Immune and inflammation-related factors alteration as a biomarker for predicting prognosis and responsiveness to PD-1 monoclonal antibody in patients with recurrent cervical cancer

ABSTRACT: Purpose: We aimed to elucidate the potential mechanisms of effective responsiveness to PD-1 monoclonal antibody and evaluate more reliable biomarkers for improving the predictive utility of the dominant populations of recurrent cervical cancer (RCC) to receive immunotherapy. Methods: Peripheral blood samples of PD-L1 positive patients with RCC treated with second-line PD-1 monoclonal antibody were collected prior to treatment initiation and after the treatment. Differentially expressed genes (DEGs) were analyzed between partial response (PR) patient and progressive disease (PD) patients.

Whole Exome Predictors Immune Checkpoint Therapy Response in Renal Cell Carcinoma

Project description:<p>Immune checkpoint therapies, including monoclonal antibodies to programmed cell death-1 (PD-1) and cytotoxic % lymphocyte associated protein-4 (CTLA-4), yield durable clinical responses across many tumor types, including metastatic melanoma, non-small cell lung cancer (NSCLC), and renal cell carcinoma (RCC). However, predictors of response to these therapies in RCC are still unknown. Genomic characterization of large clinical cohorts of patients treated with anti-CTLA-4 and anti-PD-1 agents in melanoma and NSCLC have suggested that high mutational burden, high neoantigen burden, and high expression of certain genes in pre-treatment tumors may be associated with patient response to these therapies. In this study, we sought to investigate genomic predictors of response to anti-PD1 therapy in metastatic RCC in two independent clinical cohorts using whole exome and whole transcriptome sequencing.</p>

Evaluation of anti-tumorigenic effects of anti-Lipocalin2 (anti-LCN2) monoclonal antibody against human cervical cancer xenografts in athymic nude mice

Project description:This study investigated the impact of an anti-LCN2 monoclonal antibody on STR-authenticated HeLa cells using a xenograft model in athymic nude mice. Treatment of tumors with anti-LCN2 antibody resulted in 39 – 60% reduction in tumor volume. RNA extracted from the tumors post-treatment was sequenced and aligned to human and mouse genome separately. Differential expression analysis of human mapped overexpressed genes in treated tumor revealed suppression of pro-tumorigenic TNF- and IL17 signalling pathways, whereas the corresponding mouse-mapped genes indicated pathways including activation of T-cells and T-cell mediated killing in treated tumors. M2 mouse macrophage markers showed a decline under treatment signifying tumor regression. This observed T-cell activation and debilitation of M2 mouse macrophages by anti-LCN2 underlies its potential as an immune sensitizer that can potentially devour tumor cells through T-cell mediated cytotoxicity.

mFOLFOX6+Bevacizumab+PD-1 Monoclonal Antibody in Local Advanced MSS CRC

Project description:Immunotherapy has achieved significant therapeutic effect in DNA mismatch repair-deficient or microsatellite instability-high (dMMR/MSI-H) colorectal cancer (CRC). However, for proficient mismatch repair(pMMR)/microsatellite stable(MSS) CRC, the curative effect of PD-1 monoclonal antibody was poor and most of the data came from stage Ⅳ patients with distant metastasis. Among the whole CRC patients, more than eighty-five percent were pMMR/MSS CRC. It would be very inspiring when major CRC patients(pMMR/MSS) could be benefit from immunotherapy. For T4NxM0 CRC patients, R0 resection was difficult to achieve. If the patients could not got R0 resection, which means the tumors were almost destined to recurrent and patients life time were counting down. Whether combined treatment of mFOLFOX6+ Bevacizumab+PD-1 monoclonal antibody could maximize the curative effect was still unknown. This study aims to evaluate the effect and safety of mFOLFOX6+ Bevacizumab+PD-1 monoclonal antibody treatment combinations in patients with local advanced(T4NxM0) pMMR/MSS CRC.
