Project description:Circulating monocytes are main source for tumor-associated macrophages (TAMs) that control tumor growth, angiogenesis, metastasis and therapy resistance. We raised the questions how monocyte programming is affected by growing tumors localized in colon and rectal sections, and how treatment onsets affect monocyte programming in the circulation. Patients with rectal cancer and colon cancer were enrolled in the study. Peripheral blood monocytes were characterized by transcriptomic analysis using RNA sequencing. NGS analysis identified tumor-specific transcriptional programming of monocytes in all CRC patients compared to healthy individuals. The analysis also identified difference between colon and rectal cancer monocytes and chemotherapy effects on monocyte profile in rectal cancer.

Patients with rectal cancer and colon cancer were enrolled in the study. Spatial transcriptomic analysis was applied using GeoMX DSP-NGS. The key transcriptional difference between monocytes of patients with colon and rectal cancer was increased expression of PFKFB3, activator of glycolysis that is currently considered as therapy target for major solid cancers. PFKFB3-expressing monocyte-derived macrophages massively infiltrated tumor in colon. Nanostring technology identified correlation of PFKFB3 with amount and tumor-promoting properties of TAMs in colon but not in rectal cancer.

Project description:A major population of placenta macrophages represented throughout the pregnancy consists of CD14+ macrophages, but their characteristics remain badly understood. Here we purified from placentas at term CD14+ macrophages using positive selection. The phenotyping of CD14+ macrophages performed using flow cytometry revealed that placenta CD14+ macrophages expressed a series of markers distinct of those of circulating monocytes monocyte-derived macrophages. Placenta CD14+ macrophages spontaneously matured in multinucleated giant cells (MGCs) as demonstrated by size, number of nuclei display and specific cytoskeleton organization. Placenta CD14+ macrophages and MGCs were phagocytic cells but the potential of MGCs to mount an inflammatory response was lower than that of their precursors. Placenta CD14+ macrophages and MGCs stimulated with interferon and interleukin-4 were not polarized into typical M1 or M2 profiles. Placenta macrophages exhibited specific activation transcriptional programs. Indeed, principal component analysis and hierarchical clustering show that placental macrophages formed a distinct group from circulating monocytes and monocyte-derived macrophages. Among placenta macrophages, it was also possible to distinguish CD14+ macrophages and MGCs. In addition, networks based on gene interactions were clearly different in CD14+ macrophages and MGCs. Finally, the microenvironment of placenta CD14+ macrophages governs their differentiation into MGCs because CD14+ macrophages incubated with trophoblasts exhibited exarcerbated characteristics of MGCs and because the co-incubation of circulating monocytes from working women with trophoblast supernatants resulted into the formation of MGCs whereas monocytes from non-pregnant women incubated with trophoblast supernatants did not differentiate into MGCs. Taken together, these results clearly demonstrated specific feaures of placenta CD14+ macrophages. Three replicates of each of the following: 1. We found that F4/80hi CX3CR1hi (CD11b+CD103-) cells account for 80% of mouse colonic lamina propria (cLP) MHC-IIhi cells. Both CD11c+ and CD11c- cells within this population were identified as MPs based on multiple criteria, including a MP transcriptome revealed by microarray analysis. These MPs constitutively released high levels of IL-10 at least partially in response to the microbiota via an MyD88-independent mechanism. In contrast, cells expressing low to intermediate levels of F4/80 and CX3CR1 were identified as DCs, based on phenotypic and functional analysis and comprise three separate CD11chi cell populations: CD103+CX3CR1-CD11b- DCs, CD103+CX3CR1-CD11b+ DCs and CD103-CX3CR1intCD11b+ DCs. In non-inflammatory conditions, Ly6Chi monocytes differentiated primarily into CD11c+, but not CD11c- MPs. In contrast, during colitis, Ly6Chi monocytes massively invaded the colon and differentiated into pro-inflammatory CD103-CX3CR1intCD11b+ DCs, which produced high levels of IL-12, IL-23, iNOS and TNF. These findings demonstrate the dual capacity of Ly6Chi blood monocytes to differentiate into either regulatory MPs or inflammatory DCs in the colon, and that the balance of these immunologically antagonistic cell types is dictated by microenvironmental conditions. However, the role of miRNAs in the development of macrophages is just beginning to be explored. Here, we used microarray profiling to identify miRNAs involved in this molecular circuitry. In this study, a sample of RNA extracted from human monocytes were compared with a sample of RNA extracted from the same monocytes after 6 days of CSF-1 treatment.

Project description:So far, although differentiation and activation of Ag presenting cells, including monocytes, dendritic cells (DCs) and macrophages are accompanied by programmed remodelling of cell surface glycosylated molecules with potentially biologically important consequences, no global analysis of the expression of genes involved in glycan biosynthesis (essentially glycosyltransferases (GTs) and modification (mainly sulfotransferases) has been reported in these cells. Our project aims to obtain global information relating to the expression of genes encoding GTs and sulfotransferases in human monocytes, DCs and macrophages isolated or differentiated from the same donor. We also aimed to compare the expression profile of these genes in immature versus mature (immunostimulatory phenotype) DCs and macrophages (LPS stimulation, 4 and 18 hrs). To this end we have performed three independent experiments (n = 7 x 3 samples) using the dedicated gene microarray (glycogene-chip v3). Although, globally the data are interesting(please see the summary below), they lack significance and are not yet exploitable. The relatively high variability between samples probably results from the human donor sources given their heterogeneous genetic makeup and natural history. Thus, to reach significance, we believe that additional donors need to be analyzed. Of note, the time point “4hrs” can be omitted since it does not appear to be indicative. Three x 5 samples would be thus necessary. Utilizing a focused gene microarray, we intended to compare the glycosyltransferase (GT) and sulfotransferase gene expression profile of human monocytes relative to immature dendritic cells (DCs) and to macrophages and to characterize variations in DCs and macrophages undergoing maturation. Micro-array analysis indicated that monocytes express transcripts for a full set of enzymes involved in the biosynthesis of N- and O-glycans potentially elongated by poly-LacNAc chains with type II terminal sequences. Monocytes also express, at a very good level, genes encoding enzymes involved in glycosaminoglycan biosynthesis, but seem to have a limited capacity for glycolipid biosynthesis. Immature DCs and macrophages exhibit similar pattern of GT and sulfotransferase expression with only a limited number of cell type specific gene expression. Approximately 15 % of GT and sulfotransferase transcripts varied in both DCs and macrophages, the majority of them being up-regulated compared to monocytes. Strikingly, stimulation of DCs and Ms with lipopolysaccharide caused a general alteration in gene expression, in particular in DCs, mainly affecting genes found to be positively modulated during the differentiation steps. LPS treatment also caused a relatively minor increase in gene expression in macrophages whereas in DCs, the expression levels of a significant number of genes were enhanced. Validation of this analysis was partially provided by quantitative RT-PCR and flow cytometry of cell surface glycan epitopes. Collectively, this preliminary study implies an important modification of the pattern of glycosylation in differentiated and activated APCs with potential biological consequences.
