Project description:Accumulating evidence demonstrates that the central nervous system (CNS) is not disconnected from the peripheral immune system; however, precisely how the adaptive immune system surveils the CNS remains a critical question. Recent findings reveal that channels between the dura mater and the skull facilitate the exchange of cerebrospinal fluid and immune cells between the CNS and skull bone marrow (BM) under both homeostatic and disease conditions. Skull BM serves as a source of innate immune cells for the CNS, yet its role in adaptive immune responses remains insufficiently characterized. Here, we identify lymphoid structures within the skull BM, featuring germinal center-like formations and harboring a distinct population of follicular helper-like T cells that promote B cell activation and humoral immunity through CD40L, IL-21, and IFN-γ signaling. Adaptive immune cells within these skull BM lymphoid structures surveil and respond to CNS-derived antigens, which we demonstrate to be critical for optimal antitumor immunity in murine brain cancer models. Our discovery of distinct anatomical sites in the skull BM that enable adaptive immunosurveillance of the CNS highlights promising avenues for immunotherapies targeting neurological diseases, including brain cancers.

The dura mater acts as an autonomous B cell niche supporting immune responses during chronic Trypanosoma brucei infection

Project description:The dura mater hosts a rich population of B cell progenitors, yet its capacity to sustain B cell development during systemic lymphopenia remains unclear. Using Trypanosoma brucei infection, which induces peripheral B cell depletion, we demonstrate that the murine dura mater maintains intact B cell lymphopoiesis independently of bone marrow and spleen. We performed a comparative single cell transcriptomic profiling of the calvaria bone marrow, femur bone marrow, and dura mater from naive and Trypanosoma brucei-infected female mice. The sequencing rendered high quality data from the calvaria bone marrow and the dura mater, but the femur bone marrow failed to generated reliable data and was thus excluded from downstream analyses. Adult skull bone marrow is an expanding and resilient hematopoietic reservoir

Project description:The bone marrow microenvironment is a critical regulator of hematopoietic stem cell self-renewal and fate. While it is appreciated that aging, chronic inflammation and other insults compromise bone marrow function and thereby negatively affect hematopoiesis, it is not known whether different bone compartments exhibit distinct microenvironmental properties and functional resilience. Here, we have employed imaging, pharmacological approaches, and mouse genetics to uncover specialized and highly surprising properties of bone marrow in adult and aging skull. Specifically, we show that the skull bone marrow undergoes lifelong expansion involving vascular growth, which results in an increasing contribution to total hematopoietic output. Furthermore, skull is largely protected against major hallmarks of aging, including upregulation of pro-inflammatory cytokines, adipogenesis and loss of vascular integrity. Striking dynamic and rapid changes to the skull vasculature and bone marrow are induced by physiological alterations, namely pregnancy, but also pathological challenges, such as stroke and experimental chronic myeloid leukemia. These responses are highly distinct from femur, the most extensively studied bone marrow compartment. We propose that skull harbors a protected and dynamically expanding bone marrow microenvironment, which is relevant for experimental studies but, potentially, also clinical treatments in humans.
