Ablation of ER stress-linked kinase PERK in cancer cells elicits protective T cell immunity by paraptosis immunogenic cell death-induced Type-I IFN

ABSTRACT: C57BL6 wildtype mice were injected with B16F10 melanoma transduced with either untargeted sgRNA (Scramble) or PERK (EIF2AK3) knockout (KO) sgRNA. At day 17, tumors were harvested to isolate cKit+ (CD117) precursors via magnetic bead enrichment. These cKit expressing precursors were then sequenced.

ISR pathway activation signatures in MEFs: GCN2 and PERK

Project description:In response to different cellular stressors, the ISR kinases, PERK, PKR, HRI and GCN2, activate downstream transcriptional programs. While the core ISR transcription program is well characterized, markers that are specific to each individual ISR kinase activation pathway are not known. To identify markers that are induced by PERK or GCN2, but not the other ISR kinases, we subjected WT, GCN2-/-, and PERK-/- MEFs to amino acid starvation (RPMI 1640 SILAC -Lys -Arg) or Thapsigargin (200nM) treatment for 6 hours to activate the GCN2 and PERK pathways, respectively and performed RNA sequencing.

PERK protein kinase facilitates keratinocyte collective cell migration by engagement with cell adhesion molecules independent of its kinase activity

Project description:Proximity labeling experiments (BioID) were performed on Human NTERT keratinocytes depleted of PERK (EIF2AK3, PERK-KO) re-expressing a WT PERK containing a C-terminal UltraID tag. PERK-KO and PERKUltraID cells were treated with differentiation medium containing 50 uM of exogenous Biotin for 6 h, washed three times with ice-cold PBS and harvested on ice with a modified RIPA buffer solution (25mM Tris-HCl, pH 7.5, 150 mM NaCl, 1% NP-40, 0.5% Sodium Deoxycholate, 1mM EDTA, 1x HALT protease/phosphatase inhibitor cocktail). Samples were then sonicated and incubated at 4C with rotation for 30 minutes, followed by centrifugation step to pellet cell debris. Total protein concentration was measured, and the samples were normalized to 1 mg/ml to a final volume of 1 ml. Normalized samples were incubated with 40uL of trypsin-resistant streptavidin beads with rotation overnight at 4C. The next day, the beads were collected by magnetic separation and washed three times with an IP wash solution (25 mM Tris-HCl, pH 7.5, 150 mM NaCl, 0.5% Triton X-100), followed by two washes with PBS. Finally, the beads were resuspended with 100 ul of PBS and submitted on ice for preparation and mass spectrometry analysis by the Indiana University School of Medicine Center for Proteome Analysis.

Project description:Here we report that the Th2 cytokine IL-4 and the tumor microenvironment activated protein kinase RNA-like ER kinase (PERK) ER stress signaling cascade to promote immunosuppressive M2 activation and proliferation. Lacking PERK signaling impeded mitochondrial respiration and lipid oxidation critical for M2 macrophages. In addition, PERK activation mediated the upregulation of PSAT1 and serine biosynthesis via the downstream transcription factor ATF4.

Effect of depletion of PERK on gene expression in T cells after polyclonal or allogeneic stimulation

Project description:PERK differentially regulates T cell allogeneic and anti-tumor responses. To further elucidate the mechanism, we performed transcriptomics analyses via bulk RNA sequencing on WT and PERK-deficient T cells after polyclonal or allogeneic stimulation. We here report that endoplasmic reticulum-associated degradation (ERAD)-associated genes, including Sel1l and Erlec1, were significantly increased in PERK conditional knock out (cKO) compared with WT T cells upon allogeneic but not polyclonal stimulation. SEL1L-HRD1 axis plays a central role in ERAD pathway, we also observed that PERK binds to SEL1L in activated human T-cell line. By using PERK-/-SEL1L-/- mice, we eluciated that PERK regulates T-cell allogeneic responses and GVHD induction through SEL1L-mediated ERAD pathway.

Selective activation of PERK signaling mediates fibroblast education and tumor progression by a CRELD2-dependent mechanism

Project description:It is now well-accepted that cancers co-opt the microenvironment to support their growth. However, the molecular mechanisms underlying cancer-microenvironment interactions remain poorly defined. We have found that Rho-associated kinase (ROCK) activity in the epithelial component of mammary tumors selectively actuates Protein kinase R-like endoplasmic reticulum kinase (Perk), causing the recruitment and persistent education of tumor-promoting cancer-associated fibroblasts (CAFs), a key component of the cancer microenvironment. Analysis of tumors from mice and human patients identified Cysteine-rich with EGF-like domains 2 (CRELD2), as the paracrine factor underlying PERK-mediated CAF-education downstream of ROCK. CRELD2 expression was found to be regulated by ATF4 downstream of the PERK pathway and knock-down of CRELD2 prevented tumor progression, demonstrating that the paracrine ROCK-PERK-ATF4-CRELD2 axis is a promoter of breast cancer progression and suggesting new therapeutic opportunities.
