A Novel Therapeutic Agent: Harnessing Macrophage Polarization to Combat Sepsis and Alleviate Lung Injury

Project description:Sepsis is a complex syndrome characterized by dysregulated immune responses, with macrophage polarization being pivotal in its immunomodulation. Lung injury is a prevalent and severe consequence in sepsis. This research delved into the therapeutic potential of JSNPs. Initially, investigations involving bioinformatics analysis and JHU083 treatment revealed that while JHU083 extended septic mice's survival, it had limited impact on lung injury.Subsequently, studies on JSNPs showed it could improve sepsis and lung injury in CLP and LPS models. Mechanistically, JSNPs promoted macrophage polarization towards the anti - inflammatory M2 phenotype, safeguarding neurons from sepsis - induced lung injury. Further experiments demonstrated that JSNPs's protection of lung neurons was M2 macrophage dependent, and macrophages released neurotrophic factors under JSNPs's influence to shield neurons. Overall, JSNPs emerges as a promising candidate for treating sepsis and its associated lung injury.

Low-level tragus stimulation alleviates sepsis-associated acute kidney injury by promoting renal M2 macrophage polarization.

Project description:Background: Low-level tragus stimulation (LL-TS) can reduce acute renal injury in sepsis and improve survival rates. However, the underlying molecular mechanisms remains unknown. Renal macrophages that contributed to AKI in sepsis under low-level tragus stimulation were characterized. Methods: Mice were randomly assigned to five groups following diverse treatments: Control, Sham, Sepsis, LL-TS + sepsis, and LL-TS + sepsis + Methyllycaconitine citrate (MLA). Blood creatinine testing and hematoxylin and eosin (HE) staining were used to assess kidney injury. Single-cell RNA sequencing were used to profile renal macrophages from CD45+ cells in mouse kidney. Western blot, immunofluorescence and HE staining were performed to verify the level of the alpha7 nicotinic acetylcholine receptor (α7nAChR) or interleukin-1β expression in kidney. Results: HE staining and blood creatinine testing of renal tissue indicated that LL-TS alleviated renal injury in mice with sepsis-associated acute kidney injury (AKI). A map of the renal cellular landscape was generated to identify macrophages that contributed to AKI in sepsis under LL-TS. Pseudo-timing analysis showed that LL-TS played an anti-inflammatory role by promoting renal monocyte conversion into M2 macrophages. Moreover, LL-TS significantly increased α7nAChR expression and decreased interleukin-1β expression in renal tissues. Conclusions: LL-TS alleviated sepsis-associated AKI by increasing α7nAChR expression and M2 macrophage polarization which promotes the anti-inflammatory process in sepsis. The results suggests that LL-TS may serve as a potential therapeutic strategy for sepsis-induced kidney dysfunction, ultimately improving patient outcomes.

Sepsis-induced acute lung injury is exacerbated by pulmonary Wnt signaling-dependent migration of small intestinal memory γδT17 cells to the lung

Project description:Sepsis, a worldwide health crisis, is notorious for its high mortality rate, often attributed to the development of acute lung injury. In this context, the lung, being particularly vulnerable to septic damage, emerge as a primary battleground. The possible roles of the gut-lung axis mediated by the gut microbiota and its metabolic products in the context of acute lung injury have been documented. However, the direct migration of intestinal immune cells to the lung as a mediator of acute lung injury remains unclear. Our study unveiled a process where cecal ligation and puncture-induced sepsis prompted a migration of γδ T cells from the small intestine to the lung, subsequently triggering an overwhelming inflammatory response dominated by IL-17A. Small intestinal memory γδT17 cells (CD44+ IL-7Rhigh CD8low Ly6C–) were the major subtype of γδ T cells that migrated from the small intestine to the lung and aggravated acute lung injury. Moreover, the activation of the Wnt signaling in the alveolar macrophages during sepsis was identified as a driving factor for the subsequent CCL1 upregulation, which prompted the migration of small intestinal memory γδT17 cells to the lung. S-Ketamine (S-KT) treatment demonstrated a notable capacity to alleviate acute lung injury through dampening pulmonary Wnt/β-catenin signaling-mediated migration of small intestinal γδT17 cells to the lung. Our findings highlight the significant contribution of the direct migration of immune cells from the small intestine to the lung in sepsis-induced acute lung injury, and clrify the pathological significance of the localized elevation of IL-17A in the lung.
