Grape berry response to heatwaves in a climate change scenario: a physiological, transcriptional, and metabolic overview

ABSTRACT: Grapevines cv Sauvignon Blanc were subjected to different stress regimes by simulating the most common extreme weather events occurring within the current climate change scenario. During spring, just before bud break, the vines were either flooded or kept under normal conditions. Both vines were then split in two groups and put in differnt tunnels, either undergoing a simulated heatwave or not. Berry samples were collected from the four groups of plants (control, only flooded, only heatwave, flooded + heatwave) at different timepoints before, during and after the stress. Transcriptomic analyses were carried out on these samples along with some metabolomic assessments to characterize the response to stress in the different samples and the effect of the combined stresses.

Comparative proteomics of salt stressed own rooted and 110R grafted Thompson Seedless grapevines

Project description:Salt stress is a rising threat to agriculture system. The accumulation of salts near the plant roots hampers the normal uptake of water causing osmotic stress and ionic toxicity to the plant. Thompson Seedless is a popular table grape variety of Vitis vinifera L., which is sensitive to salt stress when grown on its own roots; grafting it onto a wild rootstock such as 110 Richtor (110R) makes it tolerant to salt stress. In the present study, shotgun-proteomics approach was used for the investigation of salt stress induced molecular response of own rooted and 110R grafted Thompson Seedless grapevines. A salt stress experiment was conducted on sixteen month old potted grapevines. The grapevines were treated with 150mM NaCl solution for seven days and the control vines were irrigated with tap water. The young leaf samples were collected from control and treated vines at three time-points viz. 6 hours, 48 hours and 7 days of salt stress. The stress responsive proteins identified through statistical analysis revealed a distinct response to salinity in both the vines.

Response of Vitis vinifera to Eutypa lata infection

Project description:Three grapevines cultivars (Merlot, Cabernet-Sauvignon and Ugni Blanc) were infected by E. lata. The expression profiles of the wood part near the infection point were determined for both infected and non infected plant for each cultivars with Nimblegen microarrays vitis. Three plants were used for biological replicates. Comparisons between infected and non infected conditions allow, for each cultivars, the identifcation of genes which the expression is modified by E. lata.

Severe stunting symptoms upon nepovirus infection are reminiscent of a chronic hypersensitive-like response in grapevine

Project description:Purpose and strategy: Grapevine fanleaf virus (GFLV) causes variable symptoms in most vineyards worldwide. To better understand GFLV-grapevine interactions in relation to symptom development, field and greenhouse trials were conducted with a grapevine genotype that exhibits distinct symptoms in response to a severe and a mild strain of GFLV. Results: After validation of the infection status of the experimental vines by high throughput sequencing, the transcriptomic and metabolomic profiles in plants infected with the two viral strains were tested and compared by RNA-Seq and LC-MS, respectively, in the differentiating grapevine genotype. In vines infected with the severe GFLV strain, 1,023 genes, among which some are implicated in the regulation of the hypersensitive-type response, were specifically de-regulated, and a higher accumulation of resveratrol and phytohormones was observed. Interestingly, some experimental vines restricted the virus to the rootstock and remained symptom-less. Our results suggest that GFLV induces a strain- and cultivar-specific defense reaction similar to a hypersensitive reaction. This type of defense leads to a severe stunting phenotype in some grapevines whereas others are resistant. This work is the first evidence of a hypersensitive-like reaction in grapevine during virus infection. Conclusion: Our results suggest that GFLV induces a strain- and cultivar-specific defense reaction similar to a hypersensitive reaction. Cluster thinning effect on Cabernet Sauvignon berry ripening

Project description:The impact of yield on fruit and wine quality has been studied extensively; vine response to increased yield is well known, but responsible mechanisms – including the regulation of pathways responsible for color, aroma and mouthfeel – have not been characterized at the level of genetic regulation. To examine the impact of crop load on transcriptomic and metabolomic changes during grape berry maturation vines from two cluster thinning treatments (50% or 75% of clusters removed following fruit set) were compared to control vines (no cluster thinning). Treatments were applied to mature Cabernet Sauvignon grapevines over three consecutive vintages. Agronomic parameters, including yield per vine and the yield to pruning weight ratio, as well as fruit and wine composition, were determined at harvest. Fruit transcriptomic and metabolomic changes were measured in berry samples collected from each treatment at 7 to 10 day intervals from fruit set to harvest. The patterns of berry development by weight, sugar accumulation, and malic acid degradation indicated that vines responded differentially to crop load reduction by accelerating berry maturation progress. RNA-Seq analysis, and untargeted GC-MS and UHPLC-QTOF-MS -based metabolomic approaches were used. The indication of the acceleration of maturation transcriptional programs and of metabolite accumulation in the treated vines was suggested by multivariate analyses. To identify the transcriptional key triggers of this response we applied a two-regression step statistical approach which identified genes modulated over development dependent on the crop load. Comparing vintages we were able to estimate the environmental influence on the crop load treatments among years. The application of this multifaceted approach allowed the construction of robust models describing the impact of crop load on the genetic regulation of grape maturation.

High temperature effect on Cabernet Sauvignon berries

Project description:The experiment was conducted using four 11-year-old potted grapevines of V. vinifera L. cv. Cabernet Sauvignon grafted on SO4 (Selection Oppenheim No. 4) grown in a phytotron. Vines were trained on a Guyot trellising system and each vine carried 4-10 clusters of grapes. The experiment started approximately 1 week before veraison, when berry softening started, and continued to fruit maturity. The two temperature regimes consisted of a high day (06.00–20.00 h) temperature (max.35C) and a control (max. 25C). Under both conditions, the night-time (20.00–06.00 h) temperature was 20C.
