ABSTRACT: We revealed that tumor cells in the bleomycin-injured lungs (BLM lungs) are differently influenced by their unique lung fibrogenic microenvironment compared to those in subcutaneous tumor models in vivo or in 2-dimensional monolayer culture in vitro.

RNA-seq analysis of tumor-infiltrating effector T cells isolated from subcutaneous mouse Lewis lung carcinoma (LLC) tumors

Project description:The goal of this study was to analyze differences in gene expression between tumor-infiltrating effector T cells with higher or lower uptake of the 2-deoxyglucose analog 2-NBDG in vivo. Mice bearing subcutaneous LLC tumors were injected intravenously with the 2-deoxyglucose analog 2-(N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl) amino)-2-deoxyglucose (2-NBDG, Life Technologies, catalog: N13195). 2-NBDG was used at 25 mg/kg of mouse body weight; 3.33 mg/ml in PBS. 2-NBDG was allowed to circulate for 15 min before euthanizing the mice and excising the tumors to isolate effector (CD44+, CD62L-negative T cells) TILs. The 15% of effector CD4 or CD8 TILs with the highest 2-NBDG staining were sorted as 2-NBDG Hi cells, and the lowest 15% were sorted as 2-NBDG Lo. 4 independent mice were used. For each population, a pool of 2000 cells (500 cells per mouse) was done. RNA of the respective pools was extracted with the RNeasy Microkit (Qiagen, Catalog: 74004), and cDNA libraries were prepared from poly A RNA (mRNA) using the Smart-Seq2 single-cell protocol (Picelli, et al., 2014, Nat. Protoc. 9, 171–181) with some modifications.

Project description:Lung epithelial cell injury contributes to various lung diseases, impacting lung function and overall human health. Bleomycin (BLM)-induced murine models of pulmonary fibrosis (PF) are commonly utilized to study lung epithelial cell injury. However, the complex microenvironments in inflamed lungs hinder clear causal relationships between cell-to-cell, molecules interaction. To address this limitation, we aimed to establish an ex vivo organoid model of BLM-induced lung injury recapitulating the cellular and molecular responses of BLM-induced lung epithelial injury in vivo.

Expression data from the subclones of Lewis Lung Carcinoma (LLC) cells

Project description:Medium conditioned by LLC cells stimulates thermogenic gene expression when added onto primary adipocytes. We generated single cell colonies from parental LLC cells. Media conditioned by the subclones stimulated thermogenic gene expression in primary adipocytes at varying degrees. Subclones 2, 3, 6 and 28 produce significantly larger amount of thermogenic activity than the subclones 18, 19, 23 and 24. We compared gene expression profiles of these subclones to identify secreted factors enriched in the more thermogenic clones.

Characterization of tumor microenvironment using single cell RNA sequencing in Lewis Lung Carcinoma cell transplantation mouse model

Project description:The tumor microenvironment holds significant importance in cancer development and progression. Research conducted using mouse models of tumor cell transplantation can enhance our comprehension of the tissue microenvironment that allows tumor growth at an early stage. Among these models, those featuring the Lewis Lung Carcinoma (LLC) cell line are noteworthy due to their low immunogenic features. Since numerous cancer patients do not respond effectively to current immunotherapy, it is imperative to investigate models with low immunogenic features. Consequently, this study employed single-cell RNA sequencing on an orthotopic lung cancer mouse model utilizing the LLC cell line to profile the characteristics of diverse cell types that emerge in the lung during tumor engraftment. The lung microenvironment exhibited discernible transformations, including fibrogenic gene expression, increased cell-to-cell and cell-to-extracellular matrix adhesion, and reduced infiltration of T cells compared to lungs without tumor engraftment. In conclusion, these findings reveal cellular and molecular alterations that can be targeted for tumor prevention and treatment for low-immunogenic tumors.

TDAG8-overexpressing or vector-transfected control LLC cells (5 × e5 in 200 µL of HBSS) were injected into the lateral tail veins of 6–8-week-old C57BL/6 mice. Lungs were removed on day 19 post-injection, and total RNA was extracted.

Tff1-expressing Tregs in lung prevents exacerbation of Bleomycin-induced pulmonary fibrosis (RNA-Seq)

Project description:Bleomycin (BLM) induces lung injury, leading to inflammation and pulmonary fibrosis. Regulatory T cells (Tregs) maintain self-tolerance and control host immune responses. However, little is known about their involvement in the pathology of pulmonary fibrosis. Here we show that a unique Treg subset that expresses trefoil factor family 1(Tff1) emerges in the BLM-injured lung. These Tff1-expressing Tregs (Tff1-Tregs) were induced by IL-33. Moreover, although Tff1 ablation in Tregs had no impact, selective ablation of Tff1-Tregs using an intersectional genetic method promoted pro-inflammatory features of macrophages in the injured lung and exacerbated the fibrosis. Taken together, our study revealed the presence of a unique Treg subset expressing Tff1 in BLM-injured lungs and their critical role in the injured lung to ameliorate fibrosis.
