Pain may be related to pathogen phenotype: Transcriptome analysis of Staphylococcus aureus isolated from PJI with different VAS scores

ABSTRACT: To explore whether S. aureus contributes to the mechanism of pain, we assayed S. aureus isolates from 3 severe pain PJI (sp-PJI) and 3 mild pain PJI (mp-PJI) by RNA-sequencing.

Mass Spectrometry-Based Proteomic Profiling of Sonicate Fluid Differentiates Staphylococcus aureus Periprosthetic Joint Infection from Non-Infectious Failure: A pilot study

Project description:This study aims to use proteomic profiling of sonicate fluid samples to compare host response during Staphylococcus aureus-associated periprosthetic joint infection (PJI) and non-infected arthroplasty failure (NIAF) and investigate novel biomarkers to increase diagnostic accuracy. In this pilot study, eight sonicate fluid samples (four from NIAF and four from caused by Staphylococcus aureus PJI) were studied. Samples were reduced, alkylated and trypsinized overnight, followed by analysis using liquid chromatography-tandem mass spectrometry (LC-MS/MS) on a high-resolution Orbitrap Eclipse mass spectrometer. MaxQuant software suite was used for protein identification, filtering, and label free quantitation. Principal component analysis of the identified proteins clearly separated S. aureus PJI and NIAF samples. Overall, 810 proteins were quantified in any three samples from each group and 35 statistically significant differentially abundant proteins (DAPs) were found (2-sample t-test p-values ≤0.05 and log2fold-change values ≥2 or ≤-2). Gene ontology pathway analysis found that microbial defense responses, specifically those related to neutrophil activation, were increased in S. aureus PJI compared to NIAF samples. Proteomic profiling of sonicate fluid using LC-MS/MS, alone or in combination with complementary protein analyses, differentiated S. aureus PJI and NIAF in this pilot study.

Staphylococcus aureus gene expression during infection and antibiotic treatment with moxifloxacin in a guinea pig biofilm infection model

Project description:Staphylococcus aureus is a major agent in device related infections. Upon infection, it upregulates virulence factors and adapts to host environment. We investigated the gene expression profile of S. aureus in a guinea pig infection model. Four Teflon® cages were placed subcutaneously in each of 12 animals prior to inoculation of a S. aureus strain from a patient with prosthetic joint infection (PJI). The animals were divided into two groups of six, one treated with moxifloxacin after three days of infection and for four days, and the other untreated. Cage fluid and cages were collected for RNA sequencing analysis and metabolomics. The gene expression profiles in untreated controls on day 7 and 9 were not differentiable and resemble the profile reported for the PJI case from where the strain originated. Compared to the profiles from day 3, we found upregulation of ammonia production through urea degradation, arginine deiminase and acetoin biosynthesis pathways on day 7 and 9, while several virulence genes were downregulated. Interestingly, the transcriptome seven day after termination of moxifloxacin treatment resembled infection day 3. Overall, the gene expression profiles showed adaptation to hypoxic and acidic environment during infection development. We deem the model to be suitable for pathogenesis studies.

The Staphylococcus aureus CamS lipoprotein is a repressor of toxin production that shapes host-pathogen interaction

Project description:Lipoproteins of the opportunistic pathogen Staphylococcus aureus play a crucial role in various cellular processes and host interactions. Consisting of a protein and a lipid moiety, they support nutrient acquisition and anchor the protein to the bacterial membrane. Recently, we identified several processed and secreted small linear peptides that derive from the secretion signal sequence of S. aureus lipoproteins. Here, we show for the first time, that the protein moiety of the S. aureus lipoprotein CamS has a biological role that is distinct from its associated linear peptide staph-cAM373. The small peptide was shown to be involved in interspecies horizontal gene transfer, the primary mechanism for the dissemination of antibiotic resistance among bacteria. We provide evidence that the CamS protein moiety is a potent repressor of cytotoxins, such as α-toxin and leukocidins. The CamS-mediated suppression of toxin transcription was reflected by altered disease severity in in vivo infection models involving skin and soft tissue, as well as bloodstream infections. Collectively, we have uncovered the role of the protein moiety of the staphylococcal lipoprotein CamS as a previously uncharacterized repressor of S. aureus toxin production, which consequently regulates virulence and disease outcomes. Notably, the camS gene is conserved in S. aureus, and we also demonstrated the muted transcriptional response of cytotoxins in another S. aureus lineage. Our findings provide the first evidence of distinct biological functions of the protein moiety and its associated linear peptide for a specific lipoprotein. Therefore, lipoproteins in S. aureus consist of three functional components: a lipid moiety, a protein moiety, and a small linear peptide, with putative different biological roles that might not only determine the outcome of host-pathogen interactions but also drive the acquisition of antibiotic resistance determinants.

Staphylococcus aureus treated with fosfomycin

Project description:Staphylococcus aureus is a highly adaptable human pathogen; therefore a constant search for new effective antibiotic compounds is being preformed. Gene expression profiling can be used to determine potential targets and mechanisms of action (MOA) of known or potential drugs. The goal of our study was a development of a focused transcriptome platform to be used for confirming the MOA of new chemical entities which are designed as inhibitors of Mur ligases. A model transcriptional profile was set up for well described inhibitor of MurA ligase, fosfomycin. Moreover, we wanted to identify the pathways and processes primarily affected by this compound. S. aureus ATCC 29213 cells were treated with low concentrations of fosfomycin (1 and 4 µg/ml, respectively) and harvested at 10, 20 and 40 minutes after treatment, respectively. RNA was isolated, transcribed, labeled and hybridized to S. aureus GeneChips, representing approximately 3000 S. aureus genes. Using meta-analysis of our results and the results in the S. aureus microarray database, we have confirmed that fosfomycin induces “cell wall stimulon” genes and were able to identify genes and pathways specifically modulated by fosfomycin.

Analysis of differential gene expression in C. elegans young adults exposed to two different bacteria: E. coli strain OP50 (control), wild-type Staphylococcus aureus RN6390. Samples were analyzed at 8 hours after exposure to the different bacteria. These studies identified C. elegans genes induced by pathogen infection.
