ABSTRACT: Interleukin-18 (IL-18) was one of the inflammatory cytokines and related to major depressive disorder (MDD) and dementia. We previously investigated IL-18 knockout (Il18−/−) mice developed impaired learning and memory, more anxiety, and less motivation comparing to Il18+/+ mice. In the Il18−/− mice with acute stress, both the immobility time in the forced swim test and the tail suspension test were decreased, even though no difference was observed in the Il18+/+ mice. To reveal the mechanism, RNA-Seq was performed.

IL-18 mediates sickle cell cardiomyopathy and inducible ventricular arrhythmias

Project description:Rationale. Sickle cell cardiomyopathy is characterized by prolonged QTc, myocardial fibrosis, diastolic dysfunction, and vulnerability to ventricular tachycardia (VT). Based on previous reports, IL-18 mediates cardiac fibrosis and its promoter SNPs are associated with sudden cardiac death in a non-sickle population. We, therefore, hypothesized that IL-18 may mediate cardiomyopathy and VT in sickle cell disease. Findings. Sickle cell patients with evidence of myocardial fibrosis demonstrated greater IL18 expression. Patients with higher IL18 gene expression levels also exhibited increased QTc intervals and overall mortality. A novel SNP within IL-18, rs5744285, was associated with both QTc and IL-18 expression levels. Similar to sickle cell patients, sickle mice demonstrated increased cardiac fibrosis and prolonged action potential duration (APD) associated with higher VT inducibility. Administration of exogenous IL-18 acutely ex vivo to hearts resulted in increased triggered activity and VTs while inhibition of IL-18 resulted in reduced cardiac NFkB expression, fibrosis, and dysfunction in sickle mice. Conclusions. IL-18 is associated with prolonged QTc, myocardial fibrosis, and mortality in sickle cell patients, and prolonged APD associated with heightened VT susceptibility in sickle mice. Inhibition of IL-18 improves cardiac function, in part, via NFκB. Inflammatory cytokines contribute to the development of sickle cardiomyopathy and inducible VT.

Interleukin-18 and cytotoxic impairment are independent and synergistic causes virus-induced hyperinflammation in mice

Project description:To determine the effects of excess IL-18 on virus-induced immunopathology, we infected Il18-transgenic (Il18tg) mice with Lymphocytic Choriomeningitis Virus (LCMV, Armstrong). LCMV infection is self-limited in wild-type (WT) mice, but Prf1-/- mice develop prolonged viremia and fatal HLH. LCMV-infected Il18-transgenic (Il18tg) mice developed cachexia and hyperinflammation comparable to Prf1-/- mice, albeit with minimal mortality. We use RNA-sequencing to identify differences in the transcriptome of GP33 - specific CD8 T cells from these mice in comparison to a wild type mouse under the same conditions. We saw expression of Ifng was elevated about 4-fold in both Prf1-/- and Il18tg mice. Genes specifically upregulated in Il18tg CD8 T-cells included Csf2, Il1rl1, and Il12rb2. We also saw differences between Prf1-/- and WT/Il18tg CD8 T-cells that included Klrg1 and transcripts associated with CD8 T-cell exhaustion, including the inhibitory receptors Lag3, Havcr2, and Tigit, and the transcription factors Tox and Prdm1. These data support that IL-18 as a potent, independent, and modifiable driver of life-threatening innate and adaptive hyperinflammation, and support the rationale for an IL-18-driven subclass of hyperinflammation.

Intestinal IL-17R signaling controls liver inflammation by constraining microbiome-induced TLR9 signaling and IL-18 production

Project description:IL-17 and IL-17R signaling in the intestinal epithelium regulate the intestinal microbiome. Given the reported links between intestinal dysbiosis, bacterial translocation, and liver disease, we hypothesized that intestinal IL-17R signaling plays a critical role in mitigating hepatic inflammation. To test this, we studied intestinal epithelial-specific IL-17RA deficient mice in a model of concanavalin A hepatitis. Absence of enteric IL-17RA signaling exacerbated hepatitis and hepatocyte cell death. These mice exhibited commensal dysbiosis, increased intestinal and liver Il18, and increased liver translocation of bacterial products, specifically CpG DNA. Mechanistically, CpG DNA induced hepatic IL-18, increasing IFNγ and FasL in hepatic T-cells to drive inflammation. Thus, intestinal IL-17R regulates translocation of TLR9 ligands and constrains susceptibility to hepatitis. These data connect enteric Th17 signaling and the microbiome in hepatitis, with broader implications on the effects of impaired intestinal immunity and subsequent release of microbial products seen in other extra-intestinal pathologies.

