Transcriptome analysis of WT and lactoferrin knockout satellite cells grown for 4 days

ABSTRACT: Our phenotypic data suggest that lactoferrin knockdown reduces satellite cell proliferative capacity. The mouse muscle tissue was digested with collagenase, and satellite cells were obtained by flow sorting. We performed transcriptome analysis of wt and lactoferrin knockout cells grown for 4 days in vitro.

Project description:To identify the target genes of Dnmt1 in skeletal muscle satellite cells, we generated satellite cell specific Dnmt1 KO using Pax7-CreERT2 and Dnmt1 flox mice. After Tamoxifen (cKO) or vehicle (Control) treatment for 5 consecutive days, mice were euthanized and muscle satellite cells were collected by FACS sorting and total RNA were extracted.

They have described the relationship between PM2.5 (ambient fine particulate matter of <2.5 μm in diameter) and kidney diseases. Previous research has shown that lactoferrin showed renoprotective effects against some kidney diseases. In the current study, we analyzed the transcriptional profiles of PM2.5 and lactoferrin using RNA sequencing (RNA-seq) and identified the key signaling pathway in the human kidney proximal tubular epithelial cell line (HK-2 cells).

Project description:bulk sequencing outputs of Caco2 cells after exposure to permeability modifying and permeability rescuing agents. To identify the molecular drivers for methotrexate-induced barrier dysfunction, we conducted RNA sequencing on Caco2 spheroids treated with methotrexate and lactoferrin. Given our observation that barrier function was compromised as early as 4-6 hours after exposure to methotrexate, we isolated RNA from spheroids 4-hour post-treatment to capture the transcriptional events responsible for initiating the processes.
