The underlying mechanism might be related the changes of genes expression in digestive system. We used microarrays to detail the global programme of gene expression during this process. Jejunums from mice fed with high-glucose diet or control were selected for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain populations of genes related to lipid metabolism in order to increase the temporal resolution of expression profiles. To that end, we analyzed the expressions of genes related to lipid metabolism according to clustering analysis and references.

Gene expression data from lung tissues of C57BL/6J mice

Project description:Homoharringtonine could be a novel therapeutic agent for the treatment and the prevention of aging and age-related diseases (ARDs). Cellular senescence contributes to aging and ARDs. Removal and modulation of senescent cells (SCs) improve physical functions and extend healthspan in animal models. Thus, the development of therapeutics that target SCs (i.e., senotherapeutics) has been proposed as a promising strategy for the treatment and the prevention of aging itself and ARDs. We used drug repositioning to discover new senotherapeutics from 2,150 clinically applied compounds and found that homoharringtonine (HHT) has a senolytic activity in human dermal fibroblasts (HDFs) in vitro and in vivo models. HHT improved physical status and rescued aging phenotypes in progeroid mice and aged mice. In addition, HHT administration ameliorates bleomycin-induced lung fibrosis in mice.

Project description:We previously reported that Def (Digestive-organ expansion factor) was a pan-endodermal enriched factor that is essential for the growth of digestive organs in zebrafish using a def mutant line hi429 as model (Chen et al., 2005). To further elucidate Def function, we generated a Def over-expressed zebrafish line, namely Tg (fabp10a:def)-I, in which def expression was under the control of a liver-specific promoter fabp10a. We used microarrays to detail the global programme of gene expression in Tg (fabp10a:def)-I transgenic line compared with wild type zebrafish control, and identified distinct classes of differently regulated genes in Tg (fabp10a:def)-I line.

Project description:Here we introduce a test for lineage-specific selection of cis-regulation that can be applied to any gene set of interest. Applying the test to hundreds of gene sets in two diverged subspecies of mouse, we find evidence for selection acting on the cis-regulation of several functional classes, including those involved in both physiology (e.g. mitochondria-related genes and growth regulators) and behavior (e.g. locomotory-related genes and memory-related genes). These positively selected gene sets implicate potential targets of positive selection that are supported by quantitative trait loci, and accurately predict a number of phenotypic differences between the two subspecies, suggesting that the divergence in these traits may have been driven by adaptive evolution.
