ABSTRACT: Identification of ROS induced genes on dendritic cells Dendritic cells were incubated for 15 min with or without a ROS inhibitor (DPI), washed extensively and incubated for 30 min with a chemical allergen (DNFB), hydrogen peroxide, and vehicle alone in HBSS containing DPI or vehicle. After washed extensively, the samples were post-incubated for 5.5 h with DNFB, hydrogen peroxide, or vehicle in complete culture medium containing DPI or vehicle.

Project description:We collected protoescoleces (PSCs) from individual bovine cysts. Then we incubated 20 uL of PSCs in RPMI medium supplemented with 10% fetal bovine serum with 2.5 mM hydrogen peroxide (treated) or without hydrogen peroxide (Control) for 2 h at 37 °C. After that we collected PSCs by sedimentatios in 1.5 mL tube and washed 4 times with PBS at 37°C.

Role of the ArcAB two-component system in oxidative stress

Project description:Salmonella enterica serovar Typhimurium is a Gram-negative bacterium, facultative anaerobe and intracellular pathogen that causes enteric fever in mice. Once orally ingested, Salmonella invades and traverse the mucosal intestinal epithelia, where it is phagocytized by specialized cells including macrophages, dendritic cells and neutrophils. Within these cells, the bacterium is kept in a compartment termed Salmonella containing vacuole where it is exposed to different adverse conditions including nutrient deprivation, acid pH, reactive oxygen (ROS) as well as nitrogen (RNS) species and low oxygen levels. Among the signals encountered by the bacteria, oxidative stress is one of the main challenges that it has to overcome in order to survive. In this context, the OxyR and SoxRS proteins are the most studied regulators involved in response to ROS. However, in the past years growing evidence suggests that the ArcAB two-component system might play a key role in modulating gene expression in response to ROS. Furthermore, the global regulator ArcA is required for the resistance of Escherichia coli, S. Enteritidis and Typhimurium to hydrogen peroxide (H2O2), however, the ArcA regulon under oxidative stress conditions remains elusive. Therefore, the aim of this work was to demonstrate that ArcAB regulates the expression of genes in response to hydrogen peroxide and determine the ArcA regulon under this condition. To achieve this, we evaluated transcriptomic changes in strain 14028s, ∆arcA in response to H2O2. Total RNA was harvested from three biological replicates of wt and arcA mutant cultures exposed or unexposed to 1.5 mM hydrogen peroxide for 20 min in LB medium.

Transcriptional responses in Arabidopsis seedlings after hydrogen peroxide treatment

Project description:Excessive levels of reactive oxygen species (ROS) cause cellular stress through damage to all classes of macromolecules and result in cell death. However, ROS can also act as signaling molecules in various biological processes. In plants, ROS signaling has been documented in environmental stress perception, plant development and cell death amongst others. Knowledge on the regulatory events governing ROS signal transduction is however still scratching the surface. To further elucidate the transcriptional response and regulation upon ROS accumulation we supplemented Arabidopsis seedlings with a 10mM hydrogen peroxide (H2O2) solution to trigger oxidative stress. After growth of 7 days, hydrogen peroxide (H2O2) was added to a final concentration of 10mM. Control plants were treated with the same volume of H2O. Seedlings were grown for 24h under the same controlled conditions. Design: 3 replicates x 2 conditions (7+1 day H2O or 7+1 day H2O2)

Transcriptome profiling of H. polymorpha in response to hydrogen peroxide exposure

Project description:To gain insight into the basic mechanism of Hydrogen peroxide detoxification in the methylotrophic yeast, H. polymorpha, we analyzed changes in transcriptional profiles in response to hydrogen peroxide exposure. Total RNA samples were collected from H. polymorpha cells after 30 min incubation with 0.5mM hydrogen peroxide. Using the RNA sample obtained prior to hydrogen peroxide addition as a reference, the differential fluorescence intensities of each RNA sample prepared at the indicated time was measured after labeling with Cy3 or Cy5 fluorochromes. For all analyses, we performed dye swapping experiments to avoid dye bias.

Identification of ciliary neurotrophic factor receptor alpha as a mediator of neurotoxicity induced by extracellular alpha-synuclein

Project description:MES cells were treated with vehicle or 250 nM extracellular alpha-synuclein and plasma membrane proteins were labeled with biotin, followed by affinity purification. The biotinylated enriched membranous proteins were incubated with an alpha-synuclein peptide array membrane, washed extensively with TBS-T, followed by incubation with avidin-HRP. The positive spots were excised and the bound proteins were eluted. The elutions from consecutive spots were combined before trypsin digestion and analysis of peptides by LC-MS/MS.

Defining the response of stem cells to ROS is critical to understanding the unique sensitivity of stem cells to ROS-mediated transformation and tumor initiation. We examined both the immediate transcriptional and phospho-proteomic responses of murine ES cells induced by a single brief dose of hydrogen peroxide (H2O2) to create a more complete understanding of cell response to ROS.

Timecourse analysis of fission yeast CSL mutants treated with hydrogen peroxide

Project description:Fission yeast CSL mutants (cbf11 and cbf12 knock-outs) show altered sensitivity to hydrogen peroxide. To help elucidate the molecular basis of this phenomenon, we analyzed wild-type and mutant transcriptomes before stress and as a timecourse after treatment with hydrogen peroxide. Cells were grown to exponential phase in YES medium, untreated samples were collected (time 0), cells were treated with 0.74 mM hydrogen peroxide and harvested after 15 and 60 min post treatment. Total RNA was isolated, labelled and hybridized to custom-made two-colour microarrays. Each sample was hybridized against reference created by pooling equal aliquots from all 9 samples.
