Human Peripheral Blood mononuclear cells from postmenopausal women: grapefruit juice vs. flavanone-free placebo

ABSTRACT: Transcriptional profiling of Peripheral Blood mononuclear cells (PBMCs) in apparently healthy postmenopausal women consuming flavanones in grapefruit juice (340 ml/day providing around 210 mg of naringenin glycosides) or flavanone-free placebo drink for 6 months in a cross-over design with a 2-month washout period between consumption of two studied drinks. Goal was to determine the effects of chronic consumption of flavanones in garpefruit juice on global gene expression profiles in humans

Organic vegetable juice supplement modulates microbial community in continuous colon simulation system

Project description:In this study, we investigated the effects of organic vegetable juice supplementation on modulating the microbial community, and how its consumption ameliorates blood lipid profiles in diet-induced obese mice. Here, we analyzed the effect of organic vegetable juice on the microbial community and fatty acid synthesis via animal experiments using diet-induced obese mice and continuous colon simulation system. Organic vegetable juice supplement influenced intestinal bacterial composition from phylum to genus level, including decreased Proteobacteria in the ascending colon in the phylum. At the family level, Akkermansia which are associated with obesity, were significantly augmented in the transverse colon and descending colon compared to the control juice group. In addition, treatment with organic vegetable juice affected predicted lipid metabolism function genes related to lipid synthesis. Organic vegetable juice consumption did not have a significant effect on weight loss but helped reduce epididymis fat tissue and adipocytes. Additionally, blood lipid profiles, such as triglyceride, high-density lipoprotein, and glucose, were improved in the organic vegetable juice-fed group. Expression levels of genes related to lipid synthesis, including SREBP-1, PPARγ, C/EBPα, and Fas, were significantly decreased. Analysis of antioxidant markers, including 8-OHdG and MDA, in the vegetable juice group, indicated that blood lipid profiles were improved by the antioxidant effect. These results suggest that organic vegetable juice supplementation may modulate gut microbial community and reduce the potential role of hyperlipidemia in diet-obese mice.

Salmonella Typhimurium LT2: Control versus naringenin

Project description:Salmonellosis is one of the leading health problems worldwide. With the rise of drug resistance strains it has become imperative to identify alternative strategies. Naringenin, a flavonone, is present predominantly in grapefruit. Previously we have demonstrated that naringenin is potent inhibitor of cell-cell signaling. In the present study, we provide evidence that naringenin specifically represses 24 genes in the Salmonella pathogenicity island 1, and down-regulates 17 genes involved in flagellar and motility; thereby, attenuating virulence and cell motility, respectively. This is the first molecular evidence to demonstrate effect of naringenin on bacterial virulence and cell motility.

Project description:Alterations in stress-related gene-expression may play a role in stress-related drinking and the risk for alcohol dependence. Microarrays were used to measure changes in gene-expression in peripheral blood in non-smoking, social drinking subjects exposed to three types of personalized imagery: neutral, stressful (but not alcohol- related), and alcohol-related cues. Gene-expression was measured at baseline, immediately after, and 1 hour after stimulus presentation. Subjects were allowed to drink up to 750cc of beer in a “taste-test” following stimulus presentation in each imagery condition, and the amount of beer consumed was recorded. Gene-expression levels were compared in 2 groups of non-smoking subjects (n=11/group): heavy drinkers (HD, defined as regular alcohol use over the past year of at least 8 standard drinks/week for women and at least 15 standard drinks/week for men), and moderate drinkers (MD, defined as up to 7 standard drinks/week for women and 14 standard drinks/week for men).

Comparison of transcriptomes in five varieties of citrus fruit

Project description:To identify genes associated with citrus peel development and manifestation of peel disorders, we analyzed flavedo, albedo and juice sac tissues from five types of citrus fruit including, mandarin orange, navel orange, valencia orange, grapefruit and lemon. Fruits of five different citrus cultivars. Mature, healthy fruits of five different citrus cultivars (M-bM-^@M-^\ValenciaM-bM-^@M-^] and M-bM-^@M-^\NavelM-bM-^@M-^] orange [Citrus sinensis], mandarin [Citrus reticulata], lemon [Citrus M-CM-^W limon], grapefruit [Citrus M-CM-^W paradisi]) were purchased from a food market located in Davis, CA, USA. For all five types of fruit, three tissues (flavedo, albedo, and juice sacs) were compared separately. Each of the three tissues from each of the five types of fruit were sampled in three biological replicates, for a total of 45 samples. Samples were prepared from a 1 cm-thick equatorial disc and four sections (N, S, E, and W) were cut. Project description:Analysis of the effect of using a fruit and vegetable juice concentrate to reduce systemic inflammation in obesity. The hypothesis tested whether the presence of polyphenols in the fruit and vegetable juice concentrate could reduce the expression of systemic inflammatory genes in the blood of Obese patients with high levels of plasma CRP (≥3.0). Results provide evidence that systemic inflammatory genes/ and or pathways may be modulated by the fruit and vegetable juice concentrate.