Faecalibacterium prausnitzii promotes intestinal epithelial IL-18 production through activation of the HIF1A pathway

Project description:In this study, we investigated the regulation of interleukin-18 (IL-18) expression, crucial for intestinal barrier integrity, in inflammatory bowel disease (IBD). Through experiments with Caco-2 cells, we demonstrated that HIF1α-mediated IL18 expression induced by F. prausnitzii was dependent on HIF1α, suggesting a potential role for butyrate-producing gut bacteria in promoting mucosal healing in IBD.

A fusion protein complex that combines IL12, IL15, and IL18 signaling to induce memory-like NK cells for cancer immunotherapy

Project description:NK cells are a promising cellular therapy for cancer, with challenges in the field including persistence, functional activity, and tumor recognition. Briefly priming blood NK cells with rhIL-12, rhIL-15, and rhIL-18 (12/15/18) results in memory-like NK cell differentiation and enhanced responses against cancer. We developed a novel platform centered upon an inert tissue factor scaffold for production of heteromeric fusion protein complexes (HFPC). The first use of this platform combined IL-12, IL-15 and IL-18 receptor engagement (HCW9201) and the second adds CD16 engagement (HCW9207). HCW9201 and HCW9207 stimulated activation and proliferation signals in NK cells, but HCW9207 had decreased IL-18 receptor signaling. RNAseq and multidimensional mass cytometry revealed strong parallels between HCW9201 and 12/15/18. Moreover, HCW9201 stimulation improved NK cell metabolic fitness, and resulted in the DNA methylation remodeling characteristic of memory-like differentiation. HCW9201- and 12/15/18-primed similar increases in short-term and memory-like NK cell cytotoxicity and IFN-g production against leukemia targets, as well as equivalent control of leukemia in NSG mice. Thus, HFPCs represent a protein engineering approach that solves many problems associated with multi-signal receptor engagement on immune cells, and HCW9201-primed NK cells will be advanced as an ideal approach for clinical GMP-grade memory-like NK cell production for cancer therapy.erm and memory-like NK cell cytotoxicity and IFN-g production against leukemia targets, as well as equivalent control of leukemia in NSG mice. wdr13 knockout mice develops MD like phenotype on chronic stress

Project description:WDR13 expresses from the X chromosome and has a highly conserved coding sequence. There have been multiple associations of WDR13 with memory and psychiatric disorders. However, its detailed function in context of brain and behavior remains unknown. We characterized the behavioral phenotype of male mice lacking the homologue of WDR13 gene (Wdr13-/0). Mice were evaluated for changes in anxiety, learning and memory, and chronic stress induced behavioral deficits. Taking cue from analysis of its expression in the brain, we chose hippocampus and pre-frontal cortex for molecular studies to delineate its function. In the absence of Wdr13, there was a significant upregulation of synaptic proteins, viz., SYN1, RAB3A, CAMK2A etc accompanied with better memory retention in mice. Interestingly, three weeks social isolation stress resulted in downregulation of aforementioned synaptic proteins, decreased exploration time in central area of Open field test, increased immobility in Forced swim test, and reduced dendritic arborations of hippocampal CA1 neurons- hallmarks of Major Depressive Disorder (MDD) in Wdr13-/0 mice. This phenotype could be rescued by short imipramine treatment. We hypothesise that one of the key factors behind the MDD like phenotype in these mice was upregulation of transcription factor GATA1. Parallel work from our lab has indicated WDR13 to interact with multiple nuclear receptors and acting majorly as a co-repressor. Absence of Wdr13 resulted in de-regulated expression of a number of genes including multiple synaptic genes leading to observed phenotypes. Summarily, our data provides functional evidence for the role of Wdr13 in memory retention and in withstanding chronic stress induced depression.
