Gene expression profiling of placental villi from intrauterine and tubal ectopic pregnancies

ABSTRACT: The tubal ectopic pregnancy (TEP) is a severe pregnancy complication accounted for 10-15% pregnancy-related deaths owing to the implantation and growth of embryos in fallopian tubes. Placental villi from TEP exhibit significant difference compared with that from intrauterine pregnancy (IP) in total volumes, trophoblast differentiation and intravillous vascularization. To investigate the difference in expression profiles of placental villi from IP and TEP, placental villi were collected from each phenotype, separated from maternal tissues and analyzed by high-throughput sequencing.

Here, we performed gene expression profiling on neural crest cells isolated from these embryos, to identify the transcriptional response to p53 activation during embryogenesis.

Whole-transcriptome profiling analysis of first trimester placental villi in Indian women

Project description:Whole-transcriptome profiles of individual human placental villi samples from twenty-five (25) Indian women with normal pregnancies during 6- to 8-weeks of gestation were examined using human whole genome expression arrays (NimbleGen 135K). The present study focused on the whole-transcriptome profiling using NimbleGen135K (070925_HG18_exp__12X135K) human whole genome expression arrays of individual human placental villi samples obtained from twenty-five (25) proven-fertile women bearing normal pregnancies voluntarily terminated between 6- and 8-weeks of gestation. Gestational age was estimated from menstrual history, physical and ultrasonographic evaluation. No case of complicated pregnancy from infection, and other significant fetal and maternal clinical indications was included. These twenty five (25) samples include biological replicates of 6, 7 and 8 weeks placental villi samples.

Expression of villi-specific genes reveals the novel prognostic biomarkers in human various cancers

Project description:The relationship between oncogenesis and embryo development are surprised similarity, early placental villi development from 6 weeks to 10 weeks of pregnancy were profiled using microarrays. Through comparative villi and mature placenta data, we identified villi-specific genes that were highly expressed in villus. The large fraction of villi-specific genes dysregulated in transcriptional level across tumors and most of villi-specific genes up-regulated in human cancers. Then by the GO enrichment results, we selected 5 immune-related genes, 6 proliferation-related genes, 8 focal adhesion-related genes that were found to have reported with genomic alterations. The expression of three group genes predicted poor prognosis of a subset of tumors. Our strategy provided a fresh idea for discovering the novel prognostic biomarkers of tumors.

Exosomal microRNAs in tubal fluid may be involved in tubal epithelial structure damage and reproductive dysfunction associated with tubal endometriosis.

Project description:Endometriosis (EM) is a well-recognized disease that causes infertility in women of reproductive age. Tubal EM adversely affects fallopian tube function and female reproductive outcomes. Our study aimed to evaluate the effect of tubal EM on tubal epithelial ultrastructure and to explore the possible mechanisms of tubal infertility by analyzing the differential expression of exosomal microRNAs (miRNAs) in tubal fluid. We obtained human fallopian tube epithelium and tubal fluid samples from patients with and without tubal EM. We used scanning electron microscopy (SEM) and transmission electron microscopy (TEM) to assess ultrastructural changes. Exosomal miRNAs in tubal fluid were extracted for microarray. We visualized epithelial damage in the tubal EM group using electron microscopy. The number of organelles decreased, and organelle structure was destroyed. A total of 14 differentially expressed exosomal miRNAs were detected in tubal fluid (fold change >2 and P value <0.05). Four miRNAs (miR-1273f, miR-5699-5p, miR-6087, miR-6747-5p) were validated by quantitative real-time polymerase chain reaction (qRT-PCR). Bioinformatic analysis showed that most of the target genes participated in embryo transport, regulation of cell communication, anatomical structure morphogenesis, and immune system processes. We verified that tubal EM damaged the tubal epithelial ultrastructure in human specimens and identified differentially expressed exosomal miRNAs in tubal liquid. These findings may elucidate the pathogenesis of tubal EM-associated infertility.

