The use of tissue microRNAs to distinguish indolent and aggressive forms of prostate cancer

ABSTRACT: The low sensitivity or specificity of current parameters for prostate cancer detection leads to overdiagnosis and overtreatment of the disease. MicroRNAs are small non-coding RNAs whose level can reflect clinical and biological characteristics of the tumor. In the study, our objective was to identify miRNAs associated with disease aggressiveness, which can supplement risk stratification systems.

Description: Chronic Myeloid Leukaemia presents in chronic phase (CP) and terminates in 'blast crisis'. Despite a common abnormality, the duration of CP is variable. The aim is to compare the gene expression profiles of the indolent and aggressive forms of CML. All samples were taken within 3 months of first diagnosis. Indolent patients were defined by chronic phase CML, duration minimum 7 years. Aggressive patients were defined by chronic phase, duration maximum 3 years.

Differentially Expressed Glycoproteins in Pre- and Post-Digital Rectal Examination Urine Samples for Detecting Aggressive Prostate Cancer

Project description:Nonaggressive prostate cancer (NAG-PCa) with Gleason score of 6 is considered as a low-risk disease and does not require clinical interventions. However, current assessment of aggressiveness of PCa is invasive using needle biopsies. Urine is an appealing biospecimen for noninvasive detection of aggressive PCa. Using urine, particularly from easily obtainable from pre-DRE urine specimens, to identify AG-PCa-associated molecular markers is critical for clinical risk stratification. Herein, we acquired quantitative mass spectrometry data for glycoproteins from pre-DRE and post-DRE urine specimens from patients underwent PCa diagnosis with biopsies. Compared with urinary glycoproteins identified from post-DRE urine samples, we confirmed that three previously reported AG-PCa-associated glycoproteins identified in post-DRE urine specimens were also found to be significantly associated with AG disease in pre-DRE urine samples. In addition, new AG-PCa-associated glycoproteins were identified in pre-DRE urine specimens. Our study provides a foundation for further studies of AG-PCa biomarkers using pre-DRE urine specimens.

Transcriptional profiling of indolent and aggressive mammary lesions

Project description:We performed transcriptional profiling of tumor cells from indolent and aggressive lesions isolated from a mouse model of breast cancer. To initiate formation of mammary lesions, we delivered lentiviral particles carrying constitutively-activated Erbb2 tagged with HA and GFP (caErbB2) through the lactiferous glands of adult virgin female. Mammary glands bearing lesions were visualized under a fluorescence stereoscope, and regions harboring aggressive (>2mm, comprised of invasive lesions) and indolent (<2mm, enriched for in situ lesions) lesions were dissected away from each other. Lesions from multiple animals were pooled to obtain sufficient material for downstream analysis. Following tissue digestion, GFP-expressing epithelial cells were isolated by fluorescence-activated cell sorting (FACS), and then subjected to bulk RNA-seq.

The combined detection of Amphiregulin, Cyclin A1 and DDX20/Gemin3 expression predicts aggressive forms of Oral Squamous Cell Carcinoma

Project description:Large-scale genetic and epigenetic deregulations enable cancer cells to ectopically activate tissue-specific expression programs. A specifically designed strategy was applied to oral squamous cell carcinomas (OSCC) in order to detect ectopic gene activations and develop a prognostic stratification test. A dedicated original prognosis biomarker discovery approach was implemented using genome-wide transcriptomic data of OSCC, including training and validation cohorts. Abnormal expressions of silent genes were systematically detected, correlated with survival probabilities and evaluated as predictive biomarkers. The resulting stratification test was confirmed in a third cohort using immunohistochemistry. In order to address the potential interest of the ectopic expressions as therapeutic targets, the transcriptome of OSCC cells in which one of the predictive marker genes, CCNA1, was inactivated was analysed by RNAseq. The genome-wide expression profile of an OSCC cell line, where CCNA1 had been inactivated was found similar to that of aggressive tumours positive for the three genes. The exploration of the whole gene expression profile characterizing aggressive OSCC tumours highlights their enhanced proliferative and poorly differentiated intrinsic nature. Our experimental targeting of CCNA1 in OSCC cells is associated with a shift of transcriptomic signature towards the less aggressive form of OSCC, suggesting it could be a good target for therapeutic approaches.
