Project description:We conducted extensive transcriptome profiling studies to characterize 70 SPPARgMs and seven PPARg full agonists in 3T3-L1 adipocytes, and a subset of these ligands in adipose tissue of diabetic db/db mice. In both cases, the SPPARgMs generated attenuated gene regulatory responses, and their gene expression signatures were more enriched in metabolic pathways that are likely to mediate anti-diabetic efficacy than those of PPARg full agonists. More importantly, our profiling results demonstrated that in both 3T3-L1 adipocytes and db/db mice, SPPARgMs regulate the expression of anti-diabetic efficacy-associated genes to a greater extent than that of adverse effect-associated genes, while PPARg full agonists regulate both gene sets proportionally. We conducted 10 independent batches of profiling experiments. Within each batch, drug treatment and pool of vehicle controls were hybridized to the Agilent two channel microarray. Generally 2-3 biological replicates for each condition.

Project description:Our data indicated that activation of the PPARg nuclear receptor induces a retinoid response in human dendritic cells. In order to assess the contribution of retinoid signaling to the PPARg response we decided to use a combination of pharmacological activators and inhibitors of these pathways. Cells were treated with the synthetic PPARg ligand rosiglitazone (RSG), or with RSG along with the RARa antagonist (AGN193109) to block RARa mediated gene expression, or the RARa specific agonists (AM580) alone. This design allows one to determine if retinoid signaling is a downstream event of PPARg activation and what portion of PPARg regulated genes are regulated via induced retinoid signaling. Project description:Clinically, obesity is strongly associated with severe TH2 immunopathology, though the physiological, cellular, and molecular underpinnings of this association remain obscure. We demonstrate that obese mice are susceptible to severe atopic dermatitis (AD), a major manifestation of TH2 immunopathology and disease burden in humans. Mechanistically, we show that dysregulation of the nuclear hormone receptor peroxisome proliferator-activated receptor gamma (PPARg) in T cells is a causal link between obesity and the increased TH2 immunopathology. We find that PPARg directly controls a cellular metabolic transcriptional program that restrains nuclear gene expression of the chief TH2 effector cytokines interleukin-4 (IL-4) and interleukin-13 (IL-13). Accordingly, thiazolidinediones (TZDs), potent PPARg agonists, robustly protect obese mice from TH2 immunopathology. Collectively, these findings establish PPARg as a molecular link between obesity and TH2 immune homeostasis and identify TZDs as novel therapeutic candidates for TH2 immunopathology.

Project description:Transrepression mediated by PPARg and LXRs agonists (GW7845 and GW3965) in primary macrophages stimulated with LPS or poly I:C. Thioglycollate-elicited macrophages were isolated from mice by peritoneal lavage 3 days following peritoneal injection of 2.5 ml 3% thioglycollate (DIFCO). Cells were plated in RPMI medium 1640 and 10% fetal bovine serum, washed after 5 h the medium was removed and cells were fed with fresh medium containing 0.5% fetal bovine serum. Cells were treated with LPS (Sigma) a concentration of 100 ng/ml or poly I:C (100 ng/ml) in the absence or presence of receptor-specific agonists for PPARγ (GW7845) or LXR (GW3965) at 1 µM concentrations. Results suggest signal-dependent nuclear receptors transrepression pathways. Project description:Periodontal ligament is soft connective tissue between alveolar bone and cementum, the surface hard tissue of tooth. Periodontal ligament fibroblasts (PDLF) actively express osteo/cementogenic genes, which contribute in periodontal tissue homeostasis. However, the key factors by which PDLF retain such osteo/cementogenic abilities has not been clarified yet. Here, we find PPARg is expressed in vivo periodontal ligament tissue and its distribution pattern is highly correlated with the alkaline phosphate enzymatic activity. Knockdown of PPARg drastically deprives PDLF of osteo/cementogenic abilities in vitro. In contrast, PPARg agonists enhance it. PPARg is required for maintaining acetylation status of H3K9 and H3K27, active chromatin marks, and the supplementation of acetyl-coA, the donor of histone acetylation, is able to restore PPARg knockdown-induced decreased osteo/cementogenic abilities in PDLF. RNA-seq and ChIP-seq combined analyses identify 4 osteogenic transcripts, RUNX2, SULF2, RCAN2, and RGMA, in the PPARg-dependent active chromatin region marked by H3K27ac. Furthermore, the RUNX2 binding sites are selectively enriched in the PPARg-dependent active chromatin region. Together, these findings identify PPARg is the key transcriptional factor of PDLF to retain osteo/cementogenic abilities and reveal that global H3K27ac modification involves in the comprehensive osteo/cementogenic transcriptional alteration mediated by PPARg.

Experiment Overall Design: Monocytes were cultured for 5 days with 500 U/ml IL-4 and 800 U/ml GM-CSF, cytokine treatment was repeated at day 3. Ligands were added at the beginning of differentiation but AGN193109 (AGN) treatment was repeated at day 3. Cells were obtained from three healthy individuals (three biological replicates) and cells were treated with vehicle (DC), 2.5 uM rosiglitazone (DC RSG), 100 nM AM580 (DC AM). RSG treatment was also combined with 1 uM AGN193109 (DC RSG AGN) in this case two biological replicates was used (RNA was obtained from 2. and 3. individuals).

Project description:Beige adipocytes in mammalian white adipose tissue (WAT) can reinforce fat catabolism and energy expenditure. Promoting beige adipocyte biogenesis is a tantalizing tactic for combating obesity and its associated metabolic disorders. Here, we report that a previously unidentified phosphorylation pattern (Thr166) in the DNA-binding domain of PPARg regulates the inducibility of beige adipocytes. This unique posttranslational modification (PTM) pattern influences allosteric communication between PPARg and DNA or coactivators, which impedes the PPARg-mediated transactivation of beige cell-related gene expression in WAT. The genetic mutation mimicking T166 phosphorylation (p-T166) hinders the inducibility of beige adipocytes. In contrast, genetic or chemical intervention in this PTM pattern favors beige cell formation. Moreover, inhibition of p-T166 attenuates metabolic dysfunction in obese mice. Our results uncover a mechanism involved in beige cell fate determination. Moreover, our discoveries provide a promising strategy for guiding the development of novel PPARg agonists for the treatment of obesity and related metabolic disorders.
