Project description:The lung is the major focus of therapeutic approaches for the inherited disorder cystic fibrosis (CF) as without treatment lung disease life-limiting. However, the initiating events that predispose the CF lung to cycles of infection, inflammation and resultant tissue damage are still unclear. Observations of inflammation in the CF lung prior to birth in human and several large animal models suggested an in utero origin for the disease and encouraged a detailed investigation of cell identity prior to birth. Here we used the sheep model of CF (CFTR-/-) and age-matched wild-type sheep to investigate the single cell transcriptomes of proximal and distal lung tissue at 80- and 120 days of gestation and at term (147 days).

Project description:This project deals with the characterisation and establishment of the baseline circulating acellular proteome of sheep serum using generic methods. Serum is a readily available substrate for most experimental and clinical applications as evidenced in recent sheep studies. It involves sample preparation for the optimisation of proteomic profiling of sheep serum. The chapter therefore represents the first step in the development of a proteomics pipeline for the detection protein alterations by first establishing the normal proteomic profile of sheep serum.

Project description:Ovine mastitis is defined as the inflammation of the sheep udder, most commonly caused in response to intramammary infections. Based on the occurrence of clinical signs, mastitis is characterized as either clinical or subclinical (SCM). The impact of ovine SCM on the overall sustainability of dairy sheep farms has been documented underpinning the significance of efficient diagnosis. Although SCM can be detected in cows, the performance and the validity of the methods used do not transfer in dairy sheep. This fact challenges the development of evidence-based ovine udder health management protocols and renders the detection and control of ovine mastitis rather problematic. Currently, cell culture-based models are being successfully used in biomedical studies and have also been effectively used in the case of bovine mastitis. The objective of the present study was to culture ovine primary mammary cells for the development of 2D and 3D cell culture-based models for the study of ovine SCM. Cells were infected by mastitis-inducing pathogens mimicking the pathogenesis of SCM as derived by natural intramammary infections. The secreted proteins were subjected to mass-spectrometry resulting in the identification of 79 distinct proteins. Among those, several had already been identified in healthy or mastitic milk, while others were detected for the first time in the ovine mammary secretome. The development of cell-based models for the early detection and the overall study of SCM has the potential to be applicable and beneficial for the udder health management in dairy sheep.

Project description:Haemonchus contortus is the gastrointestinal nematode (GIN) species that most parasites sheep and goats in tropical and subtropical regions. Due to the high prevalence and pathogenicity by hematophagy, it causes severe anemia, submandibular edema and deaths. Post-genomics tools, such as proteomics, allow the identification of differentially expressed genes and differentially abundant proteins and metabolites between two conditions of a given factor. Thus, the detection of protein profiles that occur in more parasitized animals may be of great interest for the identification of sheep that effectively need anthelmintic treatment. As a result, the parasites are preserved in refugia through Targeted Selective Treatment (TST) strategies for the animals, extension of the period of efficacy of the anthelmintics and, when its use is necessary, parasite control will be more efficient. This project aims to characterize the plasma proteomic profile of sheep breeds susceptible and resistant to GIN infection, aiming at the future development of a diagnostic tool and the understanding of the resistance mechanisms involved in the Santa Inês, White Dorper and Texel sheep breeds. In this way, the development of methodologies, processes and products for the identification of sheep with higher rates of infection by GIN, in addition to the detection of breeds/individuals more resistant to these parasites and the understanding of the resistance mechanisms involved in the different hosts, is presented as a topic of great relevance for sheep farming, which can be extended to goat farming. It also addresses public health issues and the consumer population, which is increasingly demanding in terms of quality, certification and safety.

Project description:We carried out a cross species cattle-sheep array comparative genome hybridization (aCGH) experiment in order to identify copy number variations (CNVs) in the sheep genome analysing animals of Italian dairy breeds (Sarda, Bagnolese, Laticauda, Massese and Valle del Belice) using a tiling oligonucleotide array with ~385,000 probes designed on the bovine genome. We identified 135 CNV regions (CNVRs) covering about 10.5 Mb of the virtual sheep genome referred to the bovine genome (0.398%) with a mean and median equal to 77.6 kb and 55.9 kb, respectively. A comparative analysis between the identified sheep CNVRs and those reported in the cattle and goat genomes indicated that overlaps between sheep and goat and sheep and cattle CNVRs are highly significant (P<0.0001) suggesting that several chromosome regions might contain recurrent interspecies CNVRs. Many sheep CNVs affect genes with important biological functions. Further studies are needed to evaluate the functional relevance of these CNVs.

Project description:Estrogen is an important hormone that affects muscle development in female animals. Previous studies have shown that estrogen can protect muscle cells from apoptosis by inhibiting the MAPK signaling pathway. However, the molecular mechanisms by which estrogen-induced MAPK signaling regulates myoblast growth and development remain unclear. In this study, RNA-seq was performed on ovariectomized small-tailed Han (OR-STH) sheep and sham surgery small-tailed Han (STH) sheep to analyze the effects of estrogen on muscle growth and development in female animals. There were 8,721 differentially expressed circRNAs (DECs), 143 differentially expressed miRNAs (DEMs) and 2,238 differentially expressed mRNAs (DEGs) in the longissimus dorsi between the OR-STH and STH groups. Bioinformatics analysis revealed that the differentially expressed gene MAPK15 was significantly enriched in the MAPK signaling pathway, which is important for muscle development. Therefore, we constructed the ceRNA network circFAM171A1/oar-miR-485-5p/MAPK15 and explored its effect on muscle growth and development. The results of the molecular mechanism experiments indicated that circFAM171A1 can sponge oar-miR-485-5p to regulate MAPK15. The addition of the exogenous hormone estradiol (E2) to sheep myoblasts could induce circFAM171A1, regulate the expression of oar-miR-485-5p and MAPK15, and promote the proliferation of sheep myoblasts. The results showed that MAPK15 and circFAM171A1 significantly promoted the proliferation of myoblasts and inhibited the apoptosis of myoblasts in sheep, whereas oar-miR-485-5p inhibited the expression of MAPK15 and circFAM171A1, inhibited myoblast proliferation and promoted apoptosis. Furthermore, circFAM171A1 attenuated the inhibitory effect of oar-miR-485-5p on myoblasts. In summary, estrogen induced the expression of circFAM171A1 in sheep myoblasts, and circFAM171A1 can act as a sponge for oar-miR-485-5p to promote the expression of the target gene MAPK15 and ultimately regulate the proliferation of sheep myoblasts. This study provides new insights into the molecular mechanism of estrogen regulation of muscle growth and development in female animals.
