Single-cell transcriptomic profiling of pituitary adenoma from patient with Cushing's disease

Project description:We performed single-cell transcriptomic profiling of a pituitary adenoma from a patient with a rare subtype of Cushing's disease using the 10x Genomics platform. The analysis revealed the cellular landscape of the tumor, identifying distinct cell populations, their transcriptional signatures, and the degree of intratumoral heterogeneity. These findings provide insights into the cellular architecture of this uncommon ACTH-secreting pituitary adenoma variant.

The Epigenomic Landscape of Pituitary Adenomas Reveals Specific Alterations and Differentiates Among Acromegaly, Cushing's Disease and Endocrine-Inactive Subtypes

Project description:Recurrent single-nucleotide and small indel somatic mutations were infrequent among the three adenoma subtypes. However, somatic copy-number alterations (SCNA) were identified in all three pituitary adenoma subtypes. Methylation analysis revealed adenoma subtype-specific DNA methylation profiles, with GHsecreting adenomas being dominated by hypomethylated sites. Likewise, gene-expression patterns revealed adenoma subtype-specific profiles. Integrating DNA methylation and gene-expression data revealed that hypomethylation of promoter regions are related with increased expression of GH1 and SSTR5 genes in GH-secreting adenomas and POMC gene in ACTH secreting adenomas. Finally, multispectral IHC staining of immune-related proteins showed abundant expression of PD-L1 among all three adenoma subtypes.

Understanding early AutoAb signatures in Pituitary Adenomas using CDI HuProt™ (Human Proteome Microarray)

Project description:CDI HuProt™ (Human Proteome Microarray), was used to study Pituitary Adenomas PAs (Acromegaly, Cushing's and NFPA) using serum samples, from around 14 individuals (4 Healthy control, 4 Acromegaly, 3 Cushing’s and 3 NFPAs patient samples) were used study their autoantibody profiles. Patient serum samples in dilution 1:500 ratio were used for primary incubation. Secondary incubation was performed with anti-human IgG conjugated with Cy5 (Jackson Immuno Research, catalogue number 109-175-064) in 1:5000 dilutions was used. This CDI HuProt™ array was scanned with GenePix 4000B Microarray Scanner (Molecular Devices), with a PMT gain of 500, Scan Power 100 and Laser power of 1.31.

The Cushing's disease adipose gene expression profile reveals effects of long term glucocorticoids on adipose tissue lipid, protein and glucose metabolism

Project description:Glucocorticoids have major effects on adipose tissue metabolism. To study tissue mRNA expression changes induced by chronic elevated endogenous glucocorticoids, we performed RNA sequencing on subcutaneous adipose tissue from patients with Cushing’s disease (n=5) compared to patients with non-functioning pituitary adenomas (n=11). We found higher expression of transcripts involved in several metabolic pathways, including lipogenesis, proteolysis and glucose oxidation as well as decreased expression of transcripts involved in inflammation and protein synthesis. To further study this in a model system, we subjected mice to dexamethasone treatment for 12 weeks and analyzed their inguinal (subcutaneous) fat pads, which led to similar findings. Additionally, mice treated with dexamethasone showed drastic decreases in lean body mass as well as increased fat mass, further supporting the human transcriptomic data. These data provide insight to transcriptional changes that may be responsible for the co-morbidities associated with chronic elevations of glucocorticoids DESIGN: Patients with cushing's (n=5) or non-functioning pituitary adenoma (n=11) were prospectively observed from March 2011 to June 2012.
