Gene expression in NKR-P1B+ versus Ly49s3+ rat NK cells

ABSTRACT: Two major subsets of rat natural killer (NK) cells can be distinguished based on their expression of either the Ly49s3 or the NKR-P1B lectin-like receptor. Ly49s3+ NK cells, but not NKR-P1B+ NK cells, express a wide range of Ly49 receptors. Here, we performed a global gene expression profiling of sorted, single-positive NKR-P1B+ or Ly49s3+ splenic NK cells in order to get an overview of differences in expressed genes between the NKR-P1B+ and Ly49s3+ NK cell subsets.

Ly49s3+ NK cells, but not NKR-P1B+ NK cells, express a wide range of Ly49 receptors. Here, we performed a global gene expression profiling of sorted, single-positive NKR-P1B+ or Ly49s3+ splenic NK cells in order to get an overview of differences in expressed genes between the NKR-P1B+ and Ly49s3+ NK cell subsets. NKR-P1B or Ly49s3 single-positive NK cells were sorted by flow cytometry from NK-enriched mononuclear cells from rat spleens. 9 rats of the PVG.7b strain aged 8-12 weeks were used. Total RNA was harvested by Tri Reagent extraction, and subjected to microarray analysis (Affymetrix Rat Genome 230 2.0 Array).

Microarray analysis of resting Nkrp1g+ NK cells versus resting Nkrp1g- NK cells from spleens of female BALB/c mice.

Project description:Natural Killer Gene Complex (NKC)-encoded immunomodulatory C-type lectin-like receptors (CTLR) include members of the NKRP1 and CLEC2 gene families, which constitute genetically linked receptor-ligand pairs and are thought to allow for NK cell-mediated immunosurveillance of stressed or infected tissues. The mouse CTLR Nkrp1g was previously shown to form several receptor-ligands pairs with the CLEC2 proteins Clr-d, Clr-f, and Clr-g, respectively. Recently, we demonstrated a gut-restricted expression of Clr-f on intestinal epithelial cells that is spatially matched by Nkrp1g on subsets of intraepithelial lymphocytes (Leibelt et al., 2015). We now investigated expression and ligand interaction of Nkrp1g in the splenic compartment, and found an exclusive expression of Nkrp1g on a small subset of NK cells that upregulates Nkrp1g after cytokine exposure. To further characterize NKrp1g+ NK cells, we performed microarray analysis of resting Nkrp1g+ versus Nkrp1g- splenic NK cells that were purified by FACsorting. Total RNA isolation was followed by an Affymetrix microarray analysis.

Transcription profiling of mouse splenic dendritic cell subpopulations to characterize molecular differences among the cells.

Project description:To characterize differences between BALB/c splenic CD11cintB220+Gr1+ PDCs (plasmacytoid dendritic cells), CD11cintB220+CD49b+ IKDCs (interferon producing killer-dendritic cells), and CD11chighB220- cDCs (conventional dendritic cells), we performed gene expression profile analysis using Affymetrix chips. We FACS-sorted BALB/c spleen DC subpopulations. Comparison of differentially expressed genes between IKDCs and cDCs vividly revealed selective expression of multiple NK-related genes in IKDCs . These included granzymes A, B, K and M, perforin, Fas ligand, and NK receptors such as NKG2A, NKG2D, Ly49 family genes, NKR-P1, NKG7, NKp46 and Mafa (KLRG1). No NK-related genes were highly expressed in the PDCs. Experiment Overall Design: We prepared two biological samples separately for each DC population, and analyzed the expression profiles by comparing to those of cDCs (control sample).

Differential requirement for Nfil3 during NK cell development

Project description:Natural killer (NK) cells can be grouped into distinct subsets that are localized to different organs and exhibit different capacity to secrete cytokines and mediate cytotoxicity. Despite these hallmarks that reflect tissue-specific specialization in NK cells, little is known about the factors that control the development of these distinct subsets. The basic leucine zipper transcription factor nuclear factor interleukin 3 (Nfil3; E4bp4) is essential for bone marrow-derived NK cell development but it is not clear whether Nfil3 is equally important for all NK cell subsets nor how it induces NK lineage commitment. Here we show that Nfil3 is required for the formation of Eomesodermin (Eomes)-expressing NK cells, including conventional medullary and thymic NK cells, whereas TRAIL+ Eomes- NK cells develop independent of Nfil3. Loss of Nfil3 during the development of bone marrow-derived NK cells resulted in reduced expression of Eomes and, conversely, restoration of Eomes expression in Nfil3-/- progenitors rescued NK cell development and maturation. Collectively, these findings demonstrate that Nfil3 drives the formation of mature NK cell by inducing Eomes expression and reveal the differential requirements of NK cell subsets for Nfil3. Genome-wide expression study of the responses of murine NK cells and dendritic cell subsets to MCMV infection

Project description:Murine Cytomegalovirus (MCMV) infection leads to the activation of various immune cells, including dendritic cells (DCs) and Natural Killer (NK) cells. This activation is partly driven by innate cytokines including IFN-I, which are induced early after infection. The objective was to address the role of different innate cytokines in shaping DC subsets and NK cell responses, in particular the role of cell intrinsic responses to IFN-I. In order to decipher the specific impact of cell-intrinsic IFN-I on cell subsets, we performed a genome-wide expression analysis on CD45.1 WT and CD45.2 IFNAR-/- splenic conventional DC (cDC) subsets and NK cells isolated from C57BL/6 [CD45.1 WT / CD45.2 IFNAR-KO] mixed bone marow chimera mice. This study includes data from cDC subsets (CD8a and CD11b) and NK cells purified by flow cytometry sorting from the spleen of bone marrow chimera (BMC) mice, under steady-state or MCMV condition. Two independent replicates were made for each cell type, from two independent pools of spleens from uninfected or d1.5 MCMV-infected BMC 5-8 mice, and were hybridized on 3 separate batches of gene chips.

PCa_Secretome_PC3vsPC3nKR - Secretome profiling of PC3/nKR cells, a novel highly migrating prostate cancer subline derived from PC3 cells

Project description:Prostate cancer (PCa) is the most common cancer in American men. The American Cancer Society’s estimates for prostate cancer in the United States for 2017 are estimated 161.360 new cases and 26,730 deaths from PCa. To study metastatic properties to bone, PC-3 cell line is mainly used classical human prostatic carcinoma cell line, established and characterized its tumorigenicity from a human prostatic adenocarcinoma metastatic to bone is reported. In addition, PC-3/nkR cell line, natural killer(NK) cells-resistant, was isolated from mammary tumor xenograft studies in mice from PC-3 was implanted to nude mice and fecund to be tumorigenic in the early 2000s. In this study, we investigated secreted proteins of the conditioned media of PC-3 and PC-3/nkR cell lines using comparative proteomics technology to identify the molecular mechanism related to metastatic processes related to PC-3/nkR. Our study showed PC-3/nkR cells are new highly migrated and NK cells-resistant cell-line compared to PC-3 cells, as novel highly malignant tumor cells to study mechanisms of PCa metastatic.
