Adipocyte thyroid hormone receptor β determines the intracellular usage and fate of glucose and governs systemic homeostasis

ABSTRACT: To investigate the effect of thyroid hormone and its receptor on glucose and lipid metabolism in white adipose tissue (WAT), and the effect of thyroid hormone and its receptor on WAT regulation on glucose metabolism and obesity. We constructed mice with adipose tissue-specific knockout of thyroid hormone receptor beta（ATRβKO mice）. ATRβKO mice were given T3 stimulation or high-fat feeding. We then performed gene expression profiling analysis using data obtained from RNA-seq of White adipose tissue from knockout ATRβKO mice treated with T3 or HFD feeding.

Microarray Analysis of the Delipidation of White Adipose Tissue of Mice Fed Conjugated Linoleic Acid

Project description:The white adipose tissue (WAT) rapidly loses mass when mice are fed a diet containing trans-10, cis-12 conjugated linoleic acid (t10c12 CLA). A microarray analysis of WAT due to CLA feeding was performed to better define the processes and genes involved. WAT weight decreased by ca. 80% over 17 days of feeding a 0.5% t10c12 CLA diet. The lipid volume decreased by 90% and the number of adipocytes and total cells were reduced by15% and 47%, respectively. Microarray profiling of replicated pools of control and treated mice (n=140) at seven time points over the 17day feeding indicated between 2798 to 4318 genes showed mRNA changes of 2-fold or more. Transcript levels for genes of glucose and fatty acid import or biosynthesis were significantly reduced. A prolific inflammation response was indicated by the 2 to100-fold induction of many cytokine transcripts, including those for IL-6, IL1?, TNF ligands, and CXC family members We used microarrays to characterize transcriptome changes in mouse adipose tissue due to feeding trans-10 cis-12 linoleic acid . Adipose METTL14-elicited N6-methyladenosine promotes obesity, insulin resistance, and NAFLD by suppressing β adrenergic signaling and lipolysis

Project description:Obesity is associated with impaired β-adrenergic receptor (Adrb1-3) signaling and lipolysis, leading to aberrant white adipose tissue (WAT) growth. WAT research has been centered on transcriptional and posttranslational regulations, but posttranscriptional regulation and mRNA modifications are poorly understood. Here, we unveil a METTL14/N6-methyladenosine (m6A) paradigm guiding β-adrenergic signaling and lipolysis. METTL14 complex installs m6A on RNA, regulating mRNA fate and translation. We found that feeding and insulin increased adipose Mettl14 and m6A levels. Adipose Mettl14 and m6A were upregulated in high fat diet (HFD)-induced obesity. Ablation of adipose Mettl14 decreased Adrb2, Adrb3, Atgl (encoding lipase), and Cig-58 (Atgl activator) transcript m6A contents while increasing their translation and protein levels, thereby enhancing adipose β-adrenergic signaling and lipolysis. Consequently, adipocyte-specific Mettl14 knockout mice were resistant to HFD-induced obesity, insulin resistance, glucose intolerance, and NAFLD. These results unravel a METTL14/m6A-based epitranscriptomic mechanism governing β-adrenergic signaling, lipolysis, and adipose growth in health and disease.

The ApoE*3Leiden.human Cholesteryl Ester Transfer Protein (ApoE3L.CETP) mouse model manifests several features of the MetS upon high fat diet (HFD) feeding. Comparison of SSAT overexpressing mouse models

Project description:It has been shown that in a mouse model overexpressing spermidine/spermine N1-acetyltransferase (SSAT) in whole body the mice have improved glucose homeostasis, reduced white adipose tissue (WAT) mass and high basal metabolic rate. In this study we investigated the glucose and energy metabolism in another SSAT overexpressing mouse model (MT-SSAT) and in addition created a novel mouse model that has adipose tissue specific SSAT overexpression (aP2-SSAT) in order to elaborate the role of enhanced polyamine catabolism in WAT. In this experiment, gene expression profiles of WAT from wild type, SSAT, MT-SSAT and aP2-SSAT mice were studied.

Glis3 plays a crucial role in thyroid hormone production

Project description:The serum hormone levels including T3 and T4 were dramatically decreased in Glis3-null mice due to reduced production of thyroid hormones in thyroid. Gene expression profile and EdU incorporation analysis between WT and Glis3-null mice showed that the cell proliferation was greatly reduced in Glis3-null thyroid. Goitergenic diet (low iodine diet; LID) dramatically enhanced serum TSH levels in both WT and Glis3-null mice, however thyroid goiter was observed in WT mice but not in Glis3-null mice. A subset of genes associated with thyroid hormone production including Pendrin (Slc26a4), Nis (Na+/I– symporter, Slc5a5), Duoxa2 (dualoxidase A2), Tpo (thyroperoxidase), and Dio1 (Deiodinase1) was significantly induced in WT but not in Glis3-null mice by LID feeding.
