The role and function of YAP in cancer-associated fibroblasts

ABSTRACT: To investigate how YAP activity contributes to the functional properties of cancer-associated fibroblasts (CAFs), we established YAP-deficient cancer-associated fibroblast cell line, in which YAP expression can be inducibly silenced using shRNA, for RNA-Seq compared to wildtype CAFs.

We analyzed 5 untreated and 5-aza-dC-treated pancreatic cancer associated fibroblast cultures using the Affymetrix Human Exon 1.0 ST platform. Gene expression levels were compared using Partek (version 6.3beta).

Project description:Cancers adapt proteostasis to cope with the burden of misfolded proteins, stabilize key signalling nodes and sustain their malignant behaviour. Tumour stroma is subjected to similar stresses, but how they influence its aberrant status remains unclear. We show that tumour stroma presents consistent upregulation of target genes associated to the major misfolding regulator HSP90 in cancer-associated fibroblasts (CAFs), and that HSP90α is required for CAFs to remodel the extracellular matrix (ECM) and promote cancer cell motility and growth. Mechanistically, HSP90α sustains TGFβ responses and YAP protein levels required for CAF functionality. In vivo, stromal or fibroblast-specific loss of HSP90α results in reduced ECM deposition, angiogenesis, growth and dissemination of breast tumours. Clinical analyses reveal a correlation between HSP90-dependent programs and YAP activity in CAFs, that are also associated with poor patient prognosis. Our findings uncover a link between proteostasis, mechanotransduction and generation of aggressive tumour microenvironments through HSP90α.

Expression analysis of YAP and Angiomotin target genes

Project description:To assess changes in gene expression regulated by Yap1 or Angiomotin, either Yap1 or Angiomotin were knocked down in HEK293 cells using shRNA and expression was assessed by microarray analysis and compared to expression patterns of wildtype cells. Microarray analysis was performed to compare gene expression in Amot-KD, Yap-KD, and control HEK293 cells. Samples from three individual experiments for each of the three groups were analyzed.

shRNA profiling of human normal breast and breast cancer cell lines to identify genes regulated by YAP

Project description:Frequent loss of heterozygosity (LOH) at 11q22-23 in breast cancer strongly suggests that this region contains a tumor suppressor gene, yet to be identified. We and others have shown Yes-associated protein (YAP), which is located at 11q22.2, transactivates the p53 family member p73 upon DNA damage, suggesting a tumor suppressive function for YAP. Our analysis of breast tumor tissues by immunohistochemistry (IHC) showed loss of YAP protein expression in great portion of breast cancers. We used microarray to look at the targte genes regulated by YAP in normal breast luminal cell and breast cancer cell lines. Experiment Overall Design: We generated stable cell lines by introducing control vector(pRS-IRES-GFP/pmig) or YAP shRNA(pRS-IRES-GFP-YAP/pmig-YAP) in a normal breast luminal cell line 1089 luminal and breast cancer cell lines MDA MB231. RAN was extracted and hybridized on Affymetrix microarrays.We looked for new target genes regulated by YAP in normal breast luminal cell and breast cancer cell lines.

Twist1 knockdown in two gastric cancer-associated fibroblast lines

Project description:Primary human gastric cancer-associated fibroblast (CAF) cultures (CAF14 and CAF32) were establised from 2 gastrectomy specimens. Silencing the expression (loss-of-function effect) of Twist1 in CAFs showed candidate target genes regulated by Twist1 and abrogated their tumor-promoting properties. Experiment design: two gastric CAFs (CAF14 adn CAF32, respectively) were establised from two gastric cancer patients and Twist1 knockdown studies were performed using shRNA.
