Effect of hypoxic stress on Estrogen receptor alpha binding to chromatin

ABSTRACT: This project aimed at assessing the influence of hypoxic stress on Estrogen receptor alpha genome wide binding to chromatin, in presence or absence of E2 ligand. I also aimed at assessing if a 1-month hypoxic stress followed by 1 month of normoxia imprints an epigenetic memory, therefore altering genome wide ER binding (rescue condition).

Project description:To identify proteins involved in the regulation of adipogenic differentiation under hypoxic conditions, an RT2 Profiler PCR array was used to screen a panel of 84 genes associated with human adipogenesis in Bone-marrow Mesenchymal stem cells under normal and hypoxic conditions. Bone-marrow Mesenchymal stem cells were put in hypoxic chamber, set at an oxygen concentration of 0.2%. Cells under normoxia are considered as a control.

Mitochondria EglN1 drivers breast cancer progression by controlling metabolic adatation to hypoxic stress

Project description:Hypoxia is a hallmark of solid tumors. Mitochondria play essential roles in cellular adaptation to hypoxia, but the underlying mechanisms are not fully understood. Through mitochondrial proteomic profiling, we find that the prolyl hydroxylase EglN1 accumulates on mitochondria under hypoxia. EglN1 substrate binding region 23 loop is responsible for its mitochondrial translocation and contributes to tumor growth. Furthermore, we identify AMPK as an EglN1 substrate on mitochondria. The EglN1-AMPK interaction is essential for their mutual mitochondrial translocation. EglN1 prolyl-hydroxylates AMPK under normoxia, then they rapidly dissociate following prolyl-hydroxylation, leading to their immediate release from mitochondria. While hypoxia results in constant EglN1-AMPK interaction and accumulation on mitochondria, leading to the formation of CaMKK2-EglN1-AMPK complex to activate AMPK phosphorylation, consequently ensuring metabolic homeostasis and tumor growth. Our findings demonstrate EglN1 as an oxygen-sensitive metabolic checkpoint signaling hypoxic stress to mitochondria through its 23 loop, revealing a therapeutic target for solid tumors.

A multielectrode array (MEA) system was used as an in vitro method of recording real-time cardiac electrophysiological activity, highlighting the biological effects facilitated by exosomes. Additionally, miRNA-sequencing was performed to compare miRNA expressions in exosome-preconditioned and non-preconditioned IPSC-cardiomyocytes, elucidating important modulators of cardiac electrophysiology.

In vitro and in vivo analysis of hypoxic gene expression in rat gliomas

Project description:In vivo profiling of hypoxic gene expression in gliomas using the hypoxia marker EF5 and laser-capture microdissection We have employed the hypoxia marker EF5 coupled with laser capture microdissection to isolate RNA from viable hypoxic and normoxic regions of 9L experimental gliomas. We report the identification of the hypoxic gene expression profile in a solid tumor using a prototypical hypoxia marker EF5, Laser Capture Microdissection (LCM) and Microarray Gene Expression Profiling (EF5-LCM-MGEP). We have utilized LCM to isolate total RNA from normoxic and hypoxic regions in the rat 9L glioma tumor model grown in its isogenic host (Fischer rat). Microarray analysis of the isolated RNA generated an in vivo hypoxia expression profile. For comparison, we also analyzed RNA samples isolated from rat 9L glioma cells in culture exposed to hypoxia or normoxia.

LncRNAs and mRNAs profile suggested CCL / CXCL were involved in the reoxygenation reversal of hypoxic pulmonary hypertension in mice

Project description:The potential roles of mRNAs and lncRNAs in the development and the reoxygenation treatment of reversal of hypoxic pulmonary hypertension are still unknown. 24 healthy adult male C57/BL6 mice were divided into three groups (normoxia group, hypoxia group, hypoxia-normoxia group), lncRNA and mRNA microarray was performed，meanwhile，the changes of right ven-tricular systolic pressure (RVSP) and other physiological indexes were measured in the three groups. The results showed that the index of RVSP, the right ventricular hypertrophy index RV/(LV+S), pulmonary artery walls and the muscularization ratio of pulmonary arterioles were significantly increased after Hypoxia treatment, while they were attenuated after the normoxia condition restored. Moreover, the study revealed there were 62 lncRNAs and 99 mRNAs with significant difference expression were associated with the reversal of hypoxic pulmonary hypertension by reoxygenation in mice. Furthermore, the pathway enrichment and gene ontology analysis of 99 DEGS were performed. Interestingly, the result suggested that chemokine subfamily CCL2/ CXCL played a role in the occurrence, development and reversion of pulmonary hypertension by reoxygenation. This study was the first time to demonstrate the transcriptome profiles of lncRNAs and mRNAs in the lung tissue during the reversal of hypoxic pulmonary hypertension in mice by reoxygenation, which might further broaden the understanding of the pathogenesis and the therapeutic effect of oxygen recovery of hypoxic pulmonary hypertension.

Identification of hypoxia-regulated genes in the liver of common sole (Solea solea) fed different dietary lipid contents

Project description:Hepatic transcriptome of sole fed low and high lipid dietary content in the early response to hypoxic stress. Characterization of the hepatic transcriptome of S. solea in order to assess the molecular mechanisms underlying the effects of dietary lipid content on hypoxia tolerance in common sole. S. solea individuals (8 month post-hatching) were fed for two month with commercial diets containing either 11% lipids or 20% lipids. Before hypoxia challenge stress, 38 individuals (19 from each dietary group) were randomly pooled in a single tank (1 m3) and left undisturbed and unfed for 48 h. The hypoxia challenge consisted then in a decrease of water oxygenation from 100 % to 10 % within one hour, followed by a slower descent at 2 % hr-1 to 4%. As fish lost their equilibrium, they were removed and sampled. Thirty six individuals (18 from each dietary group) were also sampled in normoxic conditions, resulting in 4 experimental groups: Normoxia low-lipid, Normoxia high-lipid, Hypoxia low-lipid, Hypoxia high-lipid. The experimental design was therefore a 2-way full factorial design between two juvenile dietary lipid contents and two oxygen concentration conditions.
