Mouse Lung Response to Stainless Steel and Mild Steel Welding Fume

ABSTRACT: A/J mice are genetically predisposed to spontaneous and/or chemically-induced lung tumors while C57BL/6J (B6) mice are resistant. This genetic disparity provides a unique scenario to identify molecular mechanisms associated with the lung response to welding fume at the transcriptome level.

In addition, the applicability of this methodological approach for determining specific exposure-induced responses was examined. C57Bl/6J mice were exposed to gas metal arc-stainless steel (GMA-SS) welding fume by inhalation for 10 days (3 hours/day) at 40 mg/m3. Microarray analysis was done on lung, blood and aorta from n=4 mice exposed to welding fume or air (control).

Comparative analysis of gene expression in 9 BXH RI strains and progenitors (B6 and C3) and BXHF1.

Project description:We hypothesize that gene expression in the lungs of this RI panel are divergent thus contributing to the disparity in their phenotypes. More specifically, (1) B6 mice tend to have a much smaller lung volume than the C3 mice while the F1 appears to be like the B6 in lung structure and function. We postulate that their gene expression differences would reflect this. (2) The diversity in the phenotypes of this RI panel could be explained by a few genes whose expression spectrum across this panel could explain the physiological spectrum. In order to test these hypotheses at the gene expression level, we utilized microarray analysis to examine transcriptional differences among lungs of these strains of mice. Transcription profiling of mouse aging lung from two strains C57BL/6J and DBA/2J

Project description:We hypothesize that gene expression in the aging lungs of these two strains of mice are divergent thus contributing to the disparity in the phenotypes.re specifically, (1) Aging DBA/2J mice compared to aging C57BL/6 mice are known to be accelerated in their lung physiology andrphometry; (2) C57BL/6J are known to have longer natural longevity than DBA/2J mice. In order to test these hypotheses at the gene expression level, we utilized microarray analysis to examine transcriptional differences between aging lungs of both strains of mice. Experiment Overall Design: This study utilizes microarray analysis to test these hypotheses. Three sets of lungs were harvested from both strains at each time point (C57BL/6J: 2, 18, AND 26s; DBA/2J: 2 and 18s). RNA was isolated and used for global gene expression profiling (Affymetrixuse 430 2.0 array). Statistically significant gene expression was determined as a minimum 6 counts of 9 pairwise comparisons, minimum 1.5-fold change, and p < 0.05. Further, Absolute | FC - FC SEM | >= 1.5.

Project description:Inhaled welding fumes are deposited on all parts of the respiratory system, with the nasal cavity being the initial point of contact. We investigated the influence of fresh welding fumes on nasal epithelial model, i.e., RPMI 2650 cells, at the transcriptome level. Spark-generated welding fumes at two different concentrations (85 µg/m3 - low; 760 µg/m3 - high) were exposed to RPMI 2650 cell monolayers at air-liquid-interface continuously for 6h, followed by zero hours (or) four hours post-exposure incubation.
