XBP1-mediated activation of STING signaling in macrophages contributes to the progression of liver fibrosis

ABSTRACT: Our findings demonstrate that XBP1 controls macrophage cGAS/STING/NLRP3 activation by regulating macrophage self-mtDNA cytosolic leakage via BNIP3-mediated mitophagy modulation, providing a novel target against liver fibrosis.

Mitophagy mitigates mitochondrial dysfunction-triggered activation of mtDNA-cGAS-STING inflammation during autoimmune thyroiditis

Project description:Autoimmune thyroiditis (AIT) is a common endocrine disorder characterized by the destruction of thyroid follicles and lymphocytic infiltration, yet its pathogenic mechanisms are not fully understood. This study reveals that chronic oxidative stress during thyroid hormone synthesis in follicular epithelial cells leads to mitochondrial damage and dysfunction. A key finding is the cytosolic leakage of mitochondrial DNA (mtDNA) resulting from this damage, which activates the cGAS-STING innate immune signaling pathway, driving the production of proinflammatory cytokines and exacerbating disease pathogenesis. Conversely, we identify a protective, adaptive mitophagy pathway mediated by the ATM-CHK2 axis. Upon mitochondrial stress, activated CHK2 translocates to mitochondria and phosphorylates the selective autophagy adapter TAX1BP1 at Ser722. This phosphorylation event enhances TAX1BP1's affinity for K63-linked ubiquitin chains on damaged mitochondria, facilitating their clearance via PINK1/Parkin-dependent mitophagy. Impairment of this ATM-CHK2-TAX1BP1 pathway, or genetic ablation of PINK1/Parkin, abrogates mitophagy, leading to amplified mtDNA leakage and hyperactivation of cGAS-STING signaling. Crucially, pharmacological inhibition of STING with C-176 significantly attenuates thyroiditis progression in mouse models.

Project description:Diabetic complications frequently arise in mechanically stressed regions, yet the molecular links between biomechanical forces and metabolic dysfunction remain unclear. Here, we demonstrate that mechanical stress induces glucose accumulation and downstream metabolic stress in keratinocytes, leading to mitochondrial injury and inflammation. Mechanistically, Piezo1 activation led to intracellular glucose overload and excessive advanced glycation end-products (AGEs) accumulation, which induced mitochondrial DNA (mtDNA) leakage into the cytosol and subsequently activated the cGAS–STINGsignaling cascade. Keratinocyte-specific Piezo1 deletion markedly reduced AGEs accumulation and preserved mitochondrial integrity, while Stimulator of interferon genes (STING) ablation reproduced similar downstream protective effects. Keratinocyte-specific deletion of the mechanosensor Piezo1 reduces advanced glycation end-products (AGEs) accumulation, preserves mitochondrial integrity, and dampens inflammatory signaling. Further, we show that this injury cascade involves cytosolic leakage of mitochondrial DNA (mtDNA) and activation of the cGAS–STING pathway. Stimulator of interferon genes (STING) ablation recapitulates the protective phenotype, confirming its downstream role. Notably, we identify Cortistatin (CST), an endogenous neuropeptide, as a previously unrecognized inhibitory ligand of Piezo1. CST binding attenuates calcium intake and glucose accumulation under mechanical stress, conferring notable protection in vitro and in diabetic ulcer (DUs) models. These findings uncover a CST–Piezo1–STING regulatory axis that integrates mechanical and metabolic cues to drive keratinocyte dysfunction in diabetes.

mtDNA leakage promotes neuron-glia crosstalk to induce epilepsy by cGAS-STING-driven neuroinflammation and serine metabolic reprogramming

