Astroglial Hmgb1 regulates postnatal astrocyte morphogenesis and cerebrovascular maturation

Project description:Astrocytes are intimately linked with brain blood vessels, a relationship critical for neuronal function. However, astroglial factors driving these physical and functional associations during postnatal brain development have yet to be identified. By characterizing structural and transcriptional changes in mouse cortical astrocytes during the first two postnatal weeks, we find that high-mobility group box 1 (Hmgb1), normally upregulated with injury and involved in adult cerebrovascular repair, is highly expressed in astrocytes at birth and then decreases rapidly. Astrocyte-selective ablation of Hmgb1 at birth affects astrocyte morphology and endfoot placement, alters distribution of endfoot proteins connexin43 and aquaporin-4, induces transcriptional changes in astrocytes related to cytoskeleton remodeling, and profoundly disrupts endothelial ultrastructure. While lack of astroglial Hmgb1 does not affect the blood-brain barrier or angiogenesis postnatally, it impairs neurovascular coupling and behavior in adult mice. These findings identify astroglial Hmgb1 as a key player in postnatal gliovascular maturation.

Proteomics of TurboID-induced biotinylated proteins from astrocyte and astrocyte endeet after acute peripheral inflammation

Project description:The brain vasculature is tightly regulated to control the exchange of substances between the periphery and the brain parenchyma at an interface called the blood-brain barrier (BBB). Astrocytes are glial cells that wrap the brain vasculature with a specialised structure called the endfoot, which is an integral component of the BBB. We generated adeno-associated viruses (AAVs) that express a promiscuous biotin ligase, TurboID, specifically in astrocytes, and developed a method that allows us to define the in vivo protein composition of the astrocyte endfoot with negligible contamination of other cells or other compartments of the astrocyte. Here, we isolated the biotinylated proteins from both the whole astrocyte and astrocyte endfeet from mice with acute peripheral inflammation.

Next Generation Sequencing Facilitates Quantitative Analysis of Wild Type and Nrl-/- Retinal Transcriptomes

Project description:Purpose: To identify the role of glycosylation of dentin matrix protein1 (DMP1), S89G-DMP1 point mutation mouse model was created with changing S89 to glycine(S89G). RNA sequencing were performed to compare the transcriptome differences between the neural stem cells or astrocytes separated form S89G-DMP1 and WT mice. Methods: mRNA profiles of astrocytes and neural stem cell separated from S89G-DMP1 and WT were generated by RNA sequencing. The sequence reads that passed quality filters were analyzed at the transcript isoform level with TopHat2(for astrocytes) or with HISAT2o (for neural stem cells). Results: Differential expression analyses with DESeq2 use raw read counts: 762 transcripts were downregulated and 991 transcripts were up-regulated in S89G-DMP1 astrocytes; 475 transcripts were downregulated and 343 transcripts were up-regulated in S89G-DMP1 neural stem cells. All the differential expressed genes were used for Heatmap analysis and KEGG or GO enrichment analyses. Conclusions: Our study represents the detailed transcriptome changes of astrocytes and neural stem cells with deglycosylated DMP1.

Mitochondrial Complex I inhibition enhances astrocyte responsiveness to pro-inflammatory stimuli

Project description:Inhibition of the mitochondrial oxidative phosphorylation (OXPHOS) system can lead to metabolic disorders and neurodegenerative diseases. In primary mitochondrial disorders, reactive astrocytes often accompany neuronal degeneration and may contribute to neurotoxic inflammatory cascades that elicit brain lesions. The influence of mitochondria to astrocyte reactivity as well as the underlying molecular mechanisms remain elusive. Here we report that mitochondrial Complex I dysfunction promotes neural progenitor cell differentiation into astrocytes that are more responsive to neuroinflammatory stimuli. We show that the SWI/SNF/BAF chromatin remodeling complex takes part in the epigenetic regulation of astrocyte responsiveness, since its pharmacological inhibition abrogates the expression of inflammatory genes. Furthermore, we demonstrate that Complex I deficient human iPSC-derived astrocytes negatively influence neuronal physiology upon cytokine stimulation. Together, our data describe the SWI/SNF/BAF complex as a sensor of altered mitochondrial OXPHOS and a downstream epigenetic regulator of astrocyte-mediated neuroinflammation.

Macrophage and MDA-MB-231 coculture and mitochondrial transfer

Project description:The goals of this study were to determine the effects of lateral transfer of mitochondria on recipient cell biology. We therefore collected cancer cells with and without macrophage mitochondria, and macrophages with and without cancer cell mitochondria, and analyzed transcriptomes by single-cell RNA-sequencing. Through our analysis, we found that cancer cells that received macrophage mitochondria have an enrichment of transcripts associated with cell cycle progression compared to cancer cells without macrophage mitochondria. We further validated these findings with wet lab experiments using cell proliferation markers.
