Molecular and anatomical roadmap of stroke pathology in immunodeficient mice

ABSTRACT: Here, we assessed the stroke pathology of popular genetic immunodeficient mouse models using NOD scid gamma (NSG) and recombination activating gene 2 (Rag2–/–) mice as well as pharmacologically immunosuppressed mice and compared them to immune competent, wildtype (WT) C57BL/6J mice three weeks after injury.

Human IL-6 fosters long-term engraftment of human patient derived myeloma cells in immunodeficient mice

Project description:Multiple myeloma is a largely incurable and life-threatening malignancy of antibody-secreting plasma cells. An effective and widely available animal model that recapitulates human myeloma and related plasma cell disorders is lacking. We show that busulfan-conditioned hIL-6 transgenic NSG mice (NSG+hIL6) reliably support the engraftment of malignant and pre-malignant human plasma cells including from patients diagnosed with pre- and post-relapse myeloma, plasma cell leukemia, and AL amyloidosis. Consistent with human disease, NSG+hIL6 mice engrafted with patient-derived myeloma cells developed blood M spikes, and a majority developed anemia, hypercalcemia, and/or bone lesions. Singe cell RNAseq analyses showed non-malignant and malignant cell engraftment, the latter expressing a wide array of mRNAs associated with myeloma cell survival and proliferation. Myeloma-engrafted mice given CAR T-cells targeting plasma cells or bortezomib experienced reduced tumor burden. Our results establish that NSG+hIL6 mice provide an effective patient derived xenograft model for study and preclinincal drug development for myeloma and related plasma cell disorders.

Ageing signatures in NSG mice

Project description:Purpose: Understanding the role of an active immunesystem in ageing. Methods: Male NSG (NOD scid gamma) immunodeficient mice were maintained in normal housing condition. Islet cells were isolated at different age points and mRNA were extracted before shipping to Qiagen for analysis. Results: In NSG mice multiple of the metabolic pathways that are observed to be deregulated in ageing immunocompotent mice are infered to be sustained.

Preclinical study with a CD40 agonist on patient-derived xenograft mice for the treatment of B-cell acute lymphoblastic leukemia

Project description:To induce leukemia in NOD/SCID/γc−/− (NSG) mice, we injected 10^5 human B-ALL cells, into the tail vein of non-irradiated mice. Two different PDX mice models were studied (PDX-1; #1 and PDX-2; #2). The InVivoMAb anti-human CD40 antibody (BE0189, clone G28.5, BioXCell) was i.v. administrated (CD40) at the dose of 1 mg/kg, the same was done with the recommended isotype (Ig) (BE0083, MOPC-21, BioXCell). We administrated antibodies, at days 15 and 25. At day 35, when mice developed leukemia, B-ALL cells were purified from the BM of PDX-mice, with magnetic beads-conjugated to human CD45 antibodies. Three mice per condition were tested (replicat 1 to 3; mice m1 to m3).

Project description:Surrogate models have so far been missing that might allow the evaluation of graft-versus-leukemia effects in an individualized donor-patient-specific context. We here describe the generation of a NOD/SCID/IL2Rγnull (NSG) leukemia mouse model that ideally resembles the biology of miscellaneous patient-specific leukemias. Following transfer of patient-derived blasts into NSG mice, various forms of pediatric haematological malignancies readily engrafted NSG recipients and NSG-derived leukemic cells indeed resembled the individual leukemia with respect to phenotype, chromosomal aberrations, transcriptome and MRD marker expression.

Impact of dietary serine and glycine availability on plasma and tissue metabolite composition in mice

Project description:<p>This study examines how altering systemic serine and glycine availability through dietary intervention affects breast cancer metastasis in vivo. Female NOD-SCID-gamma (NSG) mice were maintained on either a control amino acid–replete diet or a serine/glycine-enriched diet for two weeks before and during metastatic progression of human breast cancer cells. By integrating dietary manipulation with in vivo metastasis assays, this study tests whether changes in circulating and tissue nutrient levels can influence metastatic colonization and tumor growth across organs. These data provide insight into how nutrient availability shapes the metabolic environment and metastatic potential of cancer cells.</p>
