Project description:We have established functions of the stimulus dependent MAPKs, ERK1/2 and ERK5 in DRG, motor neuron, and Schwann cell development. Surprisingly, many aspects of early DRG and motor neuron development were found to be ERK1/2 independent and Erk5 deletion had no obvious effect on embryonic PNS. In contrast, Erk1/2 deletion in developing neural crest resulted in peripheral nerves that were devoid of Schwann cell progenitors, and deletion of Erk1/2 in Schwann cell precursors caused disrupted differentiation and marked hypomyelination of axons. The Schwann cell phenotypes are similar to those reported in neuregulin-1 and ErbB mutant mice and neuregulin effects could not be elicited in glial precursors lacking Erk1/2. ERK/MAPK regulation of myelination was specific to Schwann cells, as deletion in oligodendrocyte precursors did not impair myelin formation, but reduced precursor proliferation. Our data suggest a tight linkage between developmental functions of ERK/MAPK signaling and biological actions of specific RTK-activating factors. Abstract:
The ErbB signaling pathways, which regulate diverse physiological responses such as cell survival, proliferation and motility, have been subjected to extensive molecular analysis. Nonetheless, it remains poorly understood how different ligands induce different responses and how this is affected by oncogenic mutations. To quantify signal flow through ErbB-activated pathways we have constructed, trained and analyzed a mass action model of immediate-early signaling involving ErbB1-4 receptors (EGFR, HER2/Neu2, ErbB3 and ErbB4), and the MAPK and PI3K/Akt cascades. We find that parameter sensitivity is strongly dependent on the feature (e.g. ERK or Akt activation) or condition (e.g. EGF or heregulin stimulation) under examination and that this context dependence is informative with respect to mechanisms of signal propagation. Modeling predicts log-linear amplification so that significant ERK and Akt activation is observed at ligand concentrations far below the K(d) for receptor binding. However, MAPK and Akt modules isolated from the ErbB model continue to exhibit switch-like responses. Thus, key system-wide features of ErbB signaling arise from nonlinear interaction among signaling elements, the properties of which appear quite different in context and in isolation.

It was slightly changed to make it valid SBML and to incorporate the step functions, described in the readme file and needed for inhibitor preincubation. the equilibration processes end at 1800 sec, so to reproduce the dynamics shown in the publication and supplemental material, only the time points after 1800 need to be considered. The parameter set is the hand fitted one used for Sfigure 3 in the supplemental materials. All species are in molecules, apart from HRG, EGF and Inh, which are in M.
The results shown in SFigure 3 can be calculated dividing the parameters ERK_PP
, AKT_PP
and ERB_B1_P_tot
by ERK_t
, AKT_t
and EGFR_t
, respectively. Somehow we did not find the right scaleing factor for the phosphorylated ErbB1 receptor. Therefore the model does only qualitatively reproduces the timecourses shown in the first row of Sfigure 3.

Project description:miR-16 is a potent tumor suppressor in ErbB-2 positive breast cancer. The primary objetive of this study was to identify previously uncharacterized putative mRNA targets of miR-16 in ErbB-2 postitive breast cancer cells. To achieve our objective we studied the effects of exogenously expressed miR-16 on the gene expression profile of primary cultures of ErbB-2 positive murine breast cancer tumors. The C4HD tumor line displays high levels of estrogen receptor and progesterone receptor, overexpresses ErbB-2 and ErbB-3, exhibits low ErbB-4 levels, and lacks EGF-R expression.

MEL-18, a polycomb group protein and a member of the polycomb repressive complex 1 (PRC1), have suggested as a tumor suppressor in several cancer, including breast cancer. The results provides that the depletion of MEL-18 in HER2-positive breast cancer causes the activation of ErbB signaling pathway. We proposed that MEL-18 is a novel prognostic and therapeutic marker for HER2-positive breast cancer.
