Project description:We compared the transcriptome of homozygous mutants for AtCesA4 and AtCesA6 (cellulose synthase genes) to their heterozygous counterparts that have a wild type phenotype. All plants were 4-weeks-old and grown under short day conditions. 10 samples were used in this experiment: 3 each homozygous and heterozygous AtCesA4 plants, and 2 each homozygous and heterozygous AtCesA6 plants.

Project description:R. cellulolyticum strains (the parent strain, ΔccpA, ΔccpB and Δcph) were cultivated in defined VM medium with cellulose (10 g/L).Since ΔccpA and Δcph could not grow on cellulose, we performed an incubation experiment to understand how ΔccpA and Δcph respond to cellulose. Specifically, the parent strain, ΔccpA and Δcph were grown to an OD600 of 0.5-0.6 in 50 ml defined VM media with cellobiose as the carbon source. Each strain had three biological replicates. Bacterial cells from each biological replicate were then collected by centrifugation at 4000 g and washed twice with the defined VM medium (no carbon added). Finally, washed cells from each biological replicate were inoculated into the defined VM medium with 10 g/L cellulose. During shaking incubation at 34°C, samples were collected at five time points (0,1, 3, 6, 12 hours). After centrifugation at 4°C, 5000×g for 10 min, cell pellets were immediately flash frozen with liquid nitrogen and then stored at -80°C for further use. For ΔccpB, ΔccpB and the parent strain were cultivated with six biological replicates and collected at mid-exponential growth phase.

Project description:The interplay between plant chemistry and architecture was investigated using a pharmacological approach combined with mutant analysis. Due to the high abundance and essential role of cellulose in plant development and function, it was hypothesized that perturbation of cellulose biosynthesis would have far-reaching effects on plant chemistry and resource allocation. The impact of cellulose disruption was studied through comprehensive multiphase-NMR (CMP-NMR) using the Arabidopsis thaliana cellulose synthase mutant ectopic lignification1 (eli1). CMP-NMR confirmed several known metabolic impacts of cellulose disruption including increased lignification, increased starch production, and a shift from crystalline to amorphous cellulose. It also revealed unexpected metabolic impacts such as increased methanol production, increased seed-derived lipid content, and the presence of the peptide pentaglycine, which had not been previously observed in plants. It was further hypothesized that the metabolic impacts of cellulose disruption are mediated by one or more signal molecules that would be elevated in cellulose synthase mutants such as eli1. The presence of one or more signal molecules in eli1 that induce a lignification response was confirmed by exposing wild-type seedlings to ground tissue from eli1. Treatment of wild-type seedlings with pentaglycine suggested that the peptide may be at least partially responsible for mediating this response. A case is made for glycine rich proteins (GRPs) as the source of pentaglycine in vivo, and for wall associated kinases (WAKs) as receptors that trigger either developmental or defence responses through differential binding of intact GRPs and oligoglycine peptides.

Project description:Transcriptional profiling after inhibition of cellulose synthesis by thaxtomin A and isoxaben in Arabidopsis thaliana suspension cells Perturbations in the cellulose content of the plant cell wall lead to global modifications in cellular homeostasis, as seen in cellulose synthase mutants or after inhibiting cellulose synthesis. In particular, application of inhibitors of cellulose synthesis such as thaxtomin A (TA) and isoxaben (IXB) initiates a programmed cell death (PCD) in Arabidopsis thaliana suspension cells that is dependent on de novo gene transcription. To further understand how TA and IXB activate PCD, a whole genome microarray analysis was performed on mRNA isolated from Arabidopsis suspension cells exposed to TA and IXB. More than 75% of the genes upregulated by TA were also upregulated by IXB, including genes encoding cell wall-related and calcium-binding proteins, defence/stress-related transcription factors, signalling components and cell death-related proteins. Comparisons with published transcriptional analyses revealed an important subset of genes generally induced in response to various biotic and abiotic stress.

Project description:Clostridium cellulovorans is among the most promising candidates for consolidated bioprocessing (CBP) of cellulosic biomass to liquid biofuels (e.g. ethanol, butanol). C. cellulovoranscan metabolize all the main plant polysaccharides (i.e. cellulose, hemicellusoses and pectins). Unlike other well established cellulolytic microorganisms, C. cellulovorans most abundant catabolite is butyrate. This attracted attention on this strain as potential butanol producer since most reactions involved in butyrate and butanol synthetic pathway from acetyl-coA are common. Recent studies demonstrated that the introduction of a single heterologous alcohol/aldehyde dehydrogenase can significantly divert the branching-point intermediate, i.e. butyryl-CoA, towards butanol production in this strain. In spite of the potential of C. cellulovorans for application in CBP of plant biomass, engineering its metabolic pathways towards industrial utilization still requires enhanced understanding of its metabolism. Few recent studies aimed at understanding the regulation of C. cellulovorans central carbon metabolism in response to different substrate availability, which seem insufficient for the aforementioned purposes. The present study aimed at improving comprehension of cellulose metabolism in C. cellulovorans by comparing growth kinetics, substrate consumption/product accumulation and whole-cell soluble proteome with those of C. cellulovorans grown on a soluble carbohydrate, i.e. glucose, as the main carbon source. Modulations of the central carbon metabolism in response to changes in the growth substrate were detected, including the regulation of glycolytic enzymes, fermentation pathways and nitrogen assimilation. Our data suggest that a higher energy expenditure occurs in cellulose-grown C. cellulovorans which induces up-regulation of ATP synthetic pathways, e.g. acetate production and ATP synthase.

Project description:The barley brittle stem mutants, fs2, designated X054 and M245, have reduced levels of cellulose compared with their isogenic parents Ohichi and Shiroseto. A custom-designed microarray, based on Agilent technology and including genes involved in cell wall metabolism, was used to compare transcript levels in the mutant and parental lines. For both mutants, the microarray revealed a marked decrease in mRNA for the HvCesA4 cellulose synthase gene in specific zones of stem internodes, and this was confirmed by quantitative PCR.

Project description:Interventions: Administraion of CPT-11 is twice for 4 weeks on days 1 and 15. CPT-11 is reconstituted in >=250 mL of normal saline or 5% dextrose in water and infuse 90min on day 1 for pharmacokinetics, and 90min over on day15. CPT-11 adjusted dosage is determined from 50,75,100,125 or 150mg/sqm in the heterozygous group and the homozygous group by continual reassessment method. CPT-11 dosage is fixed at 150mg/sqm in the wild group.Definision of UGT1A1 polymorphisms groups: The homozygous group is patient with homozygous genotype of UGT1A1*28/*28 or UGT1A1*6/*6, with combined heterozygous genotypes of UGT1A1*28 and UGT1A1*6. The heterozygous group is patient with heterozygous genotype of either UGT1A1*28 or UGT1A1*6. The wild group is patients with no UGT1A1*28 and UGT1A1*6 mutation.
Primary outcome(s): Maximum tolerated dose for the heterozygous group and the homozygous group, respectively. Incidence rate of dose limiting toxicities for the wild group.

Project description:In this study, we use Cross-linking Mass Spectrometry to identify the interaction sites of the microtubule binding N-terminal domain of the companion of cellulose synthase 1 protein (CC1∆C223) from Arabidopsis thaliana with tubulin. We consistently detected four cross-linked peptides of CC1∆C223 to β-tubulin (K40 to E111, K94 to E111, K96 to E111 and K96 to E158; letters and numbers indicate specific amino acids in the protein sequence of CC1∆C223 and β-tubulin, respectively) and one cross-linked peptide of CC1∆C223 to α-tubulin (K40 to D327).