Project description:Epilepsy is increasingly recognized as a disorder involving metabolic dysregulation beyond neural hyperexcitability, yet the underlying metabolic mechanisms remain poorly defined. Here, we identify a mitochondrion-immunity-metabolism axis that drives spontaneous chronic epilepsy. Brain-specific deletion of Mic19 impairs mitochondrial cristae structure and mitochondrial integrity in neurons, leading to activation of the Z-mtDNA–ZBP1–RIPK3-MLKL axis and p-MLKL-mediated pore formation on the mitochondrial membrane. This process results in cytosolic and extracellular leakage of mitochondrial DNA (mtDNA), which is subsequently taken up by microglia and triggers cGAS-STING-dependent inflammatory signaling. The resulting neuroinflammation promotes sustained activation of astrocytes. Critically, reactive astrocytes undergo profound metabolic reprogramming, marked by upregulated glycolysis and enhanced L-serine biosynthesis. Astrocyte-derived L-serine is subsequently transferred to neurons and converted into D-serine, a key NMDA receptor co-agonist that enhances neuronal excitability. This metabolic shift in astrocytes exacerbates excitotoxicity and sustains epileptic activity. Importantly, pharmacologic inhibition of STING with H-151 treatment markedly suppresses seizures, reinforcing the therapeutic potential of targeting immunometabolic crosstalk in epilepsy. Our findings reveal that mtDNA-mediated cGAS-STING activation and D-serine act as important drivers of epilepsy initiation, offering new mechanistic insights into neuron-microglia-astrocyte crosstalk and highlighting immunometabolic modulation as a promising therapeutic strategy for epilepsy.

Project description:Macroautophagy decreases with age, and this change is considered a hallmark of the aging process. It remains unknown whether mitophagy, the essential selective autophagic degradation of mitochondria, also decreases with age. In our analysis of mitophagy in multiple organs in the mito-QC reporter mouse, mitophagy was either increased or unchanged in old versus young mice. Transcriptomic analysis showed marked upregulation of the type I interferon response in the retina of old mice, an effect that correlated with increased levels of cytosolic mtDNA and activation of the cGAS/STING pathway. Crucially, these same alterations were replicated in primary human fibroblasts from elderly donors. In old mice, pharmacological induction of mitophagy with urolithin A attenuated cGAS/STING activation and ameliorated deterioration of neurological function. These findings point to induction of mitophagy as a novel strategy to decrease age-associated inflammation and increase healthspan.

Suppression of antiviral innate immunity by the COX2-PGE2-PKA signaling axis by inhibiting the cytosolic mtDNA-dependent activation of STING

Project description:The innate immune cGAS-STING pathway is activated by cytosolic doubled-stranded DNA (dsDNA) to induce type I interferon response, which is essential for antiviral and antitumor immunity. However, intrinsic STING activation during viral infection or within the tumor microenvironment is often insufficient to achieve complete viral clearance or tumor regression, indicating the presence of additional mechanisms that evades its activity. Here, we identified COX2/PGE2 as an intrinsic negative regulator of STING activation, particularly in response to cytosolic mitochondrial DNA (mtDNA) generated during HSV-1 infection. Mechanistically, we demonstrated that PGE2 signals through the EP4-PKA axis to induce mitophagy and facilitate the clearance of immunostimulatory cytosolic mtDNA, thereby dampening STING-mediated antiviral response. Furthermore, we discovered STOML2 as a direct downstream target of PKA, thereby linking mitochondrial quality control to innate immune signaling. Together, our findings established the COX2/PGE2/PKA axis as an intrinsic negative regulator of STING signaling, which may be targeted to potentiate STING-mediated antiviral and antitumor immunity.

The role of XBP1 in regulating the progression of non-alcoholic steatohepatitis [XWT/XKO BMDMs]

Project description:Our study demonstrated that XBP1 is upregulated in liver tissues of patients with NASH. Conditional knockout of Xbp1 in the hepatocytes resulted in decreased lipid accumulation in mice. Genetic specific deletion of Xbp1 in macrophages ameliorated nutritional steatohepatitis and fibrosis in mice by reducing the secretion of pro-inflammatory cytokine, increasing M2 macrophage polarization, and decreasing TGF-β1 expression. Pharmacological inhibition of XBP1 protects against steatohepatitis and fibrosis, highlighting a promising strategy for NASH therapy.
