Iron treatment induces resistance against the rice blast fungus Magnaporthe oryzae through potentiation of immune responses

ABSTRACT: To investigate the role of iron excess in rice immune responses to Magnaporthe oryzae infection. Gene expression profiling analysis were performed using data obtained from RNA-seq of rice plants grown in differential iron supply and challenged with Magnaporthe oryzae spores.

Molecular basis of drought-induced susceptibility to the rice blast fungus Magnaporthe oryzae

Project description:In order to understand the mechanisms of Drought induced susceptibility (DIS) we’ve conducted a dual RNAseq experiment on rice infected tissues by Magnaporthe oryzae. At 4 days post inoculation tissues have been collected on mock inoculated and M. oryzae inoculated plants. Rice were conducted under two type of water regime: DIS Drought during three days before inoculation, NoDIS no drought before inoculation. RNAseq was conducted both on rice and fungal RNA.

Analysis of transcript abundance during appressorium formation in the rice blast fungus Magnaporthe oryzae using HT-SuperSAGE.

Project description:The rice-blast fungus Magnaporthe oryzae is disseminated using a three-celled spore or conidium. Upon landing on the surface of a rice leaf, the conidium germinates and forms a specialised structure for entry into the host plant - the appressorium. In this study we have followed gene expression thoughout germination of the conidium and formation of the appressorium to identify genes that may be important for appressorium function. We have also compared gene expression in the wild-type germinating conidium at four hours to a mutant strain deleted for the MAP kinase pmk1. The pmk1 mutant is unable to form appressoria.

Positive roles for Osa-miR7695 in regulating defense responses against the rice blast fungus

Project description:MicroRNAs (miRNAs) are small non-coding RNAs that function as post-transcriptional regulators of gene expression in eukaryotes. In rice, miR7695 targets an alternatively spliced transcript of natural resistance-associated macrophage protein 6 (OsNramp6) encoding an iron transporter whose expression is regulated by infection with the rice blast fungus Magnaporthe oryzae. Rice plants grown under high iron supply showed blast resistance, which supports that iron is a factor in controlling blast resistance by still unknown mechanisms. Here, iron accumulated near M. oryzae appressoria, the sites of pathogen entry, and in cells surrounding infected regions of the rice leaf. Activation-tagged MIR7695 rice plants (MIR7695-Ac) exhibited enhanced resistance to M. oryzae infection. RNA-seq analysis revealed that blast resistance in MIR7695-Ac plants was associated with strong induction of defense-related genes, including pathogenesis-related and diterpenoid biosynthetic genes. Levels of phytoalexins during pathogen infection were higher in MIR7695-Ac than wild-type plants. Early phytoalexin biosynthetic genes, OsCPS2 and OsCSP4, were highly upregulated in rice plants grown under high iron supply. Our data indicate that miR7695 positively regulates rice immunity while establishing links between defense and iron signaling in rice. MiR7695-mediated regulation of OsNramp6 has great potential for the development of strategies to control rice blast disease.

Physiological stressors and invasive plant infections alter the small RNA transcriptome of the rice blast fungus, Magnaporthe oryzae

Project description:The rice blast fungus, Magnaporthe oryzae is a destructive pathogen of rice and other related crops, causing significant yield losses worldwide. Endogenous small RNAs (sRNAs), including small interfering RNAs (siRNAs) and microRNAs (miRNAs) are critical components of gene regulation in many eukaryotic organisms. Recently several new species of sRNAs have been identified in fungi. This fact along with the availability of genome sequence makes M. oryzae a compelling target for sRNA profiling. We have examined sRNA species and their biosynthetic genes in M. oryzae, and the degree to which these elements regulate fungal stress responses. To this end, we have characterized sRNAs under different physiological stress conditions, which had not yet been examined in this fungus. The resulting libraries are composed of more than 37 million total genome matched reads mapping to intergenic regions, coding sequences, retrotransposons, inverted, tandem, and other repeated regions of the genome with more than half of the small RNAs arising from intergenic regions. The 24 nucleotide (nt) size class of sRNAs was predominant. A comparison to transcriptional data of M. oryzae undergoing the same physiological stresses indicates that sRNAs play a role in transcriptional regulation for a small subset of genes. Support for this idea comes from generation and characterization of mutants putatively involved in sRNAs biogenesis; our results indicate that the deletion of Dicer-like genes and an RNA-Dependent RNA Polymerase gene increases the transcriptional regulation of this subset of genes, including one involved in virulence. Various physiological stressors and in planta conditions alter the small RNA profile of the rice blast fungus. Characterization of sRNA biosynthetic mutants helps to clarify the role of sRNAs in transcriptional control.
