Transcriptomic analyses identifies novel regulators of placental development

ABSTRACT: The placenta is a transient organ that is repsonsible for multiple processes during pregnancy. Here we aim to study the development of the placenta using mouse models at embryonic day (e) e7.5, e8.5 and e9.5 by integrating RNA-seq data at the three timepoints to identify important genes, inferring their interaction networks, and predict novel regulators of placental development.

Placental and decidual timecourse samples normalized and modeled with an undissected e17 sample

Project description:We used full genome microarrays to profile the full lifetime of the mouse placenta from embryonic day 8.5 (e8.5), at the time of chorioallantoic fusion, until postnatal day 0 (P0). For these samples, at each stage the fetal placenta and maternal decidual tissues were dissected and profiled separately (See series 1). Maternal Influenza A Virus Infection Restricts Fetal and Placental Growth and Adversely Affects the Fetal Thymic Transcriptome

Project description:Maternal influenza A viral infections in humans are associated with low birth weight, increased risk of pre-term birth, stillbirth and congenital defects. To examine the effect of maternal influenza virus infection on placental and fetal growth, pregnant C57BL/6 mice were inoculated intranasally with influenza A virus A/CA/07/2009 pandemic H1N1 or PBS at E3.5, E7.5 or E12.5, and the placentae and fetuses collecgted and weighed at E18.5. Fetal thymuses were pooled from each litter. Placentae were examined histologically, stained by IHC for CD34 and vascular channels quantified. RNA from E7.5 and E12.5 placentae and E7.5 fetal thymuses was subjected to RNA sequencing and pathway analysis. Placental weights were decreased in litters inoculated with influenza at E3.5 and E7.5. Placentae from E7.5 and E12.5 inoculated litters exhibited decreased labyrinth development and transmembrane protein 150A gene was upregulated in E7.5 placentae. Fetal weights were decreased in litters inoculated at E7.5 and E12.5 compared to controls. RNA sequencing of E7.5 thymuses indicated that 957 genes were downregulated 2-fold including Mal, which is associated with Toll-like receptor signaling and T cell differentiation. There were 28 upregulated genes. It is concluded that maternal influenza A virus infection impairs fetal thymic gene expression as well as restricting placental and fetal growth.

Blimp1/Prdm1 governs terminal differentiation of endovascular trophoblast giant cells and defines multipotent progenitors in the developing placenta

Project description:Expression profiling of Prdm1 mutant E9.5 placenta was performed using Illumina whole genome V2 arrays. The hypothesis tested in the present study was that Blimp1 regulates the transcription of key genes involved in trophoblast differentiation. We demonstrate that the invading SpA-TGCs display robust Blimp1 expression and Blimp1 functional loss selectively disrupts specification of this discrete TGC sub-type. Transcriptional profiling experiments identified additional SpA-TGC lineage restricted marker genes that potentially regulate placental morphogenesis. Prdm1BEH/+ (Vincent et al., 2005) animals were intercrossed to generate null placental tissue. Total RNA obtained from 10 Prdm1+/+ and 11 Prdm1-/- E9.5 placenta samples was hybridized to Illumina WG6_V2 beadchips.

The development of mouse trophoblast cells at the single cell level

Project description:The placenta is a poorly understood yet vital support organ. Transcriptome analysis at single cell resolution can help us understand cell compositions, developmental processes, and physiological functions. Both mice and humans have the hemochorial placenta. Profiling the single cell transcriptome of the mouse placenta is necessary for deepening our knowledge of mammalian placentation.We systematically profiled single cell transcriptomes of mouse placentae every day from E7.5 to E14.5. After stringent filtering, 15682 mouse trophoblast cells were subjected to following transcriptome analysis. We noted that trophoblast cells underwent 3 main differentiation stages: E7.5-E8.5, E9.5-E10.5, and E11.5-E14.5. With the help of advanced computational technologies from Velocyto, PAGA, monocle, and SCENIC, we have updated the recognitions of several developmental events. Firstly, P-TGCs are suggested to only derive from EPC like cells. Secondary, sinusoid trophoblast and spongiotrophoblast all derived from EPC cells, and their cell fates have been determined before the chorioallantoic fusion. Sinusoid trophoblast cells were not derived from chorion. Thirdly, SpA-TGCs were suggested to be differentiated from spongiotrophoblast cells via Gly-T cells. Furthermore, new transcription factors have been found to play roles during the differentiation of trophoblast cells. Lastly, the expression of Ifnlr1 in chorion branch cells gradually increased during placentation, which reconfirmed that mature placenta can defend the Zika virus (ZIKV) through IFN signaling. We clarified the developmental histories of mouse trophoblast cells at the branch level, especially the differentiation of sinusoid branch trophoblast cells. Meanwhile, we offered a convincing data resource for the study of mammalian placentation and etiology analysis of pregnancy-associated diseases.

Timed-pregnant CD-1 mice were obtained from Charles Rivers Labs and dissected at e7.5 to extract the ectoplacental cones (EPCs) and at e9.5 to obtain the placenta as described by Martin and Cockroft (Martin, P.; Cockroft, D. L. Culture of Postimplantation Mouse Embryos). Staging of the mouse embryos was done according to Theiler criteria (Theiler, K. Placental effects on maternal brain revealed by disrupted placental gene expression in mouse hybrids

Project description:The mammalian placenta is both the physical interface between mother and fetus, and the source of endocrine signals that target the maternal hypothalamus, priming females for parturition, lactation and motherhood. Despite the importance of this connection, the effects of altered placental signaling on the maternal brain are understudied. Here, we show that placental dysfunction alters gene expression in the maternal brain, with the potential to affect maternal behavior. Using a cross between the house mouse and the Algerian mouse in which hybrid placental development is abnormal, we sequenced late gestation placental and maternal medial preoptic area transcriptomes and quantified differential expression and placenta-maternal brain co-expression between normal and hybrid pregnancies. The expression of Fmn1, Drd3, Caln1 and Ctsr was significantly altered in the brains of females exposed to hybrid placentas. Most strikingly, expression patterns of placenta-specific gene families and Drd3 in the brains of house mouse females carrying hybrid litters matched those of female Algerian mice, the paternal species in the cross. Our results indicate that the paternally-derived placental genome can influence the expression of maternal-fetal communication genes, including placental hormones, suggesting an effect of the offspring's father on the mother’s brain.
