Sheep longissimus dorsi (LD) muscle from 40 individual progeny belonging to six Poll Dorset industry sires

ABSTRACT: Expression profiling of sheep born to Australian industry sires with high and low genetic merit (Estimated Breeding Values or EBVs) for eye muscle depth (EMD). Progeny (40) from six Poll Dorset sires representing well defined extremes of EBVs for Eye Muscle Depth (low EBV EMD and high EBV EMD) were selected for analysis. The six sires were Australian industry sires with three sires representative of low EBV EMD and three representing high EBV EMD. Microarrays were used for transcription profiling of skeletal muscle samples taken from 40 individual progeny belonging to six Poll Dorset industry sires, with 3 sires extreme for high and 3 sires extreme for low muscling (based on EBV for EMD).

Liver gene expression patterns in pigs as indicators of serum plasma levels and meat quality atributes

Project description:In pigs, most of the lipogenic activity takes place in liver and the adipose tissue. Moreover, liver is responsible for the production and release of lipoproteins, which transports lipids from the liver to target tissues such as adipose and muscle. In the context of animal production, the amount and composition of lipoprotein have an implication in the accumulation of fat in adipose deposits but also in muscle in the form or intramuscular fat. These two events have consequences in the quality of the carcasses and meat. We have measured the level of liver gene expression in 104 commercial Duroc pigs belonging to two groups with extreme phenotypes for traits strongly related with lipid deposition and composition. This has allowed us to compare the physiological and metabolic implications of selecting for each of these extreme groups. This information has been complemented with genome-wide genotyping data in order to describe the implication of the liver transcriptome in the production traits which encompass the production and marketing of the products with consumer and industry-relevant added-value characteristics. 104 liver samples form pigs belonging to two groups of animals: HIGH group (n=53) had higher carcass, plasma and muscle fat content; LOW group (n=51) had lower carcass, plasma and muscle fat content

Project description:Beef tenderness is a complex trait of economic importance for the beef industry. Understanding the genetic and epigenetic mechanisms underlying this trait may help improve the accuracy of breeding programs and deliver a better product quality to consumers. However, little is known about epigenetic effects in the muscle of Bos taurus and their implications in tenderness, and no studies have been conducted in Bos indicus. Therefore, we analyzed Reduced Representation Bisulfite Sequencing (RRBS) to search for differences in the methylation profile of Bos indicus skeletal muscle with extreme values for beef tenderness (tender = 6 animals, tough = 6 animals).

Sire modulates developmental kinetics and transcriptome in the bovine embryo

Project description:The impact of sire on pre-implantation embryonic development in cattle remains poorly understood. This study evaluated differences in embryos produced from sires with varying capacities to produce blastocysts. Sires classified as high (HP) and low-performing (LP) based on their ability to produce embryos were used in order to better understand how sire regulates embryonic development. By monitoring embryo development, it was determined that the most common arrest stage was the 5-6 cell stage. Embryos (4-6 cells) from HP and LP sires were analyzed for autophagic activity, where embryos for LP sires exhibited increased autophagy than HP-derived embryos. Transcriptome analysis of 4-cell embryos found that embryos from LP sires might have issues in sperm mitochondrial clearance, histone retention, and DNA damage, while HP sires have increased expression of genes involved in transcription, chromosome segregation, and cell division. In conclusion, LP sires have an increased proportion of embryos arresting at the 5-6 cell stage, and these embryos have higher rates of cellular stress due to paternal contributions from the spermatozoon.

Transcription profiling of mouse heart from Emd KO and controls

Project description:The present research is devoted to the identification of gene(s) severely affected by EMD mutations, leading to striated muscle laminopathies and more specifically the cardiomyopathy. For this purpose, we developped a large-scale gene expression approach on heart and skeletal tissues from Emd KO mouse model. Experiment Overall Design: In the project presented here we performed differential expression in heart from a mouse model of EDMD: a Emd KO mouse created via homologous recombination by Y.Hayashi in Japan (Ozawa, 2006). The emd knock-out mouse showed significantly delayed PR time, that could correspond to impaired atrial-ventricle node conduction. Genes were identified as differentially expressed if they met a false discovery rate threshold of 0.05 in a two-sample t-test (q-value) and showed at least a two-fold difference in expression independent of absolute signal intensity.

Yet, the miRNA gene regulatory network for the synthesis of milk protein in mammary is poorly understood. In this study, the hypothesis was that miRNAs have potential roles in bovine milk protein production. Using miRNA-seq and RNA-seq, we investigated the miRNAs profiles of mammary glands from 12 Chinese Holstein cows with six cows at peak of lactation and six in non-lactating period, from which three cows were in high and three in low milk protein percentage.