Differential miRNA profiles of pregnancy tissue in ectopic pregnancies

Project description:Abstract Objective: To investigate the physiology of ectopic pregnancy by analyzing the miRNA profiles of the pregnancy tissue from both ectopic and control pregnancies (voluntary termination of pregnancy). Design: Research study. Setting: Academic research institute in collaboration with a university hospital. Patients: Patients suffering from tubal EP and patients with a normal ongoing pregnancy scheduled for a voluntary termination of pregnancy (VTOP) were recruited. Interventions: Pregnancy tissue samples were analyzed by miRNA microarray and further validated by qPCR. Main Outcome measures: Gene expression profiles and quantitative PCR measurements. Results: Four miRNAs were found to be downregulated in EP compared to healthy trophoblast tissue, and three miRNAs were upregulated in EP trophoblast compared to control tissue samples. All genes were validated by q-PCR. We found three statistically significant miRNAs, all of which were upregulated in the EP sample group. Conclusion: We describe the alteration of seven miRNAs in EP samples that could alter pathways which are critical for correct implantation such as extracellular matrix (ECM) remodeling, or mucin biosynthesis, thus resulting in ectopic pregnancies. This study was authorized by the Institutional Review Board/Independent Ethics Committee of the Hospital Universitario La Fe, Valencia, Spain. Eight patients suffering from tubal EP and another eight patients with a normal ongoing pregnancy scheduled for a VTOP were recruited.

DNA methylation profiling of placental villi from karyotypically normal recurrent miscarriage

Project description:Miscarriage occurs in 15-20% of clinical pregnancies. While chromosomal errors are observed in over 50%, causes of karyotypically normal losses are poorly understood. DNA methylation undergoes reprogramming during development and must be appropriately set to maintain a healthy pregnancy. We hypothesize that aberrant DNA methylation may cause karyotypically normal miscarriage, particularly among women experiencing recurrent miscarriage (RM). DNA methylation in first trimester chorionic villi was assessed in chromosomally normal miscarriages from women with RM (N=33) or isolated miscarriage (M, N=21), and elective terminations (TA, N=16). Differentially methylated candidate loci were identified using the Illumina Infinium HumanMethylation27 BeadChip array by comparing 10 RM to 10 TA samples. Follow up showed a significant increase in methylation in RM and M compared to TA placentae at the CYP1A2 (p=0.002) and AXL (p=0.02) promoters and decrease at the DEFB1 (p=0.008) promoter. Gene function analysis showed an enrichment of imprinted genes (p=9.53E-10) and genes previously associated with RM (p=9.51E-06). DNA methylation was evaluated at 7 imprinted loci using bisulfite pyrosequencing. An increase of outliers at these loci was observed in RM (3.9%) compared to M (0%) and TA (0.9%) (p=0.02), with increased average methylation at the H19/IGF2 ICR1 in M samples (p<0.0001). Altered DNA methylation in the placenta at specific loci, as well as global dysregulation in specific cases, may contribute to or be a consequence of placental insufficiency in karyotypically normal miscarriage. First-trimester placental villi samples from karyotypically normal miscarriages from recurrent miscarriage patients (N, N=10) and chromosomally normal elective terminations (PZET, N=10).

Landscape of single-cell transcriptomics of human placent in pathophysiological pregnancies

Project description:Pathophysiological pregnancies amy result from gene-environmental interactions. In this study, investigation of the placental cell development in both chorionic villi and decidua of variant forms of pathophysiological pregnancies was pursued with a single cell RNA sequencing. Transcriptomic gene differential expression in variant placental cell clusters from chorionic villi and decidua of pregnancies, unique marker genes in trophoblasts that may lead to identification of biomarker for placental cell development states and cell-specific pathway activities were identified. In addition, gene expression profiles of ACE2 and TMPRSS2, the two key proteins involved in SARS-CoV-2 infection, in variant placental cell clusters was documented, which may facilitate exploration of vertical transmission of COVID-19 from mother to fetus.
