Vehicle) or DEHP 500 mg/kg/day, from E12.5-14.5 (n=2-17). Gross and histological placental morphology was analysed, and bulk RNA-sequencing conducted. Results: Placentae of Inha WT, Het and KO fetuses exhibited no gross differences, and no differentially expressed genes. However, sex differences included a significant increase in labyrinth area and placental size from WT males compared to WT females. DEHP exposure significantly increased fetal resorption at E15.5 but yielded little differences in placental morphology. Transcript analysis identified sex- and genotype-specific transcript alterations resulting from DEHP treatment. Eight transcripts were altered by DEHP exposure across all genotypes (Cts3, Ceacam13, Gm5155, Ssr4, Sec11c, Psg18, Psg-ps1, Taf7l). Discussion: Despite minimal morphological changes, transcriptomic differences suggest that spongiotrophoblast cells and syncytiotrophoblast giant cells are key targets of DEHP exposure, with CEA family transcripts susceptible to its effects. Additionally, DEHP exposure also modulates gene transcript responses to elevated activin A.

Comparison of granulosa cells from WT and Inha KO mice following gonadotropin stimulation

Project description:Inhibin α knockout (Inha-/-) female mice develop sex cord-stromal ovarian cancer with complete penetrance and previous studies demonstrate that the pituitary gonadotropins [follicle stimulating hormone (FSH) and luteinizing hormone (LH)] are influential modifiers of granulosa cell tumor development and progression in inhibin-deficient females. Recent studies have demonstrated that Inha-/- ovarian follicles develop precociously to the early antral stage in prepubertal mice without any increase in serum FSH and these studies suggested that in the absence of inhibins, granulosa cells differentiate abnormally, and thus at sexual maturity may undergo an abnormal response to gonadotropin signaling. To test this hypothesis, we stimulated immature WT and Inha-/- female mice prior to gross tumor formation with gonadotropin analogs, and subsequently examined post-gonadotropin induced ovarian follicle development, as well as preovulatory and hCG-induced gene expression changes in granulosa cells. We find that at three weeks of age, inhibin-deficient ovaries do not show further antral development nor undergo cumulus expansion. Widespread alterations in the transcriptome of gonadotropin-treated Inha-/- granulosa cells suggest that gonadotropins initiate an improper program of cell differentiation in Inha-/- cells. Overall, our experiments reveal that inhibins are essential for the normal gonadotropin-dependent response of granulosa cells.
