Gene expression in wild type and Cagal102 knockout Candida albicans strains.

ABSTRACT: CaGAL102 is a sequence homolog of Rmlb. In Candida knock out of this gene causes abnormal hyphal morphogenesis and increased sensitivity to cell wall damaging agents. The knock out strain is also avirulent in mouse model of systemic infection. To get a larger insight into the function of the protein product of this gene we carried out global transcription analysis through micro array experiment. The gene is expressed under normal growth conditions and the knock out causes the cells to become hyphal under these conditions. The RNA was checked using Bioanalyser (Agilent). Single dye experiment was carried out using Cy3 labelled pooled wild type samples and two independent mutant RNA samples.

Partial de-repression of the hyphal program does not restore hyphae formation in absence of a functional Arp2/3 complex

Project description:Deleting components of the Arp2/3 complex in Candida albicans resulted in a global lack of hyphal specific gene induction. This observation suggests that the failure in hyphal growth of Arp2/3 complex mutants could be a result of failure to activate hyphal specific genes. If the hyphal defect was primarily due to failure to activate gene expression, de-repressing hyphal-specific gene expression by deleting the NRG1 repressor could potentially suppress the defect, as deletion of NRG1 leads to constitutive filamentous growth even in the absence of any hyphal induction signals (Garcia-Sanchez et al., 2005, Kadosh & Johnson, 2005). We therefore created an nrg1Δ/Δarp2Δ/Δ mutant. When grown under non-inducing conditions, nrg1Δ/Δarp2Δ/Δ cells showed the arp2Δ/Δ mutant morphology of round and swollen cells. When induced for hyphal growth, nrg1Δ/Δarp2Δ/Δ cells also exhibited the arp2Δ/Δ cell morphology and did not form hyphae even after extended overnight incubation times.

Immunoproteomic analysis of antibody responses to extracellular proteins of Candida albicans in serum of candidemia patients revealed glycosylation effects on protein antigen recognition

Project description:During infection the yeast Candida albicans undergoes a yeast-to-hyphal transition and secretes numerous proteins required for the invasion and damage of host tissues and the modulation of the host immune responses. Still little is known about the interplay of Candida albicans secreted proteins and the host adaptive immune system. For the characterization of the extracellular proteins of C. albicans and their ability to trigger a serological response in candidemia patients we applied 2D gel electrophoresis and LC-MS/MS-based approaches. These analyses identified the secretion of 101 different proteins from C. albicans yeast cells and 410 proteins from hyphal cells. Morphology-dependent changes of the secretome were mostly associated with proteins involved in cell wall organization, general stress response and the interplay with host immune cells. Our immunoproteomics workflow for the identification of secreted C. albicans protein antigens revealed a core set of 20 immunodominant specific anti- C. albicans protein antibodies. However, some secreted protein antigens showed cross-reactivity with sera from control groups without Candida infection. Glycoprotein staining of C. albicans hyphal secretome demonstrated that some secreted proteins are strongly glycosylated. Enzymatic deglycosylation of secreted proteins uncovered a significant contribution of glycan epitopes to the recognition of the secreted aspartyl protease Sap6 by IgG antibodies from patient sera.

RNA-Seq analysis of WT and BbWor1 disruption strain in beauveria bassiana

Project description:Wor1 is a conserved fungal regulatory protein that control the phenotypic switching and pathogenicity of Candida albicans.We simulated three models of Beauveria bassiana morphological transitions, including CTH (conidia to hyphae), HTC (hyphae to conidia) and BTB (blastospore to blastospore).We find that disruption of BbWor1 (an ortholog of Wor1) resulted in a distinct reduction in the time required for germination (CTH), a significant increase in hyphal growth and a decrease in the yield of conidia (HTC), indicating that BbWor1 positively controls conidia production and negatively regulates hyphal growth in conidia-hyphae switching. Meanwhile, the ΔBbWor1 prominently decreased blastospore yield, as well as shortening G0/G1 phase and prolonging G2/M phase, under the BTB model. Importantly, BbWor1 contributed to conidia-hyphae switching and blastospore propagation via different genetic pathways, and yeast one-hybrid testing demonstrated the necessity of BbWor1 to control the transcription of an allergen-like protein gene (BBA_02580), as well as a conidial wall protein gene (BBA_09998). Moreover, the dramatically weakened virulence of ΔBbWor1 was examined by immersion and injection methods.

Role of a Candida albicans Whi5/Nrm1 homolog in cell cycle and hyphal-specific gene expression

Project description:Candida albicans is an opportunistic pathogenic fungus that is able to assume several morphologies, including yeast and hyphal growth forms. The hyphal morphology can be induced by various environmental stimuli and is accompanied by expression of a large set of hyphal-specific genes (HSGs). Cell cycle and morphogenetic programs are interconnected: notably, inhibition of cell cycle progression often causes a switch to filamentous growth. Here we identify and characterize CaNrm1, a C. albicans homolog of the S. cerevisiae Whi5 and Nrm1 transcription inhibitors that, analogous to mammalian Rb, regulate the cell cycle transcription program in the G1 phase and at the G1/S transition. CaNRM1 is able to complement the phenotypes of both whi5 and nrm1 mutants in S. cerevisiae. Deletion of CaNRM1 causes a reduction in cell size and results in increased resistance to hydroxyurea (HU), an inhibitor of DNA replication; analysis of the expression of ribonucleotide reductase, the target of HU, suggests that its transcriptional induction in response to HU is mainly dependent upon CaNrm1. Genetic epistasis analysis suggests that CaNrm1 interacts with the SBF and MBF transcription factors in S. cerevisiae and with the MBF functional homolog in C. albicans. At the transcription level, deletion of CaNRM1 causes an induction of many G1 and G1/S-specific genes. Induction of the HSGs is dampened under certain conditions in the Canrm1-/- mutant, suggesting that the cell cycle transcription program influences the morphogenetic transcription program of C. albicans. One aspect of the characterization of the C. albicans Whi5/Nrm1 gene was to examine how this molecule influences global gene expression. To that end, we isolated total RNA from log-phase nrm1-/nrm1- (as well as parent/background strain) cells in two independent replicate experiments. These samples were processed, labeled, and subsequently used for hybridization of custom C. albicans Affymetrix arrays.
